Genomic analysis of Nypa fruticans elucidates its intertidal adaptations and early palm evolution

Nypa fruticans(Wurmb),a mangrove palm species with origins dating back to the Late Cretaceous period,is a unique species for investigating long-term adaptation strategies to intertidal environments and the early evolution of ***,we present a chromosome-level genome sequence and assembly for *** integrated the genomes of *** and other palm family members for a comparative genomic analysis,which confirmed that the common ancestor of all palms experienced a whole-genome duplication event around 89 million years ago,shaping the distinctive characteristics observed in this *** also inferred a low mutation rate for the *** genome,which underwent strong purifying selection and evolved slowly,thus contributing to its stability over a long evolutionary ***,ancient duplicates were preferentially retained,with critical genes having experienced positive selection,enhancing waterlogging tolerance in ***,we discovered that the pseudogenization of Early Methionine-labelled 1(EM1)and EM6 in *** underly its crypto-vivipary characteristics,reflecting its intertidal *** study provides valuable genomic insights into the evolutionary history,genome stability,and adaptive evolution of the mangrove *** results also shed light on the long-term adaptation of this species and contribute to our understanding of the evolutionary dynamics in the palm family.

目的:分析叙事护理干预对血液透析患者心理健康与睡眠质量的影响。方法:选取2022年1月至2023年5月陆军第七十三集团军医院肾内科行血液透析治疗的患者66例作为主要对象,采用随机数字表法分为对照组和观察组,每组33例。对照组给予常规护理干预,观察组在对照组基础上给予叙事护理干预。采用匹兹堡睡眠质量指数(PSQI)比较2组患者干预前后睡眠质量的变化,采用汉密尔顿焦虑量表(HAMA)、汉密尔顿抑郁量表(HAMD)比较2组患者干预前后焦虑、抑郁状态的改善效果,采用生命质量测定量表(QLQ-C30)比较2组患者干预前后生命质量的变化。结果:干预后,观察组HAMA评分、HAMD评分、PSQI评分显著低于对照组,观察组QLQ-C30评分显著高于对照组,差异均有统计学意义(均P<0.05)。结论:将叙事护理运用在血液透析患者的护理工作中能够获得显著性优势,不仅可以改善患者的不良情绪,又可以提高其睡眠质量和生命质量,有借鉴意义和临床推广价值。

目的分析司美格鲁肽对2型糖尿病合并肾功能不全患者血糖水平的影响。方法回顾性选取陆军第七十三集团军医院于2023年1月—2024年3月收治的100例2型糖尿病合并肾功能不全患者的临床资料,根据治疗方式不同分两组,各50例。对照组三餐前皮下注射胰岛素,研究组在对照组基础上皮下注射司美格鲁肽。治疗前后,对比两组血糖水平(糖化血红蛋白、空腹血糖、餐后2 h血糖)、肾功能(血尿素氮、白蛋白、血肌酐、尿微量白蛋白)、炎症因子(白细胞介素6、超敏C反应蛋白、丙二醛)及不良反应发生情况(恶心、呕吐、腹泻、心率加快)。结果治疗后,研究组血糖水平低于对照组,差异有统计学意义(P均0.05)。结论司美格鲁肽对2型糖尿病合并肾功能不全患者能够改善血糖水平、提高肾功能、降低炎症因子水平,并且不会增加不良反应发生风险。

Two decades of suspect evidence for adaptive molecular evolution——negative selection confounding positive-selection signals

There has been a large literature in the last two decades affirming adaptive DNA sequence evolution between *** main lines of evidence are from(i) the McDonald-Kreitman(MK) test,which compares divergence and polymorphism data,and(ii) the phylogenetic analysis by maximum likelihood(PAML) test,which analyzes multispecies divergence ***,we apply these two tests concurrently to genomic data of Drosophila and *** our surprise,the> 100 genes identified by the two tests do not overlap beyond random *** the non-concordance could be due to low powers leading to high false negatives,we merge every 20-30 genes into a 'supergene'.At the supergene level,the power of detection is large but the calls still do not *** rule out methodological reasons for the *** particular,extensive simulations fail to find scenarios whereby positive selection can only be detected by either MK or PAML,but not *** molecular evolution is governed by positive and negative selection concurrently,a fundamental assumption for estimating one of these(say,positive selection) is that the other is ***,in a broad survey of primates,birds,Drosophila and Arabidopsis,we found that negative selection rarely stays constant for long in *** a consequence,the variation in negative selection is often misconstrued as a signal of positive *** conclusion,MK,PAML and any method that examines genomic sequence evolution has to explicitly address the variation in negative selection before estimating positive *** a companion study,we propose a possible path forward in two stages—first,by mapping out the changes in negative selection and then using this map to estimate positive *** now,the large literature on positive selection between species has to await reassessment.

Assembly processes underlying bacterial community differentiation among geographically close mangrove forests

Bacterial communities play pivotal roles in nutrient cycling in mangrove *** assembly of mangrove microbial communities has been found to be influenced by complex factors,such as geographic distance,physicochemical conditions,and plant identity,but the relative importance of these factors and how these factors shape the assembling process remain *** analyzed the bacterial communities sampled from three mangrove species(Aegiceras corniculatum,Bruguiera sexangula,and Kandelia obovata)at three locations along the estuarine Dongzhai Harbor in Hainan,*** revealed larger differences in rhizosphere bacterial communities among geographical locations than among plant species,indicated by differences in diversity,composition,and interaction *** found that dispersal limitation and homogeneous selection have substantial contributions to the assembly of mangrove rhizosphere bacterial communities in all three *** the phylogenetic‐bin‐based null model analysis(iCAMP)framework,we also found dispersal limitation and homogeneous selection showing dominance in some *** greater differences among geographic locations may be mainly attributed to the larger proportions of dispersal limitation even at such a short geographic *** also found that beta diversity was positively correlated with environmental distances,implying that the more similar environmental conditions(such as rich carbon and nitrogen contents)among plant species may have shaped similar bacterial *** concluded that the geographic distances,which are associated with dispersal limitation,played a key role in assembling mangrove rhizosphere bacterial communities,while physicochemical conditions and plant identity contributed less.

Expansion and adaptive evolution of the WRKY transcription factor family in Avicennia mangrove trees

Mangroves are adapted to intertidal zones,which present extreme environmental *** are among the most prominent transcription factors(TFs)in higher plants and act through various interconnected networks to regulate responses to multiple abiotic ***,based on omic data,we investigated the landscape and evolutionary patterns of WRKYs in the main mangrove genus *** found that both the number and the proportion of TFs and WRKYs in Avicennia species exceeded their inland relatives,indicating a signifcant expansion of WRKYs in *** identifed 109 WRKY genes in the representative species Avicennia *** genomic analysis showed that two recent wholegenome duplication(WGD)events played a critical role in the expansion of WRKYs,and 88%of Avicennia marina WRKYs(AmWRKYs)have been retained following these *** comparative transcriptomics on roots under experimental salt gradients,we inferred that there is high divergence in the expression of WGD-retained ***,we found that the expression of 16 AmWRKYs was stable between freshwater and moderately saline water but increased when the trees were exposed to high *** particular,14 duplicates were retained following the two recent WGD events,indicating potential neo-and *** also found that WRKYs could interact with other upregulated genes involved in signalling pathways and natural antioxidant biosynthesis to enhance salt tolerance,contributing to the adaptation to intertidal *** omic data of the WRKY family in *** broadens the understanding of how a TF family relates to the adaptive evolution of mangroves.

原发性痛经(PD)是妇科常见病,艾灸治疗PD有一定优势。时素华基于传承李志刚教授“通督启神”针法充督启阳、通督启神之理念并结合多年艾灸诊治PD的临床经验,总结出痛经具有“虚、寒、瘀、滞”的病机特点,重视情志因素对痛经的影响,在此基础上提出以“补虚散寒,逐瘀通滞为主,兼通督启神,灸畅情志”的灸治原则,临床以阴阳温经灸为主随证加减灸治PD。该灸法在改良岳美中“大灸”基础上结合重灸法,以择时逆灸,量大时久,重灸透达为主要灸治特点,具灸面大,火力足,温透力强且高效的灸疗优势。本文结合时素华对PD病因病机的认识,介绍阴阳温经灸,并结合典型病案以供参考,为临床艾灸治疗此类疾病提供新思路。

Molecular Population Genetics of Rice Domestication

Domestication is a selection process that genetically modifies species to meet human needs. A most intriguing feature of domestication is the extreme phenotypic diversification among breeds. What could be the ultimate source of such genetic variations？ Another notable outcome of artificial selection is the reduction In the fitness of domesticated species when they live In the wild without human assistance. The complete sequences of the two subspecies of rice cultlvars provide an opportunity to address these questions. Between the two subspecies, we found much higher rates of non-synonymous （N） than synonymous （S） substitutions and the N/S ratios are higher between cultivars than between wild species. Most interestingly, substitutions of highly dissimilar amino acids that are deleterious and uncommon between natural species are disproportionately common between the two subspecies of rice. We suggest strong selection in the absence of effective recombination may be the driving force, which we called the domestication-associated Hill-Robertson effect. These hitchhiking mutations may contribute to some fitness reduction in cultivars. Comparisons of the two genomes also reveal the existence of highly divergent regions in the genomes. Haplotypes in these regions often form highly polymorphic linkage blocks that are much older than speciation between wild species. Genes from such regions could contribute to the differences between indica and japonica and are likely to be involved in the diversifying selection under domestication. Their existence suggests that the amount of genetic variation within the single progenitor species Oryza rufipogon may be insufficient to account for the variation among rice cultivars, which may come from a more inclusive gene pool comprising most of the A-genome wild species.
