Trans-10,cis-12, not cis-9,trans-11, conjugated linoleic acid decreases ErbB3 expression in HT-29 human colon cancer cells

AIM： To examine whether trans-10,cis-12 CLA （t10C12） or cis-9,trans-11 CLA （c9t11） inhibits heregulin （HRG）-β- stimulated cell growth and HRG-β-ErbB3 signaling in HT-29 cells. METHODS： We cultured HT-29 cells in the absence or presence of the CLA isomers and/or the ErbB3 ligand HRG-β. MTT assay, [^3H]thymidine incorporation, Annexin V staining, RT-PCR, Western blotting, immunoprecipitation, and in vitro kinase assay were performed. RESULTS： HRG-β increased cell growth, but did not prevent DNA t10c12-induced growth inhibition. T10C12 inhibited DNA synthesis and induced apoptosis of HT-29 cells, whereas c9t11 had no effect. T10c12 decreased the levels of ErbB1, ErbB2, and ErbB3 proteins and transcripts in a dose-dependent manner, whereas c9t11 had no effect. Immunoprecipitation/ Western blot studies revealed that t10c12 inhibited HRG- β-stimulated phosphorylation of ErbB3, recruitment of the p85 subunit of phosphoinositide 3-kinase （PI3K） to ErbB3, ErbB3-associated PI3K activities, and phosphorylation of Akt. However, c9t11 had no effect on phospho Aid： levels. Neither t10c12 nor c9t11 had any effect on HRG-β-induced phosphorylation of ERK-1/2. CONCLUSION： These results indicate that the inhibition of HT-29 cell growth by t10c12 may be induced via its modulation of ErbB3 signaling leading to inhibition of Akt activation.

Effect of dietary supplementation of xylanase on apparent ileal digestibility of nutrients,viscosity of digesta,and intestinal morphology of growing pigs fed corn and soybean meal based diet

This study was to determine apparent ileal digestibility of acid detergent fiber(ADF), neutral detergent fiber(NDF), dry matter(DM), energy, organic matter(OM), crude ash, digesta viscosity, and gut morphology in nursery pigs fed diets containing xylanase(Lohmann Animal Nutrition GmbH, Cuxhaven, Germany). The diet(61% corn, 35% soybean meal, 1% poultry fat, and 3% minerals and vitamins) was mixed with 3 levels of xylanase(0,700, and 1400 LXU/kg). Thirty-six barrows(17.6 ± 3.3 kg) received one of 3 treatment diets based on a randomized complete block design with the initial body weight(BW) as a block. Pigs were individually housed and received experimental diets twice daily(0700 and 1700 h) at a fixed amount based on BW of pigs(0.09 x BW^(0.75) kg). Pigs were fed diets for 10 d, and chromium oxide(0.3%) was added to the diets from d 6 as an indigestible external marker. Pigs were euthanized at the end of d 10 for the collection of digesta and tissues. Jejunal digesta were centrifuged to measure viscosity using a viscometer(Brookfield Engineering Laboratories, Stoughton, MA). Diets and freeze-dried ileal digesta were used to measure ADF, NDF, and chromium to calculate apparent ileal digestibility of ADFand NDF. Villus height and crypt depth of jejunum were measured using a microscope(Fisher Scientific, Hampton, NH). Data were analyzed using polynomial contrasts in the MIXED procedure of SAS version 9.3(SAS Inc., Cary, NC, USA). Morphological measurements and ileal ADF digestibility were not affected by increasing xylanase. However, increasing xylanase supplementation from 0 to 1400 LXU/kg enhanced ileal digestibility of NDF(P < 0.042, linear) from 27.9 to 40.3%,DM(P < 0.006, linear) from 55.4 to 64.6%, OM(P < 0.006, linear) from 59.2 to 67.7%, and energy(P < 0.003,linear) from 58.8 to 68.0%. Gastrointestinal stromal tumor (GIST) represents the most common kind of mesenchymal tumor that arises from the alimentary tract. GIST is currently defined as a gastrointestinal tract mesenchymal tumor showing CD117 (c-kit protein) positivity at immunohistochemistry. Throughout the whole length of the gastrointestinal tract, GIST arises most commonly from the stomach followed by the small intestine, the colorectum, and the esophagus. Only 3%-5% of GISTs occur in the duodenum, and especially, if GIST arises from the C loop of the duodenum, it can be difficult to differentiate from the pancreas head mass because of its anatomical proximity. Here, we report a case of duodenal GIST, which was assessed as a pancreatic head tumor preoperatively.

BACKGROUND Accessory soleus muscle(ASM)is a rare congenital variation that is almost asymptomatic,but several papers have recently described symptomatic *** clinical features of this condition are similar to tarsal tunnel syndrome(TTS)and include pain and numbness around the medial side of the *** commonly originates from the fibula or soleus muscle and inserts into the Achilles tendon or ***,it is identified as posteromedial swelling and definitely diagnosed by magnetic resonance *** most cases,treatment is observation,but surgical excision can be considered if symptoms are *** SUMMARY A 23-year-old male Korean soldier presented with complaints of bilateral foot and ankle pain and a swelling medial to the Achilles tendon that was more pronounced on the right *** first occurred after playing soccer 10 mo before this presentation,worsened after physical exertion,and were relieved by *** had no medical history,and no one in his family had the *** results were *** tests were performed to exclude common diseases such as tumors or ***,MRI revealed a bulky accessory soleus muscle in both feet,though the patient complained of more severe pain on the right side during physical ***,surgical resection was *** surgery,a large accessory soleus muscle was noted anterior to the Achilles tendon with distinctive insertion from a normal soleus *** 12 mo after surgery,there was no pain,numbness,or swelling of the right foot or ankle,no evidence of recurrence,and the patient could do all sports *** Accessory soleus muscle should be added to the list of differential diagnosis if a patient has pain,sole numbness or swelling of the posteromedial ankle.

A recurrent pandemic with unpredictable viral nature has implied the need for a rapid diagnostic technology to facilitate timely and appropriate countermeasures against viral *** this study,conductive polymer-based nanoparticles have been developed as a tool for rapid diagnosis of influenza A(H1N1)*** distinctive property of a conductive polymer that transduces stimulus to respond,enabled immediate optical signal processing for the specific recognition of H1N1 *** poly(aniline-co-pyrrole)-encapsulated polymeric vesicles,functionalized with peptides,were fabricated for the specific recognition of H1N1 *** low solubility of conductive polymers was successfully improved by employing vesicles consisting of amphiphilic copolymers,facilitating the viral titer-dependent production of the optical *** optical response of the detection system to the binding event with H1N1,a mechanical stimulation,was extensively analyzed and provided concordant information on viral titers of H1N1 virus in 15 *** specificity toward the H1N1 virus was experimentally demonstrated via a negative optical response against the control group,***,the designed system that transduces the optical response to the target-specific binding can be a rapid tool for the diagnosis of H1N1.

Anti-inflammatory effects of natural volatile organic compounds from Pinus koraiensis and Larix kaempferi in mouse model

Natural volatile organic compounds(VOCs) extracted from conifers such as P. koraiensis and L. kaempferi have long been studied for their anti-oxidant, anti-proliferative, and anti-inflammatory effects. To evaluate the anti-inflammatory effects of VOCs from P. koraiensis and L. kaempferi, lipopolysaccharide(LPS) was administered to generate a mouse model for inflammation by the nasal route to the lungs and intraperitoneally to the whole body. VOCs of P. koraiensis and L. kaempferi were exposed to the mice by standardized wood panels with closed system. Increased levels of serum IgE and PGE2 were observed after exposure to dexamethasone and VOCs. We further determined the expression levels of inflammatory cytokine mRNA in the LPS-induced inflammation model by the reverse transcription quantitative polymerase chain reaction. Furthermore, the levels of cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β, and interleukin-13 were determined in peripheral blood mononuclear cells. Those inflammatory cytokines and the key enzyme for inflammation cyclooxygenase-2 expression in PBMCs were strongly reversed by dexamethasone and VOCs. Lung tissues after nasal LPS exposure showed increased cytokine mRNA expressions which were suppressed by treatment with dexamethasone and VOCs. Furthermore, the damage induced by LPS was attenuated by dexamethasone and VOCs. In conclusion, the results from the present study indicate that VOCs of P. koraiensis and L. kaempferi have a therapeutic potential in the treatment or prevention of local and systemic inflammation due to their immunosuppressive effects.

Formation of interfacial brittle phases sigma phase and IMC in hybrid titanium-to-stainless steel joint

The microstructures of the brazed joints for commercially pure Ti and stainless steel were investigated by the applications of various filler alloys including Ag-, Ti-, Zr- and Ni-based alloys. Generally, the dissimilar joints between Ti and stainless steel were dominated by the Ti-based intermetallic compounds （IMCs）, e.g. （Ti, Zr）2（Fe, Ni）, TiFe, TiCu, and Ti2（Fe, Ni）, due to a significant dissolution of Ti from the base metal. The （Fe-Cr） cr phase was also observed near the stainless steel due to a segregation of Cr into the interface region. This research demonstrates empirically that the brittleness of the Ti and stainless steel joint can not be avoided only by applying single braze alloy or single insert metal, and thus an introduction of additional suitable interlayer between the filler alloy and the base metal is necessary to prevent the brittleness of the joint.

Surgical outcome of pancreatic cancer using radical antegrade modular pancreatosplenectomy procedure

AIM:To evaluate the surgical outcomes following radical antegrade modular pancreatosplenectomy (RAMPS) for pancreatic cancer. METHODS:Twenty-four patients underwent RAMPS with curative intent between January 2005 and June 2009 at the National Cancer Center, South Korea. Clinicopathologic data, including age, sex, operative findings, pathologic results, adjuvant therapy, postop-erative clinical course and follow-up data were retro-spectively collected and analyzed for this study. RESULTS:Twenty-one patients (87.5%) underwent distal pancreatectomy and 3 patients (12.5%) underwent total pancreatectomy using RAMPS. Nine patients (37.5%) underwent combined vessel resection, including 8 superior mesenteric-portal vein resections and 1 celiac axis resection. Two patients (8.3%) underwent combined resection of other organs, including the colon, stomach or duodenum. Negative tangential margins were achieved in 22 patients (91.7%). The mean tumor diameter for all patients was 4.09 ± 2.15 cm. The 2 patients with positive margins had a mean diameter of 7.25 cm. The mean number of retrieved lymph nodes was 20.92 ± 11.24 and the node positivity rate was 70.8%. The median survival of the 24 patients was 18.23 ± 6.02 mo. Patients with negative margins had a median survival of 21.80 ± 5.30 mo and those with positive margins had a median survival of 6.47 mo (P = 0.021). Nine patients (37.5%) had postoperative complications, but there were no postoperative mortalities. Pancreatic fistula occurred in 4 patients (16.7%):2 patients had a grade A fistula and 2 had a grade B fistula. On univariate analysis, histologic grade, positive tangential margin, pancreatic fistula and adjuvant therapy were significant prognostic factors for survival. CONCLUSION:RAMPS is a feasible procedure for achieving negative tangential margins in patients with carcinoma of the body and tail of the pancreas.
