Background： Early pregnancy failure has a profound impact on both human reproductive health and animal production. 2/3 pregnancy failures occur during the peri-implantation period; however, the underlying mechanism（s） remains unclear. Well-organized modification of the endometrium to a receptive state is critical to establish pregnancy Aberrant endometrial modification during implantation is thought to be largely responsible for early pregnancy loss. Result： In this study, using well-managed recipient ewes that received embryo transfer as model, we compared the endometrial proteome between pregnant and non-pregnant ewes during implantation period. After embryo transfer, recipients were assigned as pregnant or non-pregnant ewes according to the presence or absence of an elongated conceptus at Day 17 of pregnancy. By comparing the endometrial proteomic profiles between pregnant and non-pregnant ewes, we identified 94 and 257 differentially expressed proteins （DEPs） in the endometrial caruncular and intercaruncular areas, respectively. Functional analysis showed that the DEPs were mainly associated with immune response, nutrient transport and utilization, as well as proteasome-mediated proteolysis. Conclusion： These analysis imply that dysfunction of these biological processes or pathways of DEP in the endometrium is highly associated with early pregnancy loss. In addition, many proteins that are essential for the establishment of pregnancy showed dysregulation in the endometrium of non-pregnant ewes. These proteins, as potential candidates, may contribute to early pregnancy loss.

Apical periodontitis is one of the most common dental pulp diseases and is the most common oral disease in clinical practice. OPN is produced by cells involved in bone morphogenesis such as osteoblasts and osteoclasts. OPN regulates various biological activities. OPN is a secreted glycoprotein with multiple glycosylation sites and phosphorylation sites. These modifications are closely related to the molecular function of OPN itself. Methods: Identification of OPN N-glycosylation sites by MS. The asparagine at position 79 of the OPN protein was mutated to glutamine, and the above plasmids were transfected into osteoblasts and osteoclasts. The effect of OPK N-glycosylation on proliferation of osteoblasts and osteoclasts was detected by CCK8. Western Blot was used to detect the expression of OPN N-glycosylation on osteoclasts and osteoblasts. Detection of N-glycosylation of OPN activated NF-κB signaling pathway to regulate osteoblasts and osteoclasts. Results:The OPN N-glycosylation site was identified as 79 by mass spectrometry. N-glycosylation of OPN can promote the proliferation of osteoclasts and inhibit the proliferation of *** N-glycosylation modulates the expression of osteoclast and osteoblast-associated factors through NF-κb signaling pathway. N-glycosylation of OPN promotes nuclear translocation of NF-κB in osteoclasts and osteoblasts. Conclusion:The N-glycosylation site of OPN is 79 *** N-glycosylation of OPN plays an important role in the biological function of OPN protein.

采用改进Hummers法制备了氧化石墨烯(GO),并将GO与经硅烷偶联剂γ-氨丙基三乙氧基硅烷(KH550)改性的纳米SiO_2进行复合,制备出纳米SiO_2-GO。通过FTIR、XRD、SEM、TEM等分析手段对SiO_2-GO进行表征。采用机械搅拌与超声分散的方法将SiO_2-GO添加到环氧树脂(EP)中。对添加不同质量分数纳米SiO_2、GO和纳米SiO_2-GO的EP基复合材料涂层的物理性能和电化学性能进行测试。结果表明,与纯EP涂层相比,SiO_2/EP、GO/EP和纳米SiO_2-GO/EP复合材料涂层的硬度、附着力和耐腐蚀性能得到显著增强,其中加入2wt%纳米SiO_2-GO/EP复合材料涂层硬度达到5 H,附着力等级达到1级,浸泡24h后涂层保护效率为99.33%。15天浸泡试验结果表明,添加1.5wt%纳米SiO_2-GO/EP复合材料涂层的硬度达到5H,附着力达到1级,涂层保护效率仍能达到97.12%。

作为一种新型碳纳米材料,石墨烯在诸多领域表现出了良好的适用性,得到了学术界及工业界的关注。在抗菌方面,石墨烯及其衍生物具有良好的抗菌性和生物相容性,同时也是抗菌活性物质的理想载体,利用石墨烯材料制备出的复合材料具有抗菌性能强、作用时间久、拉伸稳定性好、化学性质稳定等优点,可广泛应用于水处理、分离膜、医疗和制药领域。以近期文献为基础详细介绍了石墨烯抗菌机理及石墨烯复合材料在抗菌方面的应用,并阐述了石墨烯材料存在的生物安全性问题。
