Inhibition of Proliferation of Prostate Cancer Cell Line DU-145 in vitro and in vivo Using Salvia miltiorrhiza Bunge

Objective:To investigate the antiproliferative activity of Salvia miltiorrhiza Bunge.(SM)on the castration-resistant prostate cancer(CRPC)cell line DU-145 in vitro and in vivo.Methods:Prostate cancer cell line(DU-145)and normal prostate cell line(RWPE-1)were treated with SM at different concentrations(3.125,12.5,25 and 50μg/mL)to investigate the antiproliferative effects.DNA laddering analysis was performed to investigate the apoptosis of DU-145 cells.Molecular mechanism was investigated by Western blot analysis of p53,Bcl-2,prostate specific antigen(PSA),and androgen receptor(AR).Six-week-old male BALB/c nude mice were randomly divided into normal control group(n=101)and treated group(n=101)which administered 500 mg/kg SM for 2 weeks.Tumor volumes were measured.Results:Treatment with SM resulted in a dose-dependent decrease in cell number of DU-145 cells in comparison with RWPE-1.DNA laddering analysis indicated the apoptosis of DU-145 cells.Treatment with SM increased the expression of p53 and reduced the expression of Bcl-2 proteins.The levels of PSA were considerably reduced in SM-treated group compared to the controls,and a decrease in AR expression was observed when cells were treated with SM in the same pattern as a reduction in PSA.In the tumour xenograft study,SM given once a day for 2 weeks significantly inhibited tumour growth.Conclusion:SM might contribute to the anticancer actions such as induction of apoptosis and inhibition of proliferation of prostate cancer cells.

Anti-inflammatory and Antimicrobial Effects of Anthocyanin Extracted from Black Soybean on Chronic Bacterial Prostatitis Rat Model

Objective： To investigated the anti-inflammatory and antimicrobial effects of anthocyanins extracted from black soybean on the chronic bacterial prostatitis（CBP） rat model.Methods： The SpragueDawley rats were divided into 4 groups,including control,ciprofloxacin,anthocyanins and anthocyanins with ciprofloxacin groups（n=8 in each group）.Then,drip infusion of bacterial suspension（Escherichia coli Z17O2：K1：H^-） into Sprague-Dawley rats was conducted to induce CBP.In 4 weeks,results of prostate tissue,urine culture,and histological analysis on the prostate were analyzed for each group.Results： The use of ciprofloxacin,anthocyanins,and anthocyanins with ciprofloxacin showed statistically significant decreases in bacterial growth and improvements in the reduction of prostatic inflammation compared with the control group（P〈0.05）.The anthocyanins with ciprofloxacin group showed a statistically significant decrease in bacterial growth and improvement in prostatic inflammation compared with the ciprofloxacin group（P〈0.05）.Conclusions： These results suggest that anthocyanins may have anti-inflammatory and antimicrobial effects,as well as a synergistic effect with ciprofloxacin.Therefore,we suggest that the combination of anthocyanins and ciprofloxacin may be effective in treating CBP to obtain a higher rate of treatment success.

We aimed to evaluate the current nationwide trend,efficacy,safety,and quality of life(QoL)profiles of hormone treatment in real-world practice settings for prostate cancer(PCa)patients in Korea.A total of 292 men with any biopsy-proven PCa(TanyNanyMany)from 12 institutions in Korea were included in this multi-institutional,observational study of prospectively collected data.All luteinizing hormone-releasing hormone(LHRH)agonists were allowed to be investigational drugs.Efficacy was defined as(1)the rate of castration(serum testosterone≤50 ng dl^-1)at 4-week visit and(2)breakthrough(serum testosterone>50 ng dl^-1 after castration).Safety assessments included routine examinations for potential adverse events,laboratory tests,blood pressure,body weight,and bone min era I density(BMD,at baseli ne and at the last follow-up visit).QoL was assessed using the Expa nded Prostate Cancer Index Composite-26(EPIC-26).The most comm on initial therapeutic regimen was LHRH agonist with an ti-androge n(78.0%),and the most commonly used LHRH agonist for combination and mono therapy was leuprolide(64.0%for combi natio n and 58.0%for monotherapy).The castration and breakthrough rates were 78.4%and 6.6%,respectively.The laboratory results related to dyslipidemia worsened after 4 weeks of hormone treatment.In addition,the mean BMD T-score was significantly lower at the last follow-up(mean:-1.950)compared to baseline(mean:-0.195).The mean total EPIC-26 score decreased from 84.8(standard deviation[s.d.]:12.2)to 78.3(s.d.:8.1),with significant deterioration only in the urinary domain(mean:23.5 at baseli ne and 21.9 at the 4-week visit).These findi ngs dem on strate the n ationwide trend of current practice setti ngs in hormone treatment for PCa in Korea.

Antiproliferative Effects of Zinc-Citrate Compound on Hormone Refractory Prostate Cancer

Objective： To investigate the antiproliferative effects of zinc-citrate compound on hormone refractory prostate cancer （HRPC）. Methods： HRPC cell line （DU145） and normal prostate cell line （RWPE-1） were treated with zinc, citrate and zinc-citrate compound at different time intervals and concentrations to investigate the effect of zinc-citrate compound. Mitochondrial （m）-aconitase activity was determined using aconitase assay. DNA laddering analysis was performed to investigate apoptosis of DU145 cells. Molecular mechanism of apoptosis was investigated by Western blot analysis of P53, P21w^fl, Bcl-2, Bcl-xL and Bax, and also caspase-3 activity analysis. Results： Treatment with zinc-citrate compound resulted in a time- and dose-dependent decrease in cell number of DU145 cells in comparison with RWPE-1. M-aconitase activity was significantly decreased. DNA laddering analysis indicated apoptosis of DU145 cells. Zinc-citrate compound increased the expression of P21wall and P53, and reduced the expression of Bcl-2 and Bcl-xL proteins but induced the expression of Bax protein. Zinc-citrate compound induced apoptosis of DU145 cells by activation of the caspase-3 pathway. Conclusion： Zinc-citrate compound can induce apoptotic cell death in DU145, by caspase-3 activation through up-regulation of apoptotic proteins and down-regulation of antiapoptotic proteins.

Nanograin network memory with reconfigurable percolation paths for synaptic interactions

The development of memory devices with functions that simultaneously process and store data is required for efficient computation.To achieve this,artificial synaptic devices have been proposed because they can construct hybrid networks with biological neurons and perform neuromorphic computation.However,irreversible aging of these electrical devices causes unavoidable performance degradation.Although several photonic approaches to controlling currents have been suggested,suppression of current levels and switching of analog conductance in a simple photonic manner remain challenging.Here,we demonstrated a nanograin network memory using reconfigurable percolation paths in a single Si nanowire with solid core/porous shell and pure solid core segments.The electrical and photonic control of current percolation paths enabled the analog and reversible adjustment of the persistent current level,exhibiting memory behavior and current suppression in this single nanowire device.In addition,the synaptic behaviors of memory and erasure were demonstrated through potentiation and habituation processes.Photonic habituation was achieved using laser illumination on the porous nanowire shell,with a linear decrease in the postsynaptic current.Furthermore,synaptic elimination was emulated using two adjacent devices interconnected on a single nanowire.Therefore,electrical and photonic reconfiguration of the conductive paths in Si nanograin networks will pave the way for next-generation nanodevice technologies.

Wearable pressure sensors having versatile device structures have been extensively investigated to achieve high sensitivity under mechanical stimuli.Here,we introduce piezoelectric pressure sensors based on fabrics woven using polyvinylidene fluoride(PVDF)weft and polyethylene terephthalate(PET)warp yarns with different weave structures:1/1(plain),2/2,and 3/3 weft rib patterns.The dependence of the pressure-sensing performance on the weave pattern is demonstrated with an actual large-scale fabric up to the~2 m scale.An optimized pressure sensor with a 2/2 weft rib pattern produced a high sensitivity of 83 mV N−1,which was 245%higher than that of the 1/1 pattern.The detection performance of the optimal fabric was extensively evaluated with a variety of ambient input sources,such as pressing,bending,twisting,and crumpling,as well as various human motions.Further,a large allfabric pressure sensor with arrayed touch pixel units demonstrated highly sensitive and stable sensing performance.

HEAVY METAL POLLUTION IN SURFACE SEDIMENTS COLLECTED FROM DRAINAGES RECEIVING DIFFERENT ANTHROPOGENIC SOURCES FROM PENINSULAR MALAYSIA

Surface drainage sediments were collected, between January and April in 2005, from 6 sampling sites in Peninsular Malaysia suspected to have received different anthropogenic sources. These drainages included sites near a petrochemical plant, an electronic factory, a metal factory and 2 townships in addition to a relatively unpolluted site (a reference site). Based on the aqua-regia method, the metal concentrations for all the sampling sites except for the agricultural site (reference site) at Sg. Benut were between 60.92-1 019 μg/g dry weight for copper (Cu), 65.32-1 266.6μg/g dry weight for plumbum (Pb), 1.46-15.93 μg/g dry weight for cadmium (Cd), 330.10-484.14 μg/g dry weight for zinc (Zn) and 47.03-120.90 μg/g dry weight for nickel (Ni). All of these ranges were significantly (phong Kong by Chapman et al (1999) for the 5 metals. Based on the sequential extraction technique, 3 geochemical fractions [easily, freely leachable and exchangeable (EFLE), acid-reducible and oxidisable-organic] were also useful in identifying these polluted sites. The elevated metal concentrations found in the sediments might be related to point sources of industrial effluent and urban waste. Hence, the results emphasized the necessity for the treatment of effluents into this drainage area. As part of the effort to control illegal discharges and dumping into drainages, public awareness should be enhanced.

Synergistically tuning the graphitic degree,porosity,and the configuration of active sites for highly active bifunctional catalysts and Zn-air batteries

Rational design and tailoring of the structural features of Co-N-C catalysts are urgently required to construct highly efficient bifunctional non-noble metal electrocatalysts for both oxygen evolution reaction(OER)and oxygen reduction reaction(ORR).Herein,we report a series of carbon-based catalysts with varied structural features,specifically the graphitic degree of carbon,porosity,and the configuration of active sites,and their effects on bifunctional oxygen electrocatalytic reactions.Through the synergistic tuning of these structural factors,the well-tailored Co-N-C catalyst exhibits a high bifunctional electrocatalytic activity,as revealed by a half-wave potential of 0.84 V for ORR and a low overpotential of 420 mV at 10 mA·cm^(−2)for OER.More impressively,the Zn-air battery using the optimum catalyst delivers excellent performance including a peak power density of 125.2 mW·cm_(−2)and a specific capacity of 790.8 mAh·gZn^(−1),as well as stable cycling durability,outperforming the noble metalsbased catalysts.The first-principles calculations reveal that the interlayer interaction between the pyridinic N-doped graphene and the confined Co nanoparticles increases the electronic states of the active C atoms near the Fermi level,thus enhancing the adsorption of the hoo*intermediate and generating superior catalytic activity for bifunctional oxygen electrocatalysis.By comprehensively studying the structural factors of catalysts,the bifunctional catalytic behaviors,the use in a practical Zn-air device,and theoretical simulations,this work may also give inspirations to the design,use,and understanding of other kinds of catalysts.
