Polar Gini Curve:A Technique to Discover Gene Expression Spatial Patterns from Single-cell RNA-seq Data

In this work,we describe the development of Polar Gini Curve,a method for characterizing cluster markers by analyzing single-cell RNA sequencing(scRNA-seq)*** Gini Curve combines the gene expression and the 2D coordinates(“spatial”)information to detect patterns of uniformity in any clustered cells from scRNA-seq *** demonstrate that Polar Gini Curve can help users characterize the shape and density distribution of cells in a particular cluster,which can be generated during routine scRNA-seq data *** quantify the extent to which a gene is uniformly distributed in a cell cluster space,we combine two polar Gini curves(PGCs)—one drawn upon the cell-points expressing the gene(the“foreground curve”)and the other drawn upon all cell-points in the cluster(the“background curve”).We show that genes with highly dissimilar foreground and background curves tend not to uniformly distributed in the cell cluster—thus having spatially divergent gene expression patterns within the *** with similar foreground and background curves tend to uniformly distributed in the cell cluster—thus having uniform gene expression patterns within the *** quantitative attributes of PGCs can be applied to sensitively discover biomarkers across clusters from scRNA-seq *** demonstrate the performance of the Polar Gini Curve framework in several simulation case *** this framework to analyze a real-world neonatal mouse heart cell dataset,the detected biomarkers may characterize novel subtypes of cardiac muscle *** source code and data for Polar Gini Curve could be found at http://***/PGC/or https://***/projects/Polar_Gini_Curve/76749.

This work reports on the effect of commercial expandable graphite(EG)on the flammability and thermal decomposition properties of PLA-starch *** PLA-starch/EG composites were prepared by melt-mixing and their thermal stability,volatile pyrolysis products and flammability characteristics were *** char residues of the composites,after combustion in a cone calorimeter,were analyzed with environmental scanning electron microscopy(ESEM).The thermal decomposition stability of the composites improved in the presence of ***,the char content was less than expected as per the combination of the wt%EG added into PLA-starch and the%residue of *** flammability performance of the PLA-starch/EG composites improved,especially at 15 wt%EG content,due to a thick and strong worm-like char *** peak heat release rate(PHRR)improved by 74%,the total smoke production(TSP)by 40%and the specific extinction area(SEA)by 55%.The improvements are attributed to the ability of EG to exfoliate at increased temperatures during which time three effects occurred:(i)cooling due to an endothermic exfoliation process,(ii)dilution due to release of H2O,SO2 and CO2 gases,and (iii)formation of a protective intumescent char ***,the CO and CO2 yields were found to be unfavorably high due to the presence of EG.

Spatial distributions of animals can affect interactions with their natural enemies,such as parasites,and thus have important implications for host-parasite *** spatial variation in infection risk has been explored in many systems at the landscape scale,less attention has been paid to spatial structure at smaller ***,we explore a hypothesized relationship between a common spatial variable,vertical position,and risk of parasite infection in a model aquatic system,larval frogs(Rana)and trematode(Digenea)*** position is relevant to this system given evidence that the densities of snail first intermediate hosts,tadpole second intermediate hosts,and trematode infective stages can vary with *** test the effects of depth on infection risk of larval frogs by trematodes,we performed two enclosure experiments,one in the laboratory and one in the field,in which larval frogs in cages just below the water surface or near the bottom of the water column were exposed to *** with near-surface cages,mean infection load(number of cysts)in tadpoles in near-bottom cages was 83%higher after 48-h exposures in the laboratory and 730%higher after 10-day exposures in the *** findings thus indicate that infection risk depends on depth,which may have adaptive significance,as tadpoles have previously been shown to change vertical position in response to parasite *** results motivate future work examining vertical variation in infection risk and may have broader implications for host-parasite dynamics and evolution of host and parasite behavior.

Bile acids,once considered mere dietary surfactants,now emerge as critical modulators of macronutrient(lipid,carbohydrate,protein)metabolism and the systemic pro-inflammatory/anti-inflammatory *** acid metabolism and signaling pathways play a crucial role in protecting against,or if aberrant,inducing cardiometabolic,inflammatory,and neoplastic conditions,strongly influencing health and *** curative treatment exists for any bile acid influenced disease,while the most promising and well-developed bile acid therapeutic was recently rejected by the ***,we provide a bottom-up approach on bile acids,mechanistically explaining their biochemistry,physiology,and pharmacology at canonical and non-canonical *** this mechanistic model of bile acids,we explain how abnormal bile acid physiology drives disease pathogenesis,emphasizing how ceramide synthesis may serve as a unifying pathogenic feature for cardiometabolic *** provide an in-depth summary on pre-existing bile acid receptor modulators,explain their shortcomings,and propose solutions for how they may be ***,we rationalize novel targets for further translational drug discovery and provide future *** than dismissing bile acid therapeutics due to recent setbacks,we believe that there is immense clinical potential and a high likelihood for the future success of bile acid therapeutics.

In fast reactors, the inherent neutron source strength is often insufficient for monitoring the reactor start-up operation with ex-core detectors. To increase the subcritical neutron flux, an auxiliary neutron source subassembly(SSA) is generally used to overcome this problem. In this study, the estimated neutron source strength and detector count rate of an antimony-beryllium-based SSA are obtained using the deterministic transport code DORT and Monte Carlo calculations. Because the antimony activation rate is a critical parameter, its sensitivity to the capture cross section and neutron flux spectrum is studied. The reaction cross section sensitivity is studied by considering data from different evaluated nuclear data *** is observed that, because of the variation in the cross sections from different evaluated nuclear data files, the values of the saturation gamma(> 1.67 MeV) activity and neutron strength predicted by ORIGEN2 lie within ±2%.The obtained antimony activation rate and sensitivity to the neutron flux are partially validated by irradiating samples of antimony in the KAMINI reactor. The average onegroup capture cross sections of bare and cadmium-covered 123Sb samples obtained by the ratio method are 4.0 and 1.78 b, respectively. The results of the calculation predicting the activated neutron source strength as a function of operating time and sensitivity to the neutron spectrum in the irradiation region are also presented.

Bismuth-based Sillen-Aurivillius compounds are being explored as efficient photocatalyst materials for the degradation of organic pollutants due to their unique layered structure that favours effective separation of electron-hole pairs. In this work, we synthesized Sillen-Aurivillius-related Bi2YO4Cl with the bandgap of 2.5 eV by a simple solid-state reaction and sensitized with rhodium nickel (RhNi) nanoparticles (NPs) to form the RhNi/Bi2YO4Cl heterostructure. Photocatalytic activities of BiOCl, Bi2YO4Cl and the RhNi/Bi2YO4Cl heterostructure were examined for the degradation of rhodamine- 6G under visible-light illumination. Results demonstrated that the photocatalytic dye degradation efficiency of RhNi/Bi2YO4Cl heterostructures is higher than those of BiOCl and Bi2YO4Cl, attributed to the synergistic molecular-scale alloying effect of bimetallic RhNi NPs. The plausible mechanism for the degradation of rhodamine-6G and the effective electron-hole pair utilization mechanism were discussed.

AIM: To further define variables associated with increased incidences of severe toxicities following administration of yttrium-90 (90Y) ***: Fifty-eight patients undergoing 79 treatments were retrospectively assessed for development of clinical and laboratory toxicity incidence following 90Y administration. Severe toxicity events were defined using Common Terminology Criteria for Adverse Events version 4.03 and defined as grade ≥ 3. Univariate logistic regression analyses were used to evaluate the effect of different factors on the incidence of severe toxicity events. Multicollinearity was assessed for all factors with P 90Y microsphere therapy.
