B cells have an essential immunoregulatory function,which is mediated by the secretion of IL-10 and TGFβand interaction with activated CD4^(+)T cells.This antibody-independent function of B cells contributes to ameliorating autoimmune diseases or infection-induced tissue damage.In this Cellular and Molecular Immunology issue,Fu et al.demonstrated that regulatory B(Breg)cells play a critical role in resolving Pseudomonas aeruginosa-induced pneumonia[1].

In contrast to the previous belief that autoreactive B cells are eliminated from the normal repertoire of B cells,many autoreactive B cells actually escape clonal deletion and develop into mature B cells.These autoreactive B cells in healthy individuals perform some beneficial functions in the host and are homeostatically regulated by regulatory T and B cells or other mechanisms to prevent autoimmune diseases.Autoreactive B-1 cells constitutively produce polyreactive natural antibodies for tissue homeostasis.Recently,autoreactive follicular B cells were reported to participate actively in the germinal center reaction.Furthermore,the selection and usefulness of autoreactive marginal zone(MZ)B cells found in autoimmune diseases are not well understood,although the repertoire of MZ B-cell receptors(BCRs)is presumed to be biased to detect bacterial antigens.In this review,we discuss the autoreactive B-cell populations among all three major B-cell subsets and their regulation in immune responses and diseases.

We investigated the occurrence and distribution of terrestrial-derived hydroxylated isoprenoid glycerol dialkyl glycerol tetraethers(OH-GDGTs) in the Han River system and their potential impact on the application of the ring index of OH-GDGTs(RI-OH) as a sea surface temperature(SST) proxy in the eastern Yellow Sea. Thereby, we analyzed various samples collected along the Han River and from its surrounding areas(South Korea, n = 34). The OHGDGTs were found in all samples investigated. OH-GDGT-0 was the dominant OH-GDGT component in the estuary and marine samples while OH-GDGT-2 was generally dominant in the soils, the lake sediments and the river suspended particulate matter(SPM). Our results thus suggests a possible warm bias of the RI-OH-derived summer SSTs in the coastal zone to which a large amount of terrestrial organic matter is being supplied. Further studies are necessary to better assess the applicability of the RI-OH proxy in the eastern Yellow Sea.

Alzheimer's disease(AD)is the most serious age-related neurodegenerative disease and causes destructive and irreversible cognitive decline.Failures in the development of therapeutics targeting amyloid-β(Aβ)and tau;principal proteins inducing pathology in AD,suggest a paradigm shift towards the development of new therapeutic targets.The gram-negative bacteria and lipopolysaccharides(LPS)are attractive new targets for AD treatment.Surprisingly,an altered distribution of gram-negative bacteria and their LPS has been reported in AD patients.Moreover,gram-negative bacteria and their LPS have been shown to affect a variety of AD-related pathologies,such as Aβ homeostasis,tau pathology,neuroinflammation,and neurodegeneration.Moreover,therapeutic approaches targeting gram-negative bacteria or gram-negative bacterial molecules have significantly alleviated AD-related pathology and cognitive dysfunction.Despite multiple evidence showing that the gram-negative bacteria and their LPS play a crucial role in AD pathogenesis,the pathogenic mechanisms of gram-negative bacteria and their LPS have not been clarified.Here,we summarize the roles and pathomechanisms of gram-negative bacteria and LPS in AD.Furthermore,we discuss the possibility of using gram-negative bacteria and gram-negative bacterial molecules as novel therapeutic targets and new pathological characteristics for AD.

Marginal zone(MZ)B cells,which are splenic innate-like B cells that rapidly secrete antibodies(Abs)against blood-borne pathogens,are composed of heterogeneous subpopulations.Here,we showed that MZ B cells can be divided into two distinct subpopulations according to their CD80 expression levels.CD80^(high)MZ B cells exhibited greater Ab-producing,proliferative,and IL-10-secreting capacities than did CD80^(low)MZ B cells.Notably,CD80^(high)MZ B cells survived 2-Gy whole-body irradiation,whereas CD80^(low)MZ B cells were depleted by irradiation and then repleted with one month after irradiation.Depletion of CD80^(low)MZ B cells led to accelerated development of type II collagen(CII)-induced arthritis upon immunization with bovine CII.CD80^(high)MZ B cells exhibited higher expression of genes involved in proliferation,plasma cell differentiation,and the antioxidant response.CD80^(high)MZ B cells expressed more autoreactive B cell receptors(BCRs)that recognized double-stranded DNA or CII,expressed more immunoglobulin heavy chain sequences with shorter complementarity-determining region 3 sequences,and included more clonotypes with no N-nucleotides or with B-1a BCR sequences than CD80^(low)MZ B cells.Adoptive transfer experiments showed that CD21^(+)CD23^(+)transitional 2 MZ precursors preferentially generated CD80^(low)MZ B cells and that a proportion of CD80^(low)MZ B cells were converted into CD80^(high)MZ B cells;in contrast,CD80^(high)MZ B cells stably remained CD80^(high)MZ B cells.In summary,MZ B cells can be divided into two subpopulations according to their CD80 expression levels,Ab-producing capacity,radioresistance,and autoreactivity,and these findings may suggest a hierarchical composition of MZ B cells with differential stability and BCR specificity.

Abstinence from prolonged psychostimulant use prompts stimulant withdrawal syndrome.Molecular adaptations within the dorsal striatum have been considered the main hallmark of stimulant abstinence. Here we explored striatal miRNA-target interaction and its impact on circulating miRNA marker as well as behavioral dysfunctions in methamphetamine(MA) abstinence. We conducted miRNA sequencing and profiling in the nonhuman primate model of MA abstinence, followed by miRNA qPCR,LC-MS/MS proteomics, immunoassays, and behavior tests in mice. In nonhuman primates, MA abstinence triggered a lasting upregulation of miR-137 in the dorsal striatum but a simultaneous downregulation of circulating miR-137. In mice, aberrant increase in striatal miR-137-dependent inhibition of SYNCRIP essentially mediated the MA abstinence-induced reduction of circulating miR-137. Pathway modeling through experimental deduction illustrated that the MA abstinence-mediated downregulation of circulating miR-137 was caused by reduction of SYNCRIP-dependent miRNA sorting into the exosomes in the dorsal striatum. Furthermore, diminished SYNCRIP in the dorsal striatum was necessary for MA abstinence-induced behavioral bias towards egocentric spatial learning. Taken together, our data revealed circulating miR-137 as a potential blood-based marker that could reflect MA abstinence-dependent changes in striatal miR-137/SYNCRIP axis, and striatal SYNCRIP as a potential therapeutic target for striatum-associated cognitive dysfunction by MA withdrawal syndrome.

BACKGROUND While primary intestinal lymphangiectasia(PIL)is considered a rare condition,there have been several reported cases in adults.Nevertheless,the absence of clear guidance from diagnosis to treatment and prognosis poses challenges for both physicians and patients.AIM To enhance understanding by investigating clinical presentation,diagnosis,treatment,complications,and prognoses in adult PIL cases.METHODS We enrolled adult patients diagnosed with PIL between March 2016 and September 2021.The primary outcome involved examining the diagnosis and treatment process of these patients.The secondary outcomes included identifying complications(infections,thromboembolism)and assessing prognoses(frequency of hospitalization and mortality)during the follow-up period.RESULTS Among the 12 included patients,peripheral edema(100%)and diarrhea(75%)were the main presenting complaints.Laboratory tests showed that all the pati-ents exhibited symptoms of hypoalbuminemia and hypogammaglobulinemia.Radiologically,the predominant findings were edema of the small intestine(67%)and ascites(58%).The typical endoscopic finding with a snowflake appearance was observed in 75%of patients.Among the 12 patients,two responded positive-ly to octreotide and sirolimus,and eight who could undergo maintenance therapy discontinued subsequently.Complications due to PIL led to infection in half of the patients,thromboembolism in three patients,and one death.CONCLUSION PIL can be diagnosed in adults across various age groups,with different severity and treatment responses among patients,leading to diverse complications and prognoses.Consequently,tailored treatments will be necessary.We anticipate that our findings will contribute to the management of PIL,an etiology of protein-losing enteropathy.

A very high temperature reactor （VHTR） is one of the next-generation nuclear reactors chosen by the Generation IV International Forum. A Ni-base superalloy, Alloy 617, is considered as a primary candidate material for an intermediate heat exchanger （IHX） and hot gas duct （HGD） of the VHTR because of the superior creep resistance at a high temperature above 850 ~C. In this study, the microstructures of the specimens creep- ruptured at high temperatures in a helium environment were investigated. The decrease in rupture time was more pronounced with increasing creep temperature. In the specimens crept at 950 ~C, the external Cr-oxide layer of the specimens increased in thickness with increasing creep rupture time, and delaminated after a long-time creep. The high creep stress induced a deep penetration of carbide depletion along the grain boundaries at the early stage of the creep test. The creep temperature enhanced the growth rate of the decarburized zone depth clearly, but the temperature effect on the growth of the external oxide and internal oxide was not well understood as the surface reaction and creep stress affected the microstructures complexly.

笔者认为，欲学好汉语，尤其是韩国留学生要学好汉语，首先必然要学好“语音”，而“语音”学习中最关键的一项，是准确掌握“声调”。如若作为基础中之基础的“声调”出现问题，则单词掌握愈多，歧义愈大，无法与中国人进行正常交流，产生诸多“声调”引发的误解。当下，许多学者都发表了与对外汉语研究相关的论文，但是针对汉语学习者声调学习情况的调查和研究却很少，专门针对韩国学生学习情况进行研究的论文就更寥寥无几，只有中国的朱川，韩国的孟柱忆、申小喜等几名学者发表过针对韩国学生汉语声调学习情况的论文。鉴于此，确有笔者行文研究之必要。为了调查韩国学生的声调使用的实际情况，笔者设计了三个调查——认识度调查、听辨能力调查、表达能力调查。调查对象为在长春留学的50名学生，调查用问卷的方式进行。调查分析结果显示，在认识度调查上，大部分学生对声调的认识是“记忆与感觉”，认为一声和四声发音较容易，二声和三声发音则相对比较难;在听辨能力调查上，发现了学生们在一声和四声之间、二声和三声之间的混淆现象比较严重;在表达能力调查上，得知韩国学生最容易出错的声调是二声，发音程度由难到易的排序为二声、四声、一声、三声，导致错误的原因包括缺少声调知识、缺乏发音技巧、纠正能力不够高等。本文把错误发生的原因分为三大部分。第一是心里要素，包括教师的心理要素与学生的心理要素;第二是教学材料的影响，其内容主要分为缺少系统的专业教材，没有相关考试;第三是教学方法的影响，表现在声调教育顺序不规范，缺乏多样性的教学方法。本文在第四章提出了对声调教学的建议。对心理要素的建议包括“强调声调的重要性”、“纠正声调的方式”、“激发学生的学习动机”，对教学材料的建议包括“教材研发的必要性”、“选择既专业又系统的教学材料”、“设立相关考试”，对教学方法的建议包括“声调教育顺序”、“以多样的教学方法来进行教育”、“能实际应用的教学方法”等。最后，附录了汉语教师在课堂上能应用的双音节教学材料与最小对比教学材料。

Many approaches to neurodegenerative diseases that focus on amyloid-βclearance and gene therapy have not been successful.Some therapeutic applications focus on enhancing neuronal cell survival during the pathogenesis of neurodegenerative diseases,including mitochondrial dysfunction.Plasma membrane(PM)redox enzymes are crucial in maintaining cellular physiology and redox homeostasis in response to mitochondrial dysfunction.Neurohormetic phytochemicals are known to induce the expression of detoxifying enzymes under stress conditions.In this study,mechanisms of neuroprotective effects of 4-hydroxycinnamic acid(HCA)were examined by analyzing cell survival,levels of abnormal proteins,and mitochondrial functions in two different neuronal cells.HCA protected two neuronal cells exhibited high expression of PM redox enzymes and the consequent increase in the NAD^(+)/NADH ratio.Cells cultured with HCA showed delayed apoptosis and decreased oxidative/nitrative damage accompanied by decreased ROS production in the mitochondria.HCA increased the mitochondrial complexes I and II activities and ATP production.Also,HCA increased mitochondrial fusion and decreased mitochondrial fission.Overall,HCA maintains redox homeostasis and energy metabolism under oxidative/metabolic stress conditions.These findings suggest that HCA could be a promising therapeutic approach for neurodegenerative diseases.
