Fabrication, characterization, in vitro drug release and glucose uptake activity of 14-deoxy,11, 12-didehydroandrographolide loaded polycaprolactone nanoparticles

Biodegradable polymer based novel drug delivery systems brought a considerable attention in enhancing the therapeutic efficacy and bioavailability of various drugs. 14-deoxy 11, 12-didehydro andrographolide(poorly water soluble compound) loaded polycaprolactone(nanoDDA) was synthesized using the solvent evaporation technique. Nano-DDA was characterized by scanning electron microscopy(SEM) and dynamic light scattering(DLS) studies. Fourier Transform InfraRed Spectroscopy(FTIR) was used to investigate the structural interaction between the drug and the polymer. Functional characterization of the formulation was determined using drug content, cellular uptake and in vitro drug release. 2-deoxy-D-[1-~3H] glucose uptake assay was carried out to assess the antidiabetic potential of nano-DDA in L6 *** nano-DDA displayed spherical shape with a smooth surface(252.898 nm diameter), zeta potential, encapsulation and loading efficiencies of -38.9 mV, 91.98 ± 0.13% and 15.09 ± 0.18% respectively. No structural alteration between the drug and the polymer was evidenced(FTIR analysis). Confocal microscopy studies with rhodamine 123 loaded polycaprolactone nanoparticles(Rh123-PCL NPs) revealed the internalization of Rh123-PCL NPs in a time dependent manner in L6 myoblasts. A dose dependent increase in glucose uptake was observed for nano-DDA with a maximal uptake of 108.54 ± 1.42% at 100 nM on L6 myotubes, thereby proving its anti-diabetic efficacy. A biphasic pattern of in vitro drug release demonstrated an initial burst release at 24 h followed by a sustained release for up to 11 days. To conclude,our results revealed that nano-DDA formulation can be a potent candidate for antidiabetic drug delivery.

基于双流体方程和颗粒动力学理论的计算模型被广泛应用于流化床的气固两相流数值计算,高精度网格是其准确计算流动的必要条件。一些经典的微尺度阻力模型,其网格尺度决定其模拟结果的精度。亚格子过滤双流体模型是一种有效的适用于粗糙网格的计算模型,其包含的气固相间作用力和颗粒相应力本构方程是在高精度网格条件下,以微尺度双流体方程和颗粒动力学理论计算得到的气固流场为基础,对计算结果进行小尺度过滤后得出。使用亚格子过滤双流体模型替换基于颗粒动力学理论的双流体模型,针对同一物理问题,在不同网格尺度下进行了数值计算,结果表明此计算模型相比经典阻力模型具有较好的网格无关特性,并且和实验结果较为一致。同时也对颗粒动力学理论与之相结合进行了尝试,即仅使用亚格子过滤阻力模型,颗粒相应力仍然使用颗粒动力学模型,其计算结果的网格无关性及与实验值的吻合程度优于经典模型。

＜正＞ In flowering plants, penetration of the pollen tube through stigma, style and transmitting tract is essential for delivery of sperm nuclei to the egg cells embedded deeply within female tissues. Despite its importance to plant reproduction, little is known about the underlying molecular mechanisms that regulate the navigation of pollen tube through the stigma, style and transmitting tract. Here we report the identification and characterization of an Arabidopsis gene, VANGUARD1（VGD1） that encodes a pectin methylesterase（PME）-homologous protein of 595 amino acids which is required for enhancing the growth of pollen tubes in style and transmitting tract tissues. VGD1 was expressed specifically in pollen grain and pollen tube. The VGD1 protein was distributed throughout pollen grain and pollen tube, including the plasma membrane and cell wall. Functional interruption of VGD1 reduced PME activity in pollen to 18% of wild-type and greatly retarded growth of the pollen tube in the style and transmitting tract, resulting in a significant reduction of male fertility. In addition, vgd1 pollen tubes were unstable and burst more frequently when germinated and grown on in vitro culture medium, compared to wild-type pollen tubes. Our study suggests that the VGD1 product is required for growth of the pollen tube, possibly via modifying the cell wall and enhancing the interaction of the pollen tube with the female style and transmitting tract.

Hepatoprotective efficiency of methanol extract of red algae against chromium-induced oxidative damage in Wistar rats

Objective:To investigate the hepatoprotective activity of red algae Portieria hornemannii(Lyngbye)Silva(***)and Spyridia fusiformis Boergesen(***)by using the chromium treated rat liver as the animal ***:The extract of red algae at a dosage of 0.200 g/kg of whole body weight was orally administrated to Cr(VI)intoxicated rats for 28 consecutive *** effect of drug in rats was evaluated by comparing the degree of the production of enzymes responsible for antioxidant activity such lipid peroxidase,superoxide dismutase,catalase and reduced glutathione with Cr(VI)analogs in the absence of any secondary *** overall damage of liver was detected by measuring serum enzymes such as aspartate amino transferase and alanine aminotransferase activities which released into the blood from the damaged ***:It was observed that these enzyme levels were noticed in the animals treated with methanol extracts of red algae(200 mg/kg)through preventing the leakage of the above enzymes into the *** hepatoprotection obtained using LIV 52(standard reference drug)appeared relatively *** antihepatotoxic potential of red algae *** and *** might be due to their antioxidative and membrane stabilizing ***:Our results indicated that the extract of *** and *** obtained from methanol could be a promising hepatoprotective agent against chromium(VI)-induced liver damage.

Low profile antenna in communication is a new methodology. Fractal geometry is a methodology through which size reduction is achieved. A Self-similar fractal antenna using multicantor technique is proposed and experimentally studied. Space-filling cantors and self-similarity properties of fractal geometry have been adopted in the proposed antenna to miniaturize the size of antenna. The antenna is designed in such a way to operate at MICS band (Medical Implant communication Service) for wireless telemedicine application. The prototype antenna exhibits wideband characteristics and provides a good agreement of return loss (S11). Experimental return loss has been compared with that which is obtained using method of moments. The objective of using self-similar concept in antenna makes it flexible in terms of controlling the resonance and bandwidth. In this paper, the Self-similar property of fractal geometry is examined by maintaining return loss (S11) more than –30 dB approximately in all the iterations (n) and the prototype antenna has return loss greater than –10 dB and exhibits wideband characteristics.
