Uncovering the impact of alcohol on internal organs and reproductive health:Exploring TLR4/NF-kB and CYP2E1/ROS/Nrf2 pathways

This review delves into the detrimental impact of alcohol consumption on internal organs and reproductive health,elucidating the underlying mechanisms involving the Toll-like receptor 4(TLR4)/Nuclear factor kappa light chain enhancer of activated B cells(NF-kB)pathway and the Cytochrome P4502E1(CYP2E1)/reactive oxygen spe-cies(ROS)/nuclear factor erythroid 2-related factor 2(Nrf2)*** TLR4/NF-kB pathway,crucial for inflammatory and immune responses,triggers the production of pro-inflammatory agents and type-1 interferon,disrupting the balance between inflammatory and antioxidant responses when tissues are chronically exposed to ***-induced dysbiosis in gut microbes heightens gut wall permeability to pathogen-associated molecular patterns(PAMPs),leading to liver cell infection and subsequent ***,CYP2E1-mediated alcohol metabolism gen-erates ROS,causing oxidative stress and damaging cells,lipids,proteins,and deoxy-ribonucleic acid(DNA).To counteract this inflammatory imbalance,Nrf2 regulates gene expression,inhibiting inflammatory progression and promoting antioxidant *** alcohol intake results in elevated liver enzymes(ADH,CYP2E1,and catalase),ROS,NADH,acetaldehyde,and acetate,leading to damage in vital organs such as the heart,brain,and ***,alcohol negatively affects reproduc-tive health by inhibiting the hypothalamic-pituitary-gonadal axis,causing infertility in both men and *** findings underscore the profound health concerns associated with alcohol-induced damage,emphasizing the need for public awareness regarding the intricate interplay between immune responses and the multi-organ im-pacts of alcohol consumption.

Comparative chemical sensor for detection of chromium(VI) in aqueous solution

A process for purifying aqueous solutions containing heavy and toxic metals such as chromium has been investigated. One of the extremely harmful pollutants in rivers and seawater are the heavy metal ions due to their direct impact on human， animals and plants are hexavalent chromium Cr （VI）. Consequently， highly sensitive sensor to detect Chromium is essential. Surface Plasmon resonance （SPR） technique has attracted huge research interest in detecting heavy metals specifically. In this study， three types of prism-based SPR sensor; gold （Au）/silver （Ag）， Au/polyaniline （PANI） and Au/ titanium dioxide (TiO2) nanostructured films are investigated as potential sensing material to detect the presence of Cr （VI） ions in water. The base Au layer with thickness of 48.3 nm is deposited on a glass slide for all sensors. For Au/Ag， Au/PANI nanofibers and Au/ (TiO2) sensor films， the Cr （VI） concentration is varied from 1 to 15 ppm with sensitivity of 0.270 o ppm-1， 0.082o ppm-1and 0.039 o ppm-1 respectively. Based on these results， the Au/ PANI nanofibers are the most sensitive to chromium Cr （VI） among tested sensing materials.

Anti-inflammatory Effects of Ampelopsis Japonica Root on Contact Dermatitis in Mice

AIM： To investigate the effect of matrix metallopro- teinase-9 （MMP-9） on the remnant liver after massivehepatectomy in the mouse.METHODS： Age-matched, C57BL/6 wild-type （WT）, MMP-9（-/-）, and tissue inhibitors of metalloprotein- ases （TIMP）-1（-/-） mice were used. The mice received 80%-partial hepatectomy （PH）. Samples were obtained at 6 h after 80%-PH, and we used histology, immuno- histochemical staining, western blotting analysis and zymography to investigate the effect of PH on MMP-9. The role of MMP-9 after PH was investigated using a monoclonal antibody and MMP inhibitor.RESULTS： We examined the remnant liver 6 h after 80%-PH and found that MMP-9 deficiency attenuated the formation of hemorrhage and necrosis. There were significantly fewer and smaller hemorrhagic and ne- crotic lesions in MMP-9（-/-） remnant livers compared with WT and TIMP-1（-/-） livers （P 〈 0.01）, with no dif- ference between WT and TIMP-1（-/-） mice. Serum ala- nine aminotransaminase levels were significantly lower in MMP-9（-/-） mice compared with those in TIMP-I（-/-） mice （WT： 476± 83 IU/L, MMP-9（-/-）： 392 ± 30 IU/L, TIMP-I（-/-）： 673 ± 73 IU/L, P 〈 0.01）. Western blot- ting and gelatin zymography demonstrated a lack of MMP-9 expression and activity in MMP-9（-/-） mice, which was in contrast to WT and TIMP-1（-/-） mice. No change in MMP-2 expression was observed in any of the study groups. Similar to MMP-9（-/-） mice, when WT mice were treated with MMP-9 monoclonal antibody or the synthetic inhibitor GM6001, hemorrhagic and necrotic lesions were significantly smaller and fewer than in control mice （P 〈 0.05）. These results suggest that MMP-9 plays an important role in the development of parenchymal hemorrhage and necrosis in the small remnant liver.CONCLUSION： Successful MMP-9 inhibition attenuates the formation of hemorrhage and necrosis and mightbe a potential therapy to ameliorate liver injury after massive hepatectomy.

changes in Activity of Three Sulfurtransferases in Response to Exposure to Cadmium, Lead and Mercury Ions

Cadmium, lead and mercury are environmentally persistent toxicants that affect tissues and cellular components or exert an effect on generation of reactive oxygen species causing a decreased level of available antioxidant reserves. Sulfurtransferases are enzymes that are widespread in nature. Rhodanese, 3-mercaptopyruvate sulfurtransferase and γ-cystathionase play an important role in the metabolism of L-cysteine. Heavy metal ions can bind to -SH groups of cysteine residues in their active sites and, therefore, decrease the activity of these enzymes and result in changes in the level of sulfane sulfur-containing compounds, products of L-cysteine desulfuration. changes in the activity of sulfurtransferases were investigated in the kidneys, heart, brain, liver and skeletal muscle of marsh frogs (Pelophylax ridibundus) after 10 days of exposure to Pb(NO3)2 at the concentration of 28 mg/L and CdCl2 at the concentration of 40 mg or 80 mg/L, and in Xenopus laevies tissues after 7 and 14 days of exposure to HgCl2 at the concentration of 1.353 mg/L. The investigated heavy metal ions have a tendency to inhibit the activity of sulfurtransferases and decrease the level of glutathione, what can result in oxidative stress and oxidation of cysteine -SH groups to -SOH. This reversible oxidation and reduction of these redox sensitive groups can play a role in defenses against oxidative stress. Based on the presented results, one can surmise that also the expression of the three sulfurtransferases depends on heavy metal ions and/or some parameters of oxidative stress, what can explain the increase of the activity of MPST and CST in the kidney.

The Impact of Za’atari Refugee Camp on the Water Quality in Amman-Zarqa Basin

Za’atari camp is the largest refugee camp in Jordan. It was first established in 2012 to host Syrian refugees. Currently the camp hosts more than 81,000 refugees, with no proper sanitary system which might pose a major threat to surface resources in the area. An investigation was done at Za’atari refugees’ camp to find the impact of refugees settling on surface and groundwater quality. Surface water quality of surface runoff generated from thirty rain fall events were collected during the winter season of 2013/2014 from the major Wadi that passes through the camp and small ponds within the camp after the rainfall event. The collected samples were analyzed for acidity (pH), the electrical connectivity (EC), total dissolved solids (TDS), nutrients (NO3margin-left:-7px;'>- and PO4margin-left:-7px;'>3-) and selected heavy metals (Mn, Cd, Zn, Pb and Ni) in addition to biological oxygen demand (BOD5), chemical oxygen demand (COD) and intestinal worms (Total Coliform, E. cali). The results showed that there are significant variations in the EC as well as with TDS between the sites due to fluctuating amounts of water used for different activities within the camp as it was highest in the center of the camp where most of the refugees settle decreasing away from the center. The pH values were within the specifications of the World Health Organization and the Jordanian Standards. For nutrients, nitrate concentration was low with high phosphate ions which are most probably from detergents origin.
