In vitro and in vivo cytochrome P450 3A enzyme inhibition by Aframomum melengueta and Dennettia tripetala extracts

Objective: To evaluate the in vitro and in vivo inhibitory effects of two commonly used herbs, Aframomum melengueta(A. melengueta) and Dennettia tripetala(D. tripetala) on CYP 3A enzymes. Methods: In vitro inhibition of the enzymes were assessed with microsomes extracted from female albino rats using erythromycin-N-demethylation assay(EMND) method while their in vivo effects were measured by estimating simvastatin plasma concentrations in rats. Pharmacokinetic parameters were determined using non-compartmental anaysis as implemented in Win Nonlin pharmacokinetic program. Results: EMND assay with intestinal microsomes indicated that aqueous extracts of D. tripetala and A. melengueta significantly(P < 0.05) inhibited intestinal CYP 3A activity at both 50 μg and 100 μg concentrations. Petroleum ether extract of D. tripetala and ethanol extracts of A. melengueta inhibited intestinal CYP3 A activity at 100 μg but not at 50 μg concentrations. All the extracts showed an in vitrodose dependent CYP 3A inhibition with liver microsomes. In vivo analysis showed that pretreatment with the extracts enhanced systemic absorption of simvastatin with reductions in metabolizing enzymes activity as indicated in significant increases in maximal concentration, area under curve, area under moment curve and mean resident time of simvastatin(P < 0.05). Conclusions: Herbal preparations containing these plants' extracts should be used with caution especially in patients on CYP450 3A substrate medications.

Comparison of the thermal properties of clay samples as potential walling material for naturally cooled building design

The thermal properties of different clay samples obtained from locations in Akwa Ibom State, Nigeria were investigated and compared, and in order to establish their suitability as building material from energy conservation point of view. The results showed that stoneware clay has the highest solar radiation absorptivity of 22 32 m -1 while kaolin clay has the lowest radiation absoptivity of 14 46 m -1 A model for the prediction of temperature variation with thickness of the samples was developed. Results showed that kaolin would make the best choice for the design of a naturally cooled building.

Analyses of clay minerals within the Early Cretaceous Weald Basin,Southeast England reveal kaolinite,illite and chlorite as the main detrital clay minerals while glauconite and smectite are subordinates.A kaolinite-rich assemblage which characterized the sand-dominated Ashdown and Tunbridge Wells Sand formations and an illite-dominated assemblage associated mostly with the Wadhurst Clay and Weald Clay formations are *** was enriched in the Ashdown and Tunbridge Wells Sand formations during warm and humid climate with high precipitation that encouraged chemical weathering and leaching,while cold and dry conditions favoured the concentration of illite in the Wadhurst Clay and Weald Clay *** patterns associated with warm climate were drastically reduced during the drier climatic *** clay minerals are detrital in origin,with chlorite being more prominent than previously *** to previous studies and assumptions,this study revealed that authigenic clay minerals are present in the Hastings Beds,with vermiform and mica-replacive kaolinite being the most common,consistent with humid depositional *** authigenic illite is also present,along with a chloritized grain,providing evidence for *** absence of dickite and occurrence of kaolinite,suggest that authigenic illite formed in relatively shallow burial conditions,indicating a maximum burial depth of2500 m-3000 m,about 1000 m deeper than previous estimates of 1500 m-2000 *** clay minerals are absent in the Weald Clay Formation possibly because of hindered flow of meteoric water and limited growth space for authigenic *** study is significant in:1)reinforcing multiple methods to facilitate a robust and balanced knowledge of formation and transformation of clay minerals;2)investigating detrital and authigenic clay mineral assemblages when assessing the palaeoenvironments of sedimentary basins.

Effect of stocking density on growth performance of monosex Nile Tilapia (Oreochromis niloticus) in the aquaponic system integrated with lettuce (Lactuca sativa)

Aquaponics is a fish-plant recirculating system where nutrients received from the fish culture are absorbed by the plants for *** technology is relatively new for fish culture in Kenya,and the principles and operations remain largely untested for many fish *** study determined how stocking density affects the growth performance and water quality in a Nile tilapia-lettuce(Lactuca sativa)aquaponics *** experimental design included five replicates for each of the aquaponic systems stocked at densities of 150,300,and 450 fish/m^(3) for a rearing period of 56 *** treatment had a planting density of 16 lettuce/*** water quality parameters ranges during the rearing period were 3.83-5.35 mg/L for dissolved oxygen,7.44 to 7.6 for pH,0.014 mg/L to 0.032 mg/L for total ammonium nitrate(TAN),1.11-1.34 mg/L for nitrate,and 0.01-0.08 mg/L for nitrite,and all decreased with increasing stocking *** final weight of fingerlings was 25.2±4.2 g,32.0±3.8 g and 42.6±3.1 g for 450,300,and 150 fish/m^(3) *** growth rate(SGR)was reduced with increasing stocking density whereas food conversion ratio(FCR)increased with stocking *** systems with the lowest stocking densities performed better than 300 and 450 fish/m^(3) respectively.

Alcoholic liver disease(ALD)comprises a spectrum of liver pathology,including steatosis,steatohepatitis,and *** work from our group and others suggests that dietary fat,both the amount and composition,plays a pivotal role in ALD development and progression;however,the impact of specific dietary fatty acids on ALD pathogenesis is not fully *** rodent models of ALD revealed the deleterious effects of omega-6 polyunsaturated fatty acids(n-6 PUFAs),specifically linoleic acid(LA),and this may be partially attributed to the increased levels of pro-inflammatory oxidized LA *** is limited understanding regarding the role of omega-3 polyunsaturated fatty acids(n-3 PUFAs,such as alpha-linolenic acid,eicosapentaenoic acid,and docosahexaenoic acid),and bioactive n-3 PUFAderived lipid molecules in *** that majority of n-6 and n-3 PUFAs-derived metabolites are potent endogenous signaling molecules,knowledge regarding the changes in these lipid mediators may shed new light on the mechanisms contributing to ALD pathogenesis and reveal novel therapeutic targets and biomarkers of this *** current review summarizes relevant scientific literature regarding the role of dietary fat,distinct fatty acids,and bioactive fatty acid metabolites in ALD,and highlights recent advances in the field.

The need to increase bio-oil yield from biomass and enhance its fuel properties has driven research into optimizing the pyrolysis *** study investigated the influence of three key process parameters—temperature,heating rate,and nitrogen flow rate—on the pyrolysis of elephant grass(Pennisetum purpureum)in a fixed-bed *** surface methodology was used to study the im-pact of the aforementioned variables on bio-oil yield to improve its production *** analysis of the biomass revealed 79.24 wt%volatile matter,14.22 wt%fixed carbon,and 5.86%ash,with ultimate analysis showing 45.44%carbon,5.59%hydrogen,and 40.95%*** high volatile matter content and favourable carbon and hydrogen percentages indicate that elephant grass is a vi-able energy source due to its potential for high bio-oil yield and energy *** resulting bio-oil exhibited a higher heating value of 20.9 Mj/kg,indicating its suitability for various heating applications.A second-order regression model was developed for bio-oil yield,with optimal conditions identified as a temperature of 550°C,a heating rate of 17°C/min,and a nitrogen flow rate of 6 ml/*** study achieved an optimal bio-oil yield of 59.03 wt%,and the model’s high R²value of 0.8683 from analysis of variance analysis confirmed its predictive *** research highlights elephant grass as a sustainable feedstock for bio-oil production,offering valuable insights into optimizing pyrolysis conditions to enhance bio-oil yield,thus advancing biofuel technology.
