Effect of the ginsenoside Rb1 on the spontaneous contraction of intestinal smooth muscle in mice

AIM: To investigate the effect and the possible mechanism of ginsenoside Rb1 on small intestinal smooth muscle motility in mice. METHODS: Intestinal smooth muscle strips were isolated from male ICR mice (5 wk old), and the effect of ginsenoside Rb1 on spontaneous contraction was recorded with an electrophysiolograph. The effect of ginsenoside Rb1 on ion channel currents, including the voltage-gated K + channel current (IK V ), calcium-activated potassium channel currents (IK Ca ), spontaneous transient outward currents and ATP-sensitive potassium channel current (IK ATP ), was recorded on freshly isolated single cells using the whole-cell patch clamp technique. RESULTS: Ginsenoside Rb1 dose-dependently inhibited the spontaneous contraction of intestinal smooth muscle by 21.15% ± 3.31%, 42.03% ± 8.23% and 67.23% ± 5.63% at concentrations of 25 μmol/L, 50 μmol/L and 100 μmol/L, respectively (n=5,P<0.05). The inhibitory effect of ginsenoside Rb1 on spontaneous contraction was significantly but incompletely blocked by 10 mmol/L tetraethylammonium or 0.5 mmol/L 4-aminopyridine, respectively (n=5, P<0.05). However, the inhibitory effect of ginsenoside Rb1 on spontaneous contraction was not affected by 10 μmol/L glibenclamide or 0.4 μmol/L tetrodotoxin. At the cell level, ginsenoside Rb1 increased outward potassium currents, and IK V was enhanced from 1137.71 ± 171.62 pA to 1449.73 ± 162.39 pA by 50 μmol/L Rb1 at +60 mV (n=6, P<0.05). Ginsenoside Rb1 increased IK Ca and enhanced the amplitudes of spontaneous transient outward currents from 582.77 ± 179.09 mV to 788.12 ± 278.34 mV (n=5, P<0.05). However, ginsenoside Rb1 (50 μmol/L) had no significant effect on IK ATP (n=3, P<0.05). CONCLUSION: These results suggest that ginsenoside Rb1 has an inhibitory effect on the spontaneous contraction of mouse intestinal smooth muscle mediated by the activation of IK V and IK Ca , but the K ATP channel was not involved in this effect.

Experimental investigation of the effect of flow turbulence and sediment transport patterns on the adsorption of cadmium ions onto sediment particles

The mechanism of flow turbulence, sediment supply conditions, and sediment transport patterns that affect the adsorption of cadmium ions onto sediment particles in natural waters are experimentally simulated and studied both in batch reactors and in a turbulence simulation tank. By changing the agitation conditions, the sediment transport in batch reactors can be categorized into bottom sediment-dominated sediment and suspended sediment-dominated sediment. It is found that the adsorption rate of bottom sediment is much less than that of suspended sediment, but the sediment transport pattern does not affect the final （equilibrium） concentration of dissolved cadmium. This result indicates that the parameters of an adsorption isotherm are the same regardless of the sediment transport pattern. In the turbulence simulation tank, the turbulence is generated by harmonic grid-stirred motions, and the turbulence intensity is quantified in terms of eddy diffusivity, which is equal to 9.84F （F is the harmonic vibration frequency） and is comparable to natural surface water conditions. When the turbulence intensity of flow is low and sediment particles stay as bottom sediment, the adsorption rate is significantly low, and the adsorption quantity compared with that of suspended sediment is negligible in the 6 h duration of the experiment. This result greatly favors the simplification of the numerical modeling of heavy metal pollutant transformation in natural rivers. When the turbulence intensity is high but bottom sediment persists, the rate and extent of descent of the dissolved cadmium concentration in the tank noticeably increase, and the time that is required to reach adsorption equilibrium also increases considerably due to the continuous exchange that occurs between the suspended sediment and the bottom sediment. The perinucleolar compartment （PNC） is a unique nuclear substructure, forming predominantly in cancer cells both in vitro and in vivo. PNC prevalence （percentage of cells containing at least one PNC） has been found to positively correlate with disease progression in several cancers （breast, ovarian, and colon）. While there is a clear association between PNCs and cancer, the molecular function of the PNC remains unclear. Here we summarize the current understanding of the association of PNCs with cancer and its possible functions in cancer cells.

Rapid determination of pharmaceuticals from multiple therapeutic classes in wastewater by solid-phase extraction and ultra-performance liquid chromatography tandem mass spectrometry

A new analytical method utilizing ultra-performance liquid chromatography (UPLC) tandem mass spectrometry (MS/MS) has been developed to determine 16 pharmaceuticals from 8 therapeutic classes in wastewater: bezafibrate, clofibric acid, carbamazepine, caffeine, chloramphenicol, diclofenac, gemfibrozil, indomethacin, ketoprofen, mefenamic acid, metoprolol, nalidixic acid, N,N-diethyl-metatoluamide, propranolol, sulpiride and trimethoprim. Key parameters of MS/MS, UPLC and solid phase extraction (SPE) were optimized. In general, recovery of target pharmaceuticals was over 70% for the wastewater effluent samples and 50% for the influent samples. The effects of matrix suppression, loss during the pretreatment as well as instrument variability were successfully corrected by two internal standards, and acceptable relative recovery was obtained. Target compounds were quantitatively analyzed using multiple reaction monitoring (MRM) mode, and the detection limits ranged from 0.3 to 20 ng/L. A detailed study, matrix effect in effluent wastewater was also present. The method was applied to detecting pharmaceuticals in the wastewater from three wastewater treatment plants (WWTPs) in Beijing, China and the results demonstrated that most target compounds were detectable in both the influent and effluent, with the mean concentrations ranging from 20.5 to 5775.6 ng/L and 4.6 to 418.6 ng/L, respectively.

NUMERICAL MODELING OF SUSPENDED SEDIMENT TRANSPORT IN CHANNEL BENDS

An algorithm to compute three-dimensional sediment transport effect was proposed in this paper to enhance the capability of depth-averaged numerical models. This algorithm took into account of non-uniform distributions of flow velocities and suspended sediment concentrations along water depth, it significantly enhanced the applicability of 2D models in simulating open channel flows, especially in channel bends. Preliminary numerical experiments in a U-shaped and a sine-generated experimental channel indicate that the proposed method performs quite well in predicting the change of bed-deformation in channel bends due to suspended sediment transport. This method provides an effective alternative for the simulations of channel morphodynamic changes.

Trade and Rural-Urban Income Inequality： Evidence from China

Using the provincial panel data from 1978 to 2007, this paper makes an empirical analysis of the relationship between international trade and rural-urban income inequality in China. The results show that international trade has a fundamental impact on rural-urban income inequality. For the whole country, the development of international trade from 1978 to 2007 enlarged the rural-urban income gap. During the three different periods （defined in section 3.3）, the impact was also noticeable in terms of extent, direction and significance respectively at the national and regional levels. During the three periods, the relationship has an inverted ＂U＂ shape for China as a whole, central China and western China, but for eastern China the relationship has a ＂U＂ shape. At the same time, exports and imports have different influences on the rural-urban income gap in China, the influences were also different across regions; and the imports has had a more significant influence on these regions than exports has had.

Impact of physical representations in CALMET on the simulated wind field over land during Super Typhoon Meranti(2016)

A WRF(Weather Research and Forecasting Model)/CALMET(California Meteorological Model)coupled system is used to investigate the impact of physical representations in CALMET on simulations of the near-surface wind field of Super Typhoon Meranti(2016).The coupled system is configured with a horizontal grid spacing of 3 km in WRF and 500 m in CALMET,*** model performance of the coupled WRF/CALMET system is evaluated by comparing the results of simulations with observational data from 981 automatic surface stations in Fujian *** root mean square error(RMSE)of the wind speed at 10 m in all CALMET simulations is significantly less than the WRF simulation by 20%^30%,suggesting that the coupled WRF/CALMET system is capable of representing more realistic simulated wind speed than the mesoscale model *** impacts of three physical representations including blocking effects,kinematic effects of terrain and slope flows in CALMET are examined in a specified local region called Shishe *** results show that before the typhoon landfall in Xiamen,a net downslope flow that is tangent to the terrain is generated in the west of Shishe Mountain due to blocking effects with magnitude exceeding 10 m/***,the blocking effects seem to take no effect in the strong wind area after typhoon *** being affected by the typhoon strong wind or not,the slope flows move downslope at night and upslope in the daytime due to the diurnal variability of the local heat flux with magnitude smaller than 3 m/*** kinematic effects of terrain,which are speculated to play a significant role in the typhoon strong wind area,can only be applied to atmospheric flows in stable conditions when the wind field is quasinondivergent.

Most natural rivers are curved channels, where the turbulent flows have a complex helical pattern, as has been extensively studied both numerically and experimentally. The helical flow structure in curved channels has an important bearing on sediment transport, riverbed evolution, and pollutant transport study. In this article, different turbulence closure schemes i.e., the mixing-length model and the k-ε model with different pressure solution techniques i. e., hydrostatic assumptions and dynamic pressure treatments are applied to study the helical secondary flows in an experiment curved channel. The agreements of vertically-averaged velocities between the simulated results obtained by using different turbulence models with different pressure solution techniques and the measured data are satisfactory. Their discrepancies with respect to surface elevations, superelevations and secondary flow patterns are discussed.
