Use of the DSI/Ponemah jacketed external telemetry system for ECG assessments in dog telemetry

OBJECTIVE To demonstrate that the DSI /Ponemah Jet System(v5.00) can generate high quality ECG data with current methods,enabling cardiovascular function to be assessed in free moving dogs in their home *** Four male beagle dogs were used on this *** electrodes were placed on each animal and were connected to each JET *** JET device was then placed into a pocket of an externally worn animal jacket for each *** JET device transmits to a Bluetooth? *** Bluetooth? receiver can receive signals from up to 6 JET devices depending upon the device *** JET device can transmit data up to 10 meters with negligible interference and no cross-talk exists between *** animals underwent acclimatization to jackets prior to commencement of *** dog was placed into a pen and ECG measurements taken simultaneously using the DSI /Ponemah Jet *** were recorded for 1 hour pre-dose and continually for the following 23 hours under control conditions,then dosed with ascending doses of Moxifloxacin at intervals of 48 hours between each *** AND CONCLUSION Prolonged QT and QTcV(Van de Water's correction) interval was observed for animals dosed at 100 mg·kg-1 from 4-23 h *** QTcV effects(19% increase with 100 mg·kg-1 Moxifloxacin vs control).The DSI telemetry system performed well when testing the dog to the known pharmacological profile of *** sensitively and efficiently acquired ECG data from the beagle dogs and demonstrated the sensitivity of the system to detect test article induced QT *** allows a colony of dogs to be dedicated for toxicology and safety pharmacology *** DSI /ponemah JET system enables flexibility in study design and can be utilized on a range of study types to facilitate high quality ECGs and reduce the requirement for repeat dosing safety pharmacology *** data generated from this investigation demonstrate the DSI /ponemah JET system(v5.

Identification of the varietal origin of processed loose-leaf tea based on analysis of a single leaf by SNP nanofluidic array

Tea is an important cash crop, representing a $40 billion-a-year global market. Differentiation of the tea market has resulted in increasing demand for tea products that are sustainably and responsibly produced. Tea authentication is important because of growing concerns about fraud involving premium tea products. Analytical technologies are needed for protection and value enhancement of high-quality brands. For loose-leaf teas, the challenge is that the authentication needs to be established on the basis of a single leaf, so that the products can be traced back to the original varieties. A new generation of molecular markers offers an ideal solution for authentication of processed agricultural products. Using a nanofluidic array to identify variant SNP sequences, we tested genetic identities using DNA extracted from single leaves of 14 processed commercial tea products. Based on the profiles of 60 SNP markers, the genetic identity of each tea sample was unambiguously identified by multilocus matching and ordination analysis. Results for repeated samples of multiple tea leaves from the same products(using three independent DNA extractions) showed 100% concordance, showing that the nanofluidic system is a reliable platform for generating tea DNA fingerprints with high accuracy. The method worked well on green, oolong, and black teas, and can handle a large number of samples in a short period of time. It is robust and cost-effective, thus showing high potential for practical application in the value chain of the tea industry.

Off-flavour taint of aquaculture products is a global issue reducing consumer confidence in the farmed produce as they are taken up via the gills of fish,and deposited in the lipids of the *** the fish are not purged,resulting undesirable muddy earthy flavour taint can be tasted by *** undesirable flavour and odour is caused by the terpenoid compounds namely geosmin and 2-methylisoborneol,produced as secondary metabolites by certain bacteria including the cyanobacteria and *** strategies to remediate the problem rely on treating the symptoms not the cause and involve the use of time consuming purging methods and costly *** control using bacteriophages,specific to the problem causing bacteria,offers a natural alternative to chemical control,which might reduce further complications of copper based algaecides and its subsequent implications on water *** an adaptation of such biological control approach streptomycetes isolated from barramundi ponds were tested for their susceptibility to streptophages to understand whether host destruction via phage lysis would subsequently eliminate off-flavour taint productions by these *** the determination of the streptophage susceptibility of the isolates one of the most odourous streptomycete species(USC-14510)was selected to be tested further using different pond simulations resembling real-life *** was tested as the indicator of off-flavour taint production and as it has been previously reported that the cyanobacteria-actinomycete interactions occurring in ponds result in even greater levels of geosmin and 2-methylisoborneol,the geosmin levels for the isolate in the presence of cyanobacteria and streptophages were also *** indicated that the highly odourous Streptomyces species(USC-14510)once infected with streptophages,can lose its capacity to produce off-flavour *** simulation studies also revealed geosmin production

Effectiveness of a Fitbit Based Sleep and Physical Activity Intervention in an Early Intervention Psychosis (EIP) Service

Purpose: Compared to levels in the general population, people with experiences of psychosis have poorer physical fitness, more sedentary behaviour, lower physical activity, more sleep problems, and a higher incidence of insomnia. The aim of the current study was to examine the feasibility and impact of an intervention with the goal of addressing these issues. Method: Design: intervention with outcome measure data collection, with no control group. Forty-nine early intervention psychosis (EIP) service patients took part in an intervention: provision of a Fitbit, Fitbit software apps, sleep hygiene, and physical activity guidance, and three discussion sessions with clinicians. The sample consisted of 29 males and 20 females, with age range of 17 - 54 years, and average age of 29.5 years. Measures used were Fitbit activity and sleep data, and self-rated Warwick–Edinburgh Mental Wellbeing Scale (WEMWBS) and Positive and Negative Affect Schedule (PANAS). Results: WEMWBS scores significantly improved, with a medium effect size. PANAS negative affect (NA) dropped significantly. Analysis of Fitbit activity and sleep data yielded non-significant results. Conclusion: The intervention was acceptable to EIP patients and is feasible. Improvements in mental wellbeing and negative affect indicate the positive impact of the intervention on mental health. EIP services should consider assessing sleep quality and physical activity/exercise levels, and using this study’s intervention to promote wellbeing and mental health within recovery focused practice. Further research could be undertaken through a sufficiently powered randomised control trial (RCT) comparing this intervention and treatment as usual (TAU).

Highly efficient and genotype-independent barley gene editing based on anther culture

Recalcitrance to tissue culture and genetic transformation is the major bottleneck for gene manipulation in *** barley,immature embryos of Golden Promise have typically been used as explants for ***,the genotype dependence of this approach limits the genetic modification of commercial ***,we developed an anther culture-based system that permits the effective creation of transgenic and gene-edited plants from commercial barley *** protocol was tested in Golden Promise and four Australian varieties,which differed in phenology,callus induction,and green plant regeneration ***-mediated transformation was performed on microspore-derived callus to target the HvPDS gene,and T0 albinos with targeted mutations were successfully obtained from commercial *** editing of three targets was achieved with an average mutation rate of 53%in the five *** 51 analyzed T0 individuals,Cas9 induced a large proportion(69%)of single-base indels and two-base deletions in the target sites,with variable mutation rates among targets and *** ontarget and off-target activities were detected in T1 *** with immature embryo protocols,this genotype-independent platformcan deliver a high editing efficiency and more regenerant plants within a similar time *** shows promise for functional genomics and the application of CRISPR technologies for the precise improvement of commercial varieties.
