Lacidipine,thiamine pyrophosphate and their combination on the ocular ischemic syndrome induced by bilateral common carotid artery ligation

AIM:To investigate the effect of lacidipine,thiamine pyrophosphate(TPP)and the combination of lacidipine and TPP against oxidative and inflammatory eye damage induced by bilateral common carotid artery ligation in ***:Male albino Wistar rats were categorized as those who underwent sham surgery(SG),right and left common carotid cross-clamping and unclamping procedure(CCU),lacidipine+CCU(LCCU),TPP+CCU(TCCU),and combination of lacidipine and TPP(LTC)+CCU(LTCCU).One hour before anesthesia,the LCCU(n=6)received lacidipine(4 mg/kg,orally)and the TCCU(n=6)received TPP(20 mg/kg,intraperitoneally).The SG(n=6)and CCU(n=6)received the same volume of distilled water from the same *** anesthesia(60 mg/kg ketamine,intraperitoneally),the necks of the rats were opened in the *** was created for 10min by placing clips on the right and left common carotid *** in the SG only underwent subcutaneous *** 10min,the clips were removed and reperfusion was achieved for six ***,the animals were euthanized(120 mg/kg ketamine,intraperitoneally)and the levels of oxidant,antioxidant and proinflammatory cytokines in the eye tissues were *** retinal tissue of the eye was also examined ***:Lacidipine,TPP,and LTC significantly prevent the increase in malondialdehyde,tumor necrosis factoralpha,interleukin-1β(IL-1β),and IL-6 levels,decrease in total glutathione levels,superoxide dismutase and catalase activities and histopathological retinal damage in eye tissue induced by bilateral common carotid artery ligation in *** impact of these drugs on protection is determined to be LTC>lacidipine>***:As a result of the study,it is concluded that LTC may be more effective than lacidipine and TPP alone in treating ocular ischemic syndrome.

In recent years,multiple advances have been made in understanding the photosynthetic machinery in model *** transfer to horticultural important fruit crops is challenging and time-consuming due to restrictions in gene editing tools and prolonged life ***,we characterize a gene encoding a PetM domain-containing protein in *** CRISPR/Cas9 knockout lines of the PetM showed impairment in the chloroplastic electron transport rate(ETR),reduced CO_(2) assimilation,and reduction of carotenoids and chlorophylls(Chl)under several light ***,growth-condition-dependent elevation or repression of Chl a/b ratios and de-epoxidation states were identified,underlining possible impairment compensation ***,under low light and glasshouse conditions,there were basal levels in CO_(2) assimilation and ETR,indicating a potential role of the PetM domain in stabilizing the cytochrome b6f complex(Cb6f)under higher light irradiance and increasing its quantum *** suggests a potential evolutionary role in which this domain might stabilize the site of the Cb6f regulating ratios of cyclic and linear electron transport and its potential importance during the conquest of terrestrial ecosystems during which plants were exposed to higher ***,the results are discussed with regard to metabolism and their implication to photosynthesis from an agronomic perspective.

Background:High-intensity interval training(HIIT)induces similar or even superior adaptations compared to continuous endurance ***,just 6 HIIT sessions over 2 weeks significantly improves maximal oxygen uptake(VO_(2max)),submaximal exercise fat oxidation,and endurance *** even faster adaptations can be achieved with HIIT is not ***,we aimed to determine whether 2 sessions of HIIT per day,separated by 3 h,every other day for 5 days(double HIIT(HIIT-D),n=15)could increase VO_(2max),submaximal exercise fat oxidation,and endurance capacity as effectively as 6 sessions of HIIT over 2 weeks(single HIIT(HIIT-S),n=13).Methods:Each training session consisted of 10×60 s of cycling at 100%of VO_(2max)interspersed with 75 s of low-intensity cycling at 60 watt(W).Pre-and post-training assessments included VO_(2max),time to exhaustion at-80%of VO_(2max),and 60-min cycling trials at-67%of VO_(2max).Results:Similar increases(pn VO_(2max)(HIIT-D:7.7%***-S:6.0%,p>0.05)and endurance capacity(HIIT-D:80.1%***-S:79.2%,p>0.05)were *** exercise carbohydrate oxidation was reduced in the 2 groups after exercise training(HIIT-D:9.2%,p=0.014 ***-S:18.8%,p=0.012)while submaximal exercise fat oxidation was significantly increased in HIIT-D(15.5%,p=0.048)but not in HIIT-S(9.3%,p=0.290).Conclusion:Six HIIT sessions over 5 days was as effective in increasing VO_(2max)and endurance capacity and was more effective in improving submaximal exercise fat oxidation than 6 HIIT sessions over 2 weeks.

Determination of Some Trace Metals in Environmental Samples by Flame AAS Following Solid Phase Extraction with Amberlite XAD-2000 Resin after Complexing with 8-Hydroxyquinoline

A procedure for preconcentration of Mn（Ⅱ）, Fe（Ⅱ）, Co（H）, Cu（Ⅱ）, Cd（Ⅱ）, Zn（Ⅱ）, Pb（Ⅱ） and ni（Ⅱ） based on retention of their complexes with 8-hydroxyquinoline （HQ） on Amberlite XAD-2000 resin in a column was proposed for the analysis of environmental samples by flame AAS. Various parameters such as pH, eluent type, volume, concentration, flow rate and volume of sample solution, and matrix interference effect on the retention of the metal ions were investigated. The optimum pHs for the retention of metal complexes in question were about 6 except for Mn^2＋ for whose value is 8. The loading capacity of the adsorbent for these metals and their recoveries from the resin under the optimum conditions were in the range 6.82-9.26 mg·g^-1 and 95%-101%, respectively. The enrichment factor was calculated as 100 and the limit of detection was in the range 0.3-2.2 μg·L^-1 （n=20, blank＋ 3s）. The proposed enrichment method was applied to tap water, stream water and vegetable samples. The validation of the procedure was carried out by analysis of certified reference material and standard addition. The analytes were determined with a relative standard deviation lower than 6% in all samples.

Effect of sodium tungstate on visual evoked potentials in diabetic rats

AIM： To evaluate the effect of sodium tungstate on visual evoked potentials（VEPs） in diabetic rats.· METHODS： Wistar rats were randomly divided into three groups as normal control, diabetic control and diabetic rats treated with sodium tungstate. Diabetes was induced by single intraperitoneal injection of streptozotocin（50 mg/kg）. Sodium tungstate [40 mg/（kg·d）]was administered for 12 wk and then VEPs were ***, thiobarbituric acid reactive substance（TBARS） levels were measured in brain tissues.·RESULTS： The latencies of P1, n1, P2, n2 and P3 waves were significantly prolonged in diabetic rats compared with control group. Diabetes mellitus caused an increase in the lipid peroxidation process that was accompanied by changes in VEPs. However, prolonged latencies of VEPs for all components returned to control levels in sodium tungstate-treated group. The treatment of sodium tungstate significantly decreased brain TBARS levels and depleted the prolonged latencies of VEP components compared with diabetic control group.· COnCLUSIOn： Sodium tungstate shows protective effects on visual pathway in diabetic rats, and it can be worthy of further study for potential use.

Background:Cholangiocarcinoma(CCA)is a cancer type that builds in the bile ducts that carry digestive fluid,*** ducts connect the liver to the gallbladder and the small *** disease is often diagnosed at an advanced stage,resulting in a low 5-year survival *** study aims to evaluate the concentrations of serum thiol/disulfide in CCA patients and healthy volunteers and investigate the association between oxidative activity and clinical and pathological characteristics in CCA *** examining the relationship between reduced thiol/disulfide measures and tumorigenesis of the disease,we can potentially identify an unfavourable prognosis in CCA ***:To assess the status of thiol/disulfide in the blood of Cholangiocarcinoma patients using a novel automated homeostasis assay,we recruited 55 individuals for this study(CCA patients,n=27;healthy volunteers,n=28).We measured the levels of serum total thiol(TT)(–SH+–S-S–),native thiol(nT)(–SH),and disulfide(DD)(–S-S–)in both the CCA group and the control ***,we calculated the ratio of thiol/disulfide(–SH/–S-S–).Results:We explored the relationship between oxidative activity and clinical and pathological characteristics in CCA *** study found no significant differences in age and sex between CCA patients and *** and TT levels were higher in the control group compared to the CCA ***,the disulfide level did not significantly differ between the two ***'s correlation matrix analysis revealed positive correlations between TT and nT levels and negative correlations between nT/TT%,DD level,and DD/TT%.Conclusion:Our findings suggest a statistical difference in serum thiol/disulfide *** a marker of total oxidant status,thiol and disulfide levels have decreased in patients with ***,no correlation was found between the tumour markers CEA and CA19-9 and thiol levels.A decrease in blood thiol and disulfide concentrations or the ratio

*** 2003 we identified a family with familial hypocalciuric hypercalcemia(FHH)(heterozygous CASR gene mutation L173P)and a mutation in the pancreatic secretory trypsin inhibitor gene(SPInK1)(n34S).While family members with an isolated calcium-sensing receptor gene(CASR)mutation remained healthy,a combination of the CASR and SPInK1 gene mutation caused chronic pancreatitis(CP).We thus speculate that the combination of two genetic defects affecting calcium homeostasis and pancreatic enzyme activation might represent a novel approach in chronic inherited pancreatic *** therefore sought to explore whether CASR gene mutations were prevalent in a cohort of patients with CP and confirmed SPInK1 *** and methods.A cohort of 19 families(n = 170)with a history of idiopathic CP(ICP)was screened for mutations within the CASR gene;104 members of that cohort had a mutation(n34S)within the SPInK1 gene and 66 of those were suffering from *** entire CASR gene was screened for single strand conformation polymorphism under varying polyacrylamide gel conditions and subjected to direct dideoxy nucleotide sequencing of amplified ***-strand conformation polymorphisms were observed in 59 samples,clustering of exons 3,4 and *** sequence analysis revealed a yet unreported missense mutation in exon 7(R896H)and two conservative mutations in exon 4(F391F)and exon 7(E790E).Furthermore,an intronic polymorphism in nucleotide position 493-19 G > A was detected in 19 out of 170 members of that *** identified three novel calcium-sensing receptor gene mutations(1 missense mutation,2 silent mutations and 1 intronic polymorphism)in a cohort of 19 families with *** particular,the kindred with the R896H mutation presenting with a similar pedigree to the family described above may indicate a role for CASR gene mutations in SPInK1-related ***,only the patient with the combination of both CASR and n34S SPInK1 gene mutation developed
