The membrane-associated transcription factor, bZlP28, is relocated from the endoplasmic reticulum （ER） to the Golgi and proteolytically released from the membrane mediated by two proteases, SlP and S2P, in response to ER stress in Arabidopsis. The activated N-terminal domain recruits nuclear factor Y （NF-Y） subunits in the nucleus to regulate ER stress downstream genes. Little is known about the functions of the bZIP28 C-terminal lumen-facing domain. Here, we provide novel insights into how the ER lumen-facing domain affects the biological function and organelle-to-organelle movement of bZIP28 in the ER stress response. First, we demonstrated the functional redundancy of bZlP28 and bZIP60 by generation and analysis of the bZIP28 and bZIP60 double mutant zip28zip60. Subsequent genetic complementation experiments in zip28zip60 background with deletions on bZlP28 lumen-facing domain highlighted the importance of lumen-facing domain for its in vivo function of bZIP28 in the ER stress response. The protein subcellular localization and Western blotting results further revealed that the bZIP28 lumen-facing domain contains ER retention signal which is important for the proteolytic activation of bZIP28. Thus, the bZIP28 lumen-facing C-terminus plays important roles in the ER-to-Golgi movement of bZlP28, which may contribute to the sensing of the ER stress.

Soil salinity is a widespread abiotic stress constraint threatening agricultural production, as it severely inhibits growth and development of crops. Several salt stress signaling pathways have been discovered in the model plant Arabidopsis thaliana, which include components for sensing the stress, transmitting the signal and regulating the downstream genes （Deinlein et al. 2o14; Julkowska and Testerink 2015）. In nature, plant roots directly encounter heterogeneous soils and therefore their responses to saline environments require dynamic changes in growth, a process in which plant cell walls play important roles （Tenhaken 2o15）. However, the molecular mechanisms underlying cell wall remodeling under stress conditions are still unclear.

Protein folding in the endoplasmic reticulum （ER） is a fundamental process in plant cells that is vulnerable to many environmental stresses. When unfolded or misfolded proteins accumulate in the ER, the well-conserved unfolded protein response （UPR） is initiated to mitigate the ER stress by enhancing the protein folding capability and/or accelerating the ER-associated protein degradation. Here, we report the conservation of the activation mechanism of OsbZIP74 （also known as OsbZIP50）, an important ER stress regulator in monocot plant rice （Oryza sativa L.）. Under normal conditions, OsbZIP74 mRNA encodes a basic leucine-zipper transcription factor with a putative transmembrane domain. When treating with ER stress-inducing agents such as tunicamycin and DTT, the conserved double stem-loop structures of OsbZIP74 mRNA are spliced out. Thereafter, the resulting new OsbZIP74 mRNA produces the nucleus-localized form of OsbZIP74 protein, eliminating the hydrophobic region. The activated form of OsbZIP74 has transcriptional activation activity in both yeast cells and Arabidopsis leaf protoplasts. The induction of OsbZIP74 splicing is much suppressed in the OsIRE1 knock- down rice plants, indicating the involvement of OslRE1 in OsbZIP74 splicing. We also demonstrate that the unconventional splicing of OsbZIP74 mRNA is associated with heat stress and salicylic acid, which is an important plant hormone in systemic acquired resistance against pathogen or parasite.

Consolidation of high replacement ratio stone column-reinforced ground:Analytical solutions incorporating clogging effect

The utilization of stone columns has emerged as a popular ground improvement strategy,whereas the drainage performance can be adversely hampered by clogging *** the ample progress of calculation methods for the consolidation of stone column-improved ground,theoretical investigations into the clogging effect have not been thoroughly ***,it is imperative to involve the column consolidation deformation to mitigate computational error on the consolidation of composite ground with high replacement *** this context,an analytical model accounting for the initial clogging and coupled time and depth-dependent clogging of stone columns is ***,the resulting governing equations and analytical solutions are obtained under a new flow continuity relationship to incorporate column consolidation *** accuracy and reliability of the proposed model are illustrated by degradation analysis and case studies with good ***,the computed results of the current study are juxtaposed against the existing models,and an in-depth assessment of the impacts of several crucial parameters on the consolidation behavior is *** results reveal that ignoring column consolidation deformation leads to an overestimate of the consolidation rate,with maximum error reaching up to 16%as the replacement ratio ***,the initial clogging also has a significant influence on the consolidation ***,the increment of depth and time-clogging factors a and b will induce a noticeable retardation of the consolidation process,particularly in the later stage.

Virtual synchronous generators(VSGs)are widely introduced to the renewable power generation,the variablespeed pumped storage units,and so on,as a promising gridforming *** is noted that VSGs can provide virtual inertia for frequency support,but the larger inertia would worsen the synchronization stability,referring to keeping synchronization with the grid during voltage ***,this paper presents a transient damping method of VSGs for enhancing the synchronization stability during voltage *** is revealed that the loss of synchronization(LOS)of VSGs always accompanies with the positive frequency deviation and the damping is the key factor to remove LOS when the equilibrium point *** order to enhance synchronization stability during voltage dips,the transient damping is proposed,which is generated by the frequency deviation in active power ***,the proposed method can realize seamless switching between normal state and grid ***,detailed control design for transient damping gain is given to ensure the synchronization stability under different inertia requirements during voltage ***,the experimental results are presented to validate the analysis and the effectiveness of the improved transient damping method.

Grain size and shape are important determinants of grain weight and yield in rice. Here, we report a new major quantitative trait locus （QTL）, qTGW3, that controls grain size and weight in rice. This locus, qTGW3, encodes OsSK41 （also known as OsGSK5）, a member of the GLYCOGEN SYNTHASE KINASE 3/SHAGGY-like family. Rice near-isogenic lines carrying the loss-of-function allele of OsSK41 have increased grain length and weight. We demonstrate that OsSK41 interacts with and phosphorylates AUXIN RESPONSE FACTOR 4 （OsARF4）. Co-expression of OsSK41 with OsARF4 increases the accumulation of OsARF4 in rice protoplasts. Loss of function of OsARF4 results in larger rice grains. RNA-sequencing analysis suggests that OsARF4 and OsSK41 repress the expression of a common set of downstream genes, including some auxin-responsive genes, during rice grain development. The loss-of-function form of OsSK41 at qTGW3 represents a rare allele that has not been extensively utilized in rice breeding. Suppression of OsSK41 function by either targeted gene editing or QTL pyramiding enhances rice grain size and weight. Thus, our study reveals the important role of OsSK41 in rice grain development and provides new candidate genes for genetic improvement of grain yield in rice and perhaps in other cereal crops.

Ethane chemical looping oxidative dehydrogenation(CL-ODH)to ethylene is a new technology for ethylene ***_(2)O_(3)/MgO oxygen carrier was prepared using the co-precipitation *** influence of added NiO and its different loadings on Fe_(2)O_(3)/MgO were ***,a series of oxygen carriers were applied in the CL-ODH of the ethane cycle ***-Emmett-Teller(BET),X-ray diffractometry(XRD),X-ray photoelection spectroscopy(XPS),and H2-temperature programmed reduction(TPR)were used to characterize the physicochemical properties of these oxygen *** was confirmed that an interaction between NiO and Fe_(2)O_(3) occurred based on the XPS and H2-TPR *** on the CL-ODH activity performance tests conducted in a fixed-bed reactor,it was revealed that ethylene selectivity was significantly improved after NiO ***_(2)O_(3)-10%NiO/MgO showed the best activity performance with 93%ethane conversion and 50%ethylene selectivity at a reaction temperature of 650℃,atmospheric pressure,and space velocity of 7500 mL/(g·h).

The rapid development of the anti-missile weapon technology brings new challenges to the cooperative penetration strategy solution and the guidance law design for Hypersonic Vehicles(HVs).This paper studies the coordinated game penetration guidance problem for multiple hypersonic vehicles faced with space threat areas.A scheme for seeking cooperative game penetration guidance strategy under safety critical control framework is *** this scheme,a multiHV cooperative game model is proposed in a minimum optimization form which can simplify the solving process and accelerate the computing ***,a second-order control barrier function is developed to transform the implicit nonlinear constraints of the proposed model into linear *** order to obtain better performance of guidance strategy,a composite guidance law under the safety critical control framework is presented to allocate guidance strategies appropriately in the whole *** is shown that the proposed scheme can guarantee successful penetration while avoiding threat ***,a comparative simulation with a two-on-three game is conducted to verify the effectiveness of the proposed method.

The actively heated fiber-optic(AHFO)technology has become emerged as a research focus due to its advantages of distributed,real-time measurement and good *** attributes have led to the gradual application of AHFO technology to the water content measurement of in situ ***,all existing in situ applications of AHFO technology fail to consider the effect of soilesensor contact quality on water content measurements,limiting potential for the wider application of AHFO *** address this issue,the authors propose a method for determining the soilesensor thermal contact resistance based on the principle of an infinite cylindrical heat *** is then used to establish an AHFO water content measurement technology that considers the thermal contact *** reliability and validity of the new measurement technology are explored through a laboratory test and a field case study,and the spatial-temporal evolution of the soil water content in the case is *** results demonstrate that method for determining the soilesensor thermal contact resistance is highly effective and applicable to all types of *** method requires only the moisture content,dry density,and thermal response of the in situ soil to be *** the field case,the measurement error of soil water content between the AHFO method,which takes into account the thermal contact resistance,and the neutron scattering method is only *** water content of in situ soil exhibits a seasonal variation,with an increase in spring and autumn and a decrease in summer and ***,the response of shallow soils to precipitation and evaporation is *** findings contribute to the enhancement of the accuracy of the AHFO technology in the measurement of the water content of in situ soils,thereby facilitating the dissemination and utilization of this technology.
