FSD1 inhibits glioblastoma diffuse infiltration through restriction of HDAC6-mediated microtubule deacetylation

The infiltration of glioblastoma multiforme(GBM) is predominantly characterized by diffuse spread, contributing significantly to therapy resistance and recurrence of GBM. In this study, we reveal that microtubule deacetylation, mediated through the downregulation of fibronectin type III and SPRY domain-containing 1(FSD1), plays a pivotal role in promoting GBM diffuse infiltration. FSD1 directly interacts with histone deacetylase 6(HDAC6) at its second catalytic domain, thereby impeding its deacetylase activity on α-tubulin and preventing microtubule deacetylation and depolymerization. This inhibitory interaction is disrupted upon phosphorylation of FSD1 at its Ser317 and Ser324 residues by activated CDK5, leading to FSD1 dissociation from microtubules and facilitating HDAC6-mediated α-tubulin deacetylation. Furthermore, increased expression of FSD1 or interference with FSD1 phosphorylation reduces microtubule deacetylation, suppresses invasion of GBM stem cells, and ultimately mitigates tumor infiltration in orthotopic GBM xenografts. Importantly, GBM tissues exhibit diminished levels of FSD1 expression, correlating with microtubule deacetylation and unfavorable clinical outcomes in GBM patients. These findings elucidate the mechanistic involvement of microtubule deacetylation in driving GBM cell invasion and offer potential avenues for managing GBM infiltration.

Operation Risk Analysis of Electric Vehicle With Ultra-fast Charging Integrated to Distribution Network Considering the Bearing Capacity of Power System

At present, the access of distributed generation and electric vehicles with ultra-fast charging to the distribution network will increase the operational risk of the system. Aiming at the problem that the existing risk assessment indicators fail to consider the system capacity effectively, this paper proposed a comprehensive index of risk assessment of distribution networks considering the power system's capacity. The distributed generation output model and the electric vehicle load model with ultra-fast charging were first constructed. Then, the index based on the distribution entropy theory was used to measure the risk caused by the over-limit of state variables such as voltage and power and the uncertainty of their distribution. The index based on the Theil entropy theory was introduced to measure the risk caused by the equilibrium degree of the incremental distribution of state variables after the load impacted the system. The weight analysis method combining the order relationship method and the objective weighting method was used to evaluate the system risk comprehensively. Finally, the rationality of the comprehensive index was calculated and analyzed by testing the IEEE 33-node system. The results show that the comprehensive index proposed can effectively evaluate the potential risk when the system does not exceed the limit and consider that the ultra-fast charging of electric vehicles will change the time series distribution characteristics of comprehensive risk. The research in this paper can provide theoretical guidance for the safe operation of electric vehicles. with ultra-fast charging connected to the distribution network. © 2025 Power System Technology Press. There is a need to develop interventions to slow or reverse the degeneration of dopamine neurons in Parkinson’s disease after *** that preclinical and clinical studies suggest benefits of dietary n-3 polyunsaturated fatty acids,such as docosahexaenoic acid,and exercise in Parkinson’s disease,we investigated whether both could synergistically interact to induce recovery of the dopaminergic ***,mice received a unilateral stereotactic injection of 6-hydroxydopamine into the striatum to establish an animal model of nigrostriatal *** weeks after lesion,animals were fed a docosahexaenoic acid-enriched or a control diet for the next 8 *** this period,the animals had access to a running wheel,which they could use or *** acid treatment,voluntary exercise,or the combination of both had no effect on(i)distance traveled in the open field test,(ii)the percentage of contraversive rotations in the apomorphine-induction test or(iii)the number of tyrosine-hydroxylase-positive cells in the substantia nigra pars ***,the docosahexaenoic acid diet increased the number of tyrosine-hydroxylase-positive terminals and induced a rise in dopamine concentrations in the lesioned *** to docosahexaenoic acid treatment or exercise alone,the combination of docosahexaenoic acid and exercise(i)improved forelimb balance in the stepping test,(ii)decreased the striatal DOPAC/dopamine ratio and(iii)led to increased dopamine transporter levels in the lesioned *** present results suggest that the combination of exercise and docosahexaenoic acid may act synergistically in the striatum of mice with a unilateral lesion of the dopaminergic system and provide support for clinical trials combining nutrition and physical exercise in the treatment of Parkinson’s disease.

分布式电源（DG）以及大量恒功率直流负荷接入交直流配电系统使得母线暂态电压表现出正负极耦合且多波动的特性，现有暂态电压稳定性评估方法准确率低且适用性较差。推导分析了双极交直流配电系统直流侧线路、负荷参数不平衡时影响系统母线暂态电压稳定的因素。基于二元表判据思想，提出一种改进的自适应母线暂态电压稳定性评估指标。针对参数不平衡时含DG的双极交直流配电系统，引入稳定性保持因子，提出母线与系统的暂态电压稳定裕度计算方法。基于所提方法利用MATLAB/Simulink仿真平台在改进的IEEE 33节点与IEEE 69节点双极交直流配电系统中量化影响系统暂态电压稳定性的因素，并对比验证了所提指标的有效性和优越性。
