Clinical lesson learned from genetic analysis in patients prior to surgical repair of hypospadias

In Indonesia,undervirilisation in 46,XY males is the most common form of difference of sex development(DSD).This can include hypospadias(misplacement of the urethra),micropenis,bifid scrotum,and undescended testis[1].Undervirilisation or 46,XY DSD can be associated with a number of congenital syndromes,including Smith-Lemli-Opitz Syndrome(OmIm 602858),caused by an inborn error of cholesterol synthesis,and characterised by growth delay,intellectual disability,microcephaly,distinctive facial features,cleft palate,limb anomalies,and hypospadias[2]or Opitz syndrome(also known as Opitz G/BBB syndrome).Opitz syndrome can be caused by variants in the X-linked midline 1(mID1)gene(Type I)or in an autosomal dominant manner by monoallelic variants in sperm antigen with calponin homology and coiled-coil domains 1-like(SPECC1L)on chromosome 22q11.2(Type II)[3].Opitz syndrome is characterised by hypospadias,hypertelorism,cleft lip/palate,and heart defects[4].The prevalence of X-linked Opitz syndrome is estimated to be from 1 in 50000 to 1 in 100000 males[5].Recognition of a syndrome informs appropriate clinical management and patient ***,although these syndromes are rare,hypospadias may be diagnosed before the emergence of other comorbidities meaning that it is crucial for clinicians to perform a thorough clinical evaluation with syndromic causes in mind.

Fluoride is an impurity in nickel sulfate production,which is required for electric vehicle *** zirconium oxide(hZO)was evaluated for removing fl uoride from nickel sulfate *** fluoride removal occurred at ph value 4 and optimal ph value is 4–5,considering Zr *** availability decreases with ph due to hydrogen fluoride and zirconium fluoride aqueous *** removal is initially rapid,with 50 wt.%removal in 7 min,followed by slow removal up to 68 wt.%after 72 h and follows second order rate *** removal was dominated by an ion exchange mechanism and resulting Zr–F bonds were observed using Fourier-transform infrared *** presence of nickel sulfate decreased loading capacity compared to a salt-free *** maintained adsorption capacity through five cycles of loading and regeneration.

Systematic mutational analysis of epitope-grafted ED3 immunogenicity reveals a DENV3-DENV4 bi-serospecific ED3 mutant

Objective:To identify the residue determinants of the serospecificity and sero-cross-reactivity of dengue virus(DENV)envelope protein domain 3(ED3),which contains two major putative epitopes of ***:We constructed ED3 from DENV3(3ED3)and DENV4(4ED3),and six epitope-grafted variants,where we transferred epitope 1(L304I,K305D,V309m,and S310A)and/or epitope 2(D383N,K384S,K387T,and N389h)of 4ED3 onto 3ED3 and *** albino mice aged 3-4 weeks were immunized against wildtype and epitope-grafted ED3 variants and anti-ED3 IgG antibody responses were determined using ***:mouse immunization using 3ED3 and 4ED3 generated serotype-specific antisera,as ***,most epitope-grafted ED3s produced antisera serospecific to the template ED3 with little or no cross-recognition of ED3 of the serotype from which the epitopes were *** indicated that a mere grafting of the epitope was not sufficient to transfer serospecificity,contrary to our ***,one epitope-grafted ED3 mutant,where epitope 1 of 3ED3 was grafted onto 4ED3(4ED3^(epi1)),generated antisera that was serospecific to both 4ED3 and ***:The 4ED3^(epi1)is a chimeric ED3 that produces antisera possessing serospecificity to both 3ED3 and 4ED3 onto a common 4ED3 *** 4ED3^(epi1),therefore,provides a unique tool for analyzing serospecificity and sero-cross-reactivity in *** believe that chimeric ED3 may provide a template for future recombinant ED3 possessing serospecificity of multiple DENVs onto a single scaffold and may pave a way developing tri-and/or tetravalent anti-DENV antisera.

metal complexes of ciprofloxacin were synthesized to study the effect of the drug on the metals present in the body or metals co-administered as multivitamin therapy. These complexes were then characterized by IR, UV, 1 h NmR, ChN and AA analyses. It was found that the metals coordinated through the β-keto carboxylic group, forming four coordinated complexes having square planar geometry. The effect of the antibacterial activity of these complexes with respect to parent drug was also studied against 14 different Gram + ve and Gram – ve organisms by the disk susceptibility technique. Some complexes showed improve activity as compared with the stated drug.

Analysis of Immune Responses and Serological Cross Reactivities among Vibrio cholerae O1,Shigella flexneri 2a and haemophilus influenzae b

Antigenic determinants expressed on the bacterial cell surface are of importance in the serological characterization and microbiological diagnosis. The bacterial strains carrying these identical or similar antigenic epitopes might react with antibodies produced against other strains. In this study, strong immunogenicity and antigenic cross reactivity were demonstrated among V. cholerae O1, S. flexnerii 2a and h. influenzae b surface components. The enzyme linked immunosorbent assay （ELISA） results were supported by Western blot analysis, where at least 20 antigenic bands, were obtained in each of the reactions, when the surface components were reacted with the homologous antisera. The indirect ELISA results also demonstrated high degree of antigenic relatedness between the surface components of these species, where each surface component was reacted with the heterologous antisera. Western blot analysis also revealed cross reactions between the surface components suggesting common distribution of antigens/epitopes in these bacterial species. This study, thus, gave a clear idea of the level of antigenic sharing and variations among the pathogenic V. cholerae O1, S. flexneri 2a and h. influenzae b strains, which in future, may help in selecting a proper candidate for vaccines and immunodiagnostics development.

Background We aimed to systematically review the clinical and laboratory features of patients with the multisystem inflam-matory syndrome in pediatrics diagnosed during the COVID-19 *** sources A literature search in Web of Science,Pubmed,Scopus,and Science Direct was made up to June 29,*** Analysis of 15 articles(318 COVID-19 patients)revealed that although many patients presented with the typical multisystem inflammatory syndrome in pediatrics,Kawasaki-like features as fever(82.4%),polymorphous maculopapular exanthema(63.7%),oral mucosal changes(58.1%),conjunctival injections(56.0%),edematous extremities(40.7%),and cervical lymphadenopathy(28.5%),atypical gastrointestinal(79.4%)and neurocognitive symptoms(31.8%)were also *** had elevated serum lactic acid dehydrogenase,D-dimer,C-reactive protein,procalcitonin,interleukin-6,troponin I levels,and *** 77.0%developed hypotension,and 68.1%went into shock,while 41.1%had acute kidney *** care was needed in 73.7%of cases;13.2%were intubated,and 37.9%required mechanical *** immunoglobulins and steroids were given in 87.7%and 56.9%of the patients,respectively,and anticoagulants were utilized in 67.0%.Pediatric patients were discharged after a hospital stay of 6.77 days on average(95%CI 4.93-8.6).Conclusions Recognizing the typical and atypical presentation of the multisystem inflammatory syndrome in pediatric COVID-19 patients has important implications in identifying children at *** cardiac and renal decompensation and early interventions in patients with multisystem inflammatory syndrome is critical to prevent further morbidity.

The Impacts of Sea Level Rise on Basrah City, Iraq

Sea Level Rise (SLR) above the mean Sea Level (mSL) is more likely to cause a significant risk to the coastal regions. This research explores the potential impact of sea level rise, due to climate change, on coastal areas. It examines the impact of sea level rise on Basrah city and adjacent cities in Iraq. A digital elevation model (DEm) was used to create a model of Potentially Inundated Areas, manipulated and processed in Geographical Information System version 10.7 (ArcGIS 10.7). Through this model, the impact of sea level rise was assessed on the surface area. After the susceptible areas were delineated, it was estimated that at worst case scenario of 5 m sea level rise will impact Basrah city by losing 38 percent of its total surface area.

Selective C3-alkenylation of oxindole with aldehydes using heterogeneous CeO2 catalyst

We report herein that a commercially available CeO2 is an active and reusable catalyst for the C3-selective alkenylation of oxindole with aldehydes under solvent-free conditions. This catalytic method is generally applicable to different aromatic and aliphatic aldehydes, giving 3-alkyledene-oxindoles in high yields(87%–99%) and high stereoselectivities(79%–93% to E-isomers). This is the first example of the catalytic synthesis of 3-alkenyl-oxindoles from oxindole and various aliphatic aldehydes. The Lewis acid-base interaction between Lewis acid sites on CeO2 and benzaldehyde was studied by in situ IR. The structure-activity relationship study using CeO2 catalysts with different sizes suggests that defect-free CeO2 surface is the active site for this reaction.

Impact of hepatitis C virus core mutations on the response to interferon-based treatment in chronic hepatitis C

AIm To determine whether hepatitis C virus(hCV) core substitutions play a role in the response to interferon-based treatment in Caucasian patients. mEThODS One hundred eight hCV chronically infected patients initiating treatment with pegylated IFN plus ribavirin for 48 wk were tested for baseline substitutions at codons 70 and 91 of the viral core protein(Big Dye Terminator vers.3.1, Applied Biosystems,) and for genetic polymorphisms in host IL28 B gene rs12979860(Custom Taqm an 5' allelic discrimination assay; Applied Biosystems).RESULTS Of the patients, all were infected with hCV genotype 1b, 44.4% had low baseline hCV viral load, and 37.9% had mild/moderate fibrosis. Only 38.9% achieved therapeutic success, defined as sustained virological response(SVR). Eighty-eight percent of the patients presented at least one substitution at core position 70(R70Q/h) or/and position 91(L91m). The favorable IL28 B CC polymorphism was detected in only 17.6% of the patients. In the univariate analysis, young age(P mutations(P = 0.005), achievement of rapid virologic response(P h SVR. A multivariate analysis revealed three independent predictors of therapeutic success: young age(P he model correctly classified 75.9% of SVR cases with a positive predictive value of 80.7%. CONCLUSION hCV core mutations can help distinguish between patients who can still benefit from the affordable IFNbased therapy from those who must be treated with DAAs to prevent the evolution towards end-stage liver disease.

Cloning of Three Antiporter Genes from Arabidopsis and Rice for Over-Expressing Them in Farmer Popular Tomato Varieties of Bangladesh

Salinity is one of the most critical environmental problems, which causes plant growth retardation by disturbing intracellular ion homeostasis. The Na+/h+ antiporter plays an important role in resistance to salt stress by sequestering Na+ in exchange for h+ across the vacuolar membranes. In the current study, the coding regions of two Arabidopsis antiporters (AtNhX1 and AtNhX2) and one rice antiporter (OsNhX1) were amplified by target specific PCR. PCR amplicons were first cloned into pENTR/D-TOPO and later recombined with a destination vector (pK7WG2.0) by LR reaction. Positive clones were selected by PCR, restriction digestion (RD) and sequencing. They were then transformed into Agrobacterium tumefaciens (LBA4404 strain) for subsequent transformation of farmer popular tomato varieties.
