A new device used to detect the low contrast target acquisition of photoelectric theodolite is designed and its reliability is experimentally *** adjustable contrast optical target device,which can simulate the sky background luminance and a low contrast target,is *** utilizes a big integrating sphere and a small one to simulate the luminance of the background and target ***,by controlling the luminous flux of the two integrating spheres,the targets and background radiance can be continuously adjustable under the condition of constant color ***,the contrast can be controlled continuously in the range of 0%-90% and its stability is better than 1%.The biggest background luminance exceeds 60 W m-2str-1 in the spectral range of 400-800 nm.

Fine particles associated with haze pollution threaten the health of over 400 million people in China. Owing to excellent non-destructive fingerprint recognition characteristics, Raman and surface-enhanced Raman scattering(SERS) are often used to analyze the composition of fine particles to determine their physical and chemical properties as well as reaction mechanisms. However, there is no comprehensive Raman spectral library of fine particles. Furthermore, various studies that used SERS for fine-particle composition analysis showed that the uniqueness of the SERS substrates and different excitation wavelengths can produce a different spectrum for the same fine-particle component. To overcome this limitation, we conducted SERS experiments with a portable Raman spectrometer using two common SERS substrates(silver(Ag) foil and gold nanoparticles(Au NPs)) and a 785 nm laser. Herein, we introduced three main particle component types(sulfate-nitrate-ammonium(SNA), organic material, and soot) with a total of 39 chemical substances. We scanned the solid Raman, liquid Raman, and SERS spectra of these substances and constructed a fine-particle reference library containing 105 spectra. Spectral results indicated that for soot and SNA, the differences in characteristic peaks mainly originated from the solid-liquid phase transition;Ag foil had little effect on this difference, while the Au NPs caused a significant red shift in the peak positions of polycyclic aromatic hydrocarbons. Moreover, with various characteristic peak positions in the three types of spectra, we could quickly and correctly distinguish substances. We hope that this spectral library will aid in the future identification of fine particles.

The present work reports a sensitive and selective fluorescent sensor for the detection of mercury ion, Hg（II）, by hybridizing carbon nanodots （C-dots） and gold nanoclusters （Au NCs） through intrinsic interactions of the two components. The C-dots serve as the reference signal and the Au NCs as the reporter. This method employs the specific high affinity metallophilic Hg2^-Au＋ interactions which can greatly quench the red fluorescence of Au NCs, while the blue fluorescence of C-dots is stable against Hg（II）, leading to distinct ratiometric fluorescence changes when exposed to Hg（II）. A limit of detection of 28 nM for Hg（II） in aqueous solution was estimated. Thus we applied the sensor for the detection of Hg（II） in real water samples including tap water, lake water and mineral water samples with good results. We further demonstrated that a visual chemical sensor could be manufactured by immobilizing the nanohybrid probe on a cellulose acetate circular filter paper. The paper-based sensor immediately showed a distinct fluorescence color evolution from pink to blue after exposure to a drop of the Hg（II） solution

Cowpea(Vigna imguicuiata) is an important legume crop with diverse uses. The species is presently a minor crop, and evaluation of its genetic diversity has been very limited. In this study, a total of 200 genic and 100 genomic simple sequence repeat(SSR) markers were developed from cowpea unigene and genome sequences, respectively. Among them, 27 genic and 27 genomic SSR markers were polymorphic and were used for assessment of genetic diversity and population structure in 105 selected cowpea accessions. A total of 155 alleles and 2.9 alleles per marker were identified, and the average polymorphic information content(PIC) value was 0.3615. The average PIC of genomic SSRs(0.3996) was higher than that of genic SSRs(0.3235), and most of the polymorphic genomic SSRs were composed of di-and trinucleotide repeats(51.9% and 37.0% of all loci, respectively). The low level of detected genetic diversity may be attributed to a severe genetic bottleneck that occurred during the cowpea domestication process. The accessions were classified by structure and cluster analysis into four subgroups that correlated well with their geographic origins or collection sites. The classification results were also consistent with the results from principal coordinate analysis and can be used as a guide during future germplasm collection and selection of accessions as breeding materials for cultivar improvement. The newly developed genic and genomic SSR markers described in this study will be valuable genomic resources for the assessment of genetic diversity, population structure, evaluation of germplasm accessions, construction of genetic maps, identification of genes of interest,and application of marker-assisted selection in cowpea breeding programs.

To correctly assess and properly manage the public health risks associated with exposure to contaminated water,it is necessary to identify the source of fecal pollution in a *** this study,we evaluated the efficacy of our two previously developed real time-quantitative PCR(qPCR)assays for the detection of swine-associated Bacteroidales genetic markers(gene 1-38,gene 3-53)in the Yangtze Delta watershed of southeastern *** results indicated that the gene 1-38 and 3-53 markers exhibited high accuracy(92.5%,91.7%conditional probability,respectively)in detecting Bacteroidales *** water *** to binary logistic regression(BLR),these two swine-associated markers were well correlated(P<0.05)with fecal indicators(Escherichia coli and Enterococci spp.)and zoonotic pathogens(*** O157:H7,Salmonella *** Campylobacter spp.)in water *** contrast,concentrations of conventional fecal indicator bacteria(FIB)were not correlated with zoonotic pathogens,suggesting that they are noneffective at detecting fecal pollution ***,the results obtained in this study demonstrated that a swinetargeted qPCR assay based on two Bacteroidales genes markers(gene 1-38,gene 3-53)could be a useful tool in determining the swine-associated impacts of fecal contamination in a watershed.

Genetic diversity assessment of a set of introduced mung bean accessions(Vigna radiata L.)

Genetic resources from other countries or regions play an important role in broadening the genetic background of local breeding varieties. Here we describe observations on the adaptability of mung bean germplasm obtained from the United States Department of Agriculture and their genetic diversity assessment using SSR markers. Several accessions were shown to be mixtures,based on their phenotypes for some characters. Most accessions were able to complete their lifecycles when grown in Beijing, China, making them ideal for crossbreeding without day length control. High diversity was revealed by the SSR markers, with an average of 4.2 alleles per locus and a PIC value of 0.650 per locus. STRUCTURE analysis divided the accessions into six *** was no obvious trend of accessions forming groups according to their geographical origin,owing mainly to germplasm exchange and an uneven distribution of accessions. The present results indicate that this germplasm would enrich the local gene pool, and provide information for the further use of germplasm in breeding programs.

Bruchid beetle(Callosobruchus chinensis) poses a serious threat to the production and storage of mung bean(Vigna radiata). Mapping bruchid resistance(Br) will provide an important basis for cloning the responsible gene(s) and elucidating its functional mechanism, and will also facilitate marker-assisted selection in mung bean breeding. Here, we report the construction of the genetic linkage groups of mung bean and mapping of the Br1 locus using an RIL population derived from a cross between Berken, a bruchid-susceptible line, and ACC41, a bruchid-resistant line. A total of 560 markers were mapped onto 11 linkage groups,with 38.0% of the markers showing distorted segregation. The lengths of the linkage groups ranged from 45.2 to 117.0 c M with a total coverage of 732.9 c M and an average interval of1.3 c M between loci. Br1 was located on LG9 between BM202(0.7 c M) and Vr2-627(1.7 c M).Based on 270 shared SSR markers, most of the linkage groups were assigned to specific chromosomes. These results should further accelerate the genetic study of this crop.
