Inhibition of Proliferation of Prostate Cancer Cell Line DU-145 in vitro and in vivo Using Salvia miltiorrhiza Bunge

Objective:To investigate the antiproliferative activity of Salvia miltiorrhiza Bunge.(SM)on the castration-resistant prostate cancer(CRPC)cell line DU-145 in vitro and in vivo.Methods:Prostate cancer cell line(DU-145)and normal prostate cell line(RWPE-1)were treated with SM at different concentrations(3.125,12.5,25 and 50μg/mL)to investigate the antiproliferative effects.DNA laddering analysis was performed to investigate the apoptosis of DU-145 cells.Molecular mechanism was investigated by Western blot analysis of p53,Bcl-2,prostate specific antigen(PSA),and androgen receptor(AR).Six-week-old male BALB/c nude mice were randomly divided into normal control group(n=101)and treated group(n=101)which administered 500 mg/kg SM for 2 weeks.Tumor volumes were measured.Results:Treatment with SM resulted in a dose-dependent decrease in cell number of DU-145 cells in comparison with RWPE-1.DNA laddering analysis indicated the apoptosis of DU-145 cells.Treatment with SM increased the expression of p53 and reduced the expression of Bcl-2 proteins.The levels of PSA were considerably reduced in SM-treated group compared to the controls,and a decrease in AR expression was observed when cells were treated with SM in the same pattern as a reduction in PSA.In the tumour xenograft study,SM given once a day for 2 weeks significantly inhibited tumour growth.Conclusion:SM might contribute to the anticancer actions such as induction of apoptosis and inhibition of proliferation of prostate cancer cells.

Anti-inflammatory and Antimicrobial Effects of Anthocyanin Extracted from Black Soybean on Chronic Bacterial Prostatitis Rat Model

Objective： To investigated the anti-inflammatory and antimicrobial effects of anthocyanins extracted from black soybean on the chronic bacterial prostatitis（CBP） rat model.Methods： The SpragueDawley rats were divided into 4 groups,including control,ciprofloxacin,anthocyanins and anthocyanins with ciprofloxacin groups（n=8 in each group）.Then,drip infusion of bacterial suspension（Escherichia coli Z17O2：K1：H^-） into Sprague-Dawley rats was conducted to induce CBP.In 4 weeks,results of prostate tissue,urine culture,and histological analysis on the prostate were analyzed for each group.Results： The use of ciprofloxacin,anthocyanins,and anthocyanins with ciprofloxacin showed statistically significant decreases in bacterial growth and improvements in the reduction of prostatic inflammation compared with the control group（P〈0.05）.The anthocyanins with ciprofloxacin group showed a statistically significant decrease in bacterial growth and improvement in prostatic inflammation compared with the ciprofloxacin group（P〈0.05）.Conclusions： These results suggest that anthocyanins may have anti-inflammatory and antimicrobial effects,as well as a synergistic effect with ciprofloxacin.Therefore,we suggest that the combination of anthocyanins and ciprofloxacin may be effective in treating CBP to obtain a higher rate of treatment success.

We aimed to evaluate the current nationwide trend,efficacy,safety,and quality of life(QoL)profiles of hormone treatment in real-world practice settings for prostate cancer(PCa)patients in Korea.A total of 292 men with any biopsy-proven PCa(TanyNanyMany)from 12 institutions in Korea were included in this multi-institutional,observational study of prospectively collected data.All luteinizing hormone-releasing hormone(LHRH)agonists were allowed to be investigational drugs.Efficacy was defined as(1)the rate of castration(serum testosterone≤50 ng dl^-1)at 4-week visit and(2)breakthrough(serum testosterone>50 ng dl^-1 after castration).Safety assessments included routine examinations for potential adverse events,laboratory tests,blood pressure,body weight,and bone min era I density(BMD,at baseli ne and at the last follow-up visit).QoL was assessed using the Expa nded Prostate Cancer Index Composite-26(EPIC-26).The most comm on initial therapeutic regimen was LHRH agonist with an ti-androge n(78.0%),and the most commonly used LHRH agonist for combination and mono therapy was leuprolide(64.0%for combi natio n and 58.0%for monotherapy).The castration and breakthrough rates were 78.4%and 6.6%,respectively.The laboratory results related to dyslipidemia worsened after 4 weeks of hormone treatment.In addition,the mean BMD T-score was significantly lower at the last follow-up(mean:-1.950)compared to baseline(mean:-0.195).The mean total EPIC-26 score decreased from 84.8(standard deviation[s.d.]:12.2)to 78.3(s.d.:8.1),with significant deterioration only in the urinary domain(mean:23.5 at baseli ne and 21.9 at the 4-week visit).These findi ngs dem on strate the n ationwide trend of current practice setti ngs in hormone treatment for PCa in Korea.

Wearable pressure sensors having versatile device structures have been extensively investigated to achieve high sensitivity under mechanical stimuli.Here,we introduce piezoelectric pressure sensors based on fabrics woven using polyvinylidene fluoride(PVDF)weft and polyethylene terephthalate(PET)warp yarns with different weave structures:1/1(plain),2/2,and 3/3 weft rib patterns.The dependence of the pressure-sensing performance on the weave pattern is demonstrated with an actual large-scale fabric up to the~2 m scale.An optimized pressure sensor with a 2/2 weft rib pattern produced a high sensitivity of 83 mV N−1,which was 245%higher than that of the 1/1 pattern.The detection performance of the optimal fabric was extensively evaluated with a variety of ambient input sources,such as pressing,bending,twisting,and crumpling,as well as various human motions.Further,a large allfabric pressure sensor with arrayed touch pixel units demonstrated highly sensitive and stable sensing performance.

目的传统不带神经吻合的股外侧皮瓣会在一定程度上影响感觉的恢复,保护性感觉缺乏容易导致磨损、压疮、烧伤和冻伤等。为了克服传统股外侧皮瓣在感觉功能恢复上的缺陷,通过前瞻性研究股前外侧皮瓣修复四肢软组织缺损时,吻合股前外侧皮神经与皮瓣感觉恢复的相关关系,探究股前外侧皮瓣术中吻合神经能否对恢复皮瓣感觉有作用。方法术前制定本研究的病例纳入标准和排除标准;术中根据患者意愿和预后期望前瞻性规划带股前外侧皮神经的股前外侧皮瓣修复术(神经吻合组),及不带神经吻合的股前外侧皮瓣修复术(非吻合组);术后设计随访计划,评估患者的皮瓣感觉恢复参数(温觉、触觉和两点辨别觉),采用Semmes-Weinstein单丝触觉法和透明薄膜-扫描-电子测量方式定量分析触觉的恢复时间和恢复面积,根据Semmes-Weinstein的保护性触觉阈值标准,评估触觉恢复程度和恢复时间;采用Baseline两点辨别测试法测量两点辨别觉恢复时间和恢复比例;采用冷热试管法测量冷热觉恢复时间;我院修复重建外科自2017年4月至2021年8月共收治需皮瓣修复的四肢软组织缺损患者72例,根据术前的纳入标准和排除标准以及患者参与实验的意愿,纳入53余例,最终根据术后康复治疗和随访的依从性,完整地收集并分析30例四肢软组织缺损患者的有效数据;本研究中的软组织缺损均采用股前外侧皮瓣由同组医生手术修复,针对其中预后期望较高的14例患者采用带股前外侧皮神经的股前外侧皮瓣,术中镜下吻合股前外侧皮神经并经不同组医生检查神经吻合质量。结果30例患者的皮瓣均完全存活,未发生血管危象;术后随访9～23月,分析皮瓣感觉恢复面积所需时间:非吻合组在Semmes-Weinstein单丝触觉恢复面积为20%时所需时间为3.63±1.60月,恢复30%所需时间为4.69±2.59月,40%为5.5±3.31月,50%为6.5±3.34月,60%为7.5±4.04月,70%为7.67±1.89月,达到80%所需时间为8.62±3.31月,达到90%所需时间为13.63±3.07月,面积恢复最大值为91.75%,面积恢复最小值为68.68%,非吻合组的16例均未达到完全恢复;神经吻合组的14例在随访后均达到触觉面积完全恢复,Semmes-Weinstein单丝触觉恢复面积为20%时所需时间为2.58±1.17月,恢复30%所需时间为3.17±1.17月,40%为3.75±1.26月,50%为5.05±1.87月,60%为5.75±2.06月,70%为6.85±2.66月,80%为8.25±2.98月,90%为10.13±3.52月,100%为11.5±3.87月。以上数据行Wilcoxon秩和检验后Z=2.524,p<0.05,显示神经吻合组及非吻合组对触觉的恢复时间和恢复面积的差异具有统计学意义。对于温觉、两点辨别觉和Semmes-Weinstein保护性触觉恢复同样采用Wilcoxon秩和检验进行评估,结果示:14例神经吻合组的皮瓣在术后达到部分性保护性感觉恢复时间(Semmes-Weinstein数值<4.93)为3.57±0.79月,达到完全的保护性感觉恢复时间(Semmes-Weinstein数值<4.31)为7.75±2.87月;非吻合组达到保护性感觉部分恢复时间为6.5±1.17月,16例中无任何一例达到完全的保护性感觉恢复。吻合组和非吻合组达到部分性保护性感觉恢复时间经Wilcoxon秩和检验后Z=3.309,p<0.01,显示神经吻合组及非吻合组的对保护性触觉恢复的差异具有统计学意义。神经吻合组在4.28±1.03月后开始有冷热感受,而非吻合组在6.63±1.26月开始有冷热觉感受;神经吻合组14例在10.42±1.63月达到两点辨别觉恢复,非吻合组16例仅有7例达到两点辨别觉恢复,恢复时间为14.28±1.27月;以上数据行Wilcoxon秩和检验,Z=2.949,p<0.05(两点辨别觉);Z=2.207,p<0.05(温觉),显示神经吻合组及非吻合组对两点辨别觉和温觉的恢复差异具有统计学意义。结论采用股前外侧皮瓣修复四肢软组织缺损时,吻合股前外侧皮神经对术后皮瓣感觉的恢复有统计学差异。因此带感觉神经的股前外侧皮瓣修复术是治疗四肢缺损的理想方法之一。

Gastrointestinal health depends on the adaptive immune system tolerating the foreign proteins in food1,2.This tolerance is paradoxical because the immune system normally attacks foreign substances by generating inflammation.Here we addressed this conundrum by using a sensitive cell enrichment method to show that polyclonal CD4+T cells responded to food peptides,including a natural one from gliadin,by proliferating weakly in secondary lymphoid organs of the gut-liver axis owing to the action of regulatory T cells.

Possibility of Solid Hydrometeor Growth Zone Identification Using Radar Spectrum Width

In this study,the correlation between simulated and measured radar velocity spectrum width(σ_(v))is investigated.The results show that the dendrites growth zones(DGZs)and needles growth zones(NGZs)mostly contain dendrites(DN)and needles(NE),respectively.Clearσ_(v) zones(1.11.2 m s^(-1),a differential reflectivity(ZDR)higher than 0 dB,and a cross-correlation coefficient(ρhv)less than 0.96.In contrast,prolate particles with relatively uniform shapes were observed in the NGZ withσ_(v)<0.6 m s^(-1),a ZDR less than 0 dB,andρhv higher than 0.97.The simulation results show that the DN exhibited a largerσ_(v) compared to the NE,and this observedσ_(v) was strongly dependent on the wind fluctuations(v’)due to turbulence or wind shear.In contrast,the NE exhibited a significantly smallσ_(v)~0.55 m s^(-1),which converges irrespective of v’.In addition,a strong correlation between the measuredσ_(v) values at five radar elevation angles(θ=6.2°,9.1°,13.1°,19°,and 80°)and those simulated in this study confirmed the significance of the analysis results.

The validity of the Reynolds equation in spool valve analysis considering cavitation

The purpose of this research is to investigate the validity of the Reynolds equation in spool valve analysis under cavitation. This study is carried out for a laminar and isothermal flow between a sleeve and spool with grooves. The pressure and lateral force obtained from the Reynolds equation and the Navier-Stokes equation are compared with variations of aspect ratio, cross sectional area, and number of grooves. The cavitation phenomenon is considered by using a cavitation model in the Navier-Stokes equation and the Reynolds cavitation boundary condition in the Reynolds equation. A large difference of more than 20% is found between the equations for a lateral force of spool valve with many grooves. It was found that the Reynolds equation is not suitable for calculation of a lateral force of the spool valve with multiple-grooves of which the width and depth are even larger than the clearance under cavitation.
