Mycobacterium avium subspecies paratuberculosis causes Crohn's disease in some inflammatory bowel disease patients

Crohn’s disease(CD)is a chronic inflammatory condition that plagues millions all over the *** debilitating bowel disease can start in early childhood and continue into late *** and symptoms are usually many and multiple tests are often required for the diagnosis and confirmation of this ***,little is still understood about the cause(s)of *** a result,several theories have been proposed over the *** theory in particular is that Mycobacterium avium subspecies paratuberculosis(MAP)is intimately linked to the etiology of *** fastidious bacterium also known to cause Johne’s disease in cattle has infected the intestines of animals for *** is believed that due to the thick,waxy cell wall of MAP it is able to survive the process of pasteurization as well as chemical processes seen in irrigation purification *** meat,dairy products and water serve as key vehicles in the transmission of MAP infection to humans(from farm to fork)who have a genetic predisposition,thus leading to the development of *** challenges faced in culturing this bacterium from CD are *** include its extreme slow growth,lack of cell wall,low abundance,and its mycobactin *** this review article,data from 60 studies showing the detection and isolation of MAP by PCr and culture techniques have been *** this review may not be 100%comprehensive of all studies,clearly the majority of the studies overwhelmingly and definitively support the role of MAP in at least30%-50%of CD *** is very possible that lack of detection of MAP from some CD patients may be due to the absence of MAP role in these *** latter statement is conditional on utilization of methodology appropriate for detection of human MAP ***,stratification of CD and inflammatory bowel disease patients for the presence or absence of MAP is necessary for appropriate and effective treatment which may lead to a cure.

12-Aryl-8,9,10,12-tetrahydrobenzo[a]xanthene-11-one derivatives were synthesized by a convenient and environmentally benign procedure involving multicomponent condensation reactions of substituted aromatic aldehydes with 2,7-dihydroxynaphthalene/2-naphthols/2,6-dihydroxynaphthlene and cyclic 1,3-dicarbonyl compounds in task specific acidic ionic liquid[NMP]HPOat 80 ℃.This protocol has proved to be efficient in terms of good yields,operational simplicity,easy workup,recyclability of reaction medium/catalyst,and short reaction time.

Identification of mir414 and Expression Analysis of Conserved mirNAs from Stevia rebaudiana

MicrorNAs （mirNAs） usually contain 19-24 nucleotides and have been identified as important eukaryotic gene regulators. Applications of various computational approaches have simplified the task by predicting mirNAs from available sequence data sources. In this study, we identified a conserved mir414 from a computational analysis of EST sequence data available from Stevia rebaudiana. In addition, we also identified six conserved mirNAs namely mir169, mir319, mir414, mir164, mir167 and mir398 using stem-loop rT-PCr analysis. Hence, mir414 was commonly identified using both methods. The expression analysis of these mirNAs documented their roles in growth and development of Stevia. Furthermore, the detected mirNAs were found to target genes involved in plant growth, development, metabolism and signal transducfion. This is the first study reporting these conserved mirNAs and their expression in Stevia.

Small rNAs （srNAs） are 18-30 nt non-coding regulatory elements found in diverse organisms, which were initially identified as small double-stranded rNAs in Caenorhabditis elegans. With the development of new and improved technologies, srNAs have also been identified and characterized in plant systems. Among them, micro rNAs （mirNAs） and small interfering rNAs （sirNAs） are found to be very important riboregulators in plants. Various types of srNAs differ in their mode of biogenesis and in their function of gene regulation, srNAs are involved in gene regulation at both transcriptional and post-transcriptional levels. They are known to regulate growth and development of plants. Furthermore, srNAs especially plant mirNAs have been found to be involved in various stress responses, such as oxidative, mineral nutrient deficiency, dehydration, and even mechanical stimulus. Therefore, in the present review, we focus on the current understanding of biogenesis and regulatory mechanisms of plant srNAs and their responses to various abiotic stresses.

Premna integrifolia Linn. is a medicinal plant used in ＂Dhasamula＂ drug preparation of Ayurvedic systems of medicine in the treatment of various ailments like bronchitis, dyspepsia, liver disorders, piles, constipation,hyperlipidemia and fever. The anti-atherosclerotic activity of hydroalcoholic extract（HAE） of root bark of *** was evaluated in high fat diet induced atherosclerosis rats. Sixty Wistar rats were divided into six groups： the first group served as control, the second group was fed with high fat diet and the other three groups were fed with high fat diet along with various concentrations of HAE and the last group was treated with atorvastatin for 30 days. Lipid and lipoprotein profile, atherogenic index, and cardiac markers and histopathological evaluation of aorta were determined in high fat diet induced atherosclerosis rats. HAE of *** produced a significant and dose-dependent anti-atherosclerotic activity in terms of reduction in lipids and lipoprotein profile, atherogenic index, HMG-Co A reductase activity, marker enzymes such as lactate dehydrogenase（LDH）, creatine phosphokinase（CPK）, aspartate transaminase（AST）, alanine transaminase（ALT） and alkaline phosphatase（ALP）, alteration in collagen and calcium contents, mild mineralization and focal rupture of intima and media of aorta was noticed in treated groups as compared to the control. The results suggested that anti-atherosclerotic activity of HAE of P. integrifolia Linn. was due to its modulatory activity on metabolic pathway of lipid. The results contribute to the validation of the traditional use of Agnimantha in high fat diet induced atherosclerosis rats.

One pot three component synthesis of spiro [indolo-3,10’-indeno[1,2-b]quinolin]-2,4,11’-triones as a new class of antifungal and antimicrobial agents

A simple and efficient three component procedure has been developed for the synthesis of highly substituted spiro[indolo-3,100-indeno[1,2-b]quinolin]-2,4,11＇-triones by one pot three component condensation of enaminones, isatin and indane-1,3-dione in ethanol：water（1：1） in presence of ceric ammonium nitrate（CAN） as catalyst. This method provides several advantages such as lesser reaction time, high yield of products and operational simplicity. The antimicrobial activity of some of the compounds has been investigated against six microbial strains, some of the tested compounds showed good antimicrobial activity.

Cytotoxic and antiproliferative activity of kanchnar guggulu,an Ayurvedic formulation

Objective： Kanchnar guggulu is a compound Ayurvedic formulation used in clinical practice for the treat- ment of benign and malignant tumors. The present study investigates its cytotoxic and antiproliferative ***： The hydro-alcoholic （50%） extract of kanchnar guggulu was prepared. Its antimitotic activity was assessed in an Allium cepa assay, while its antiproliferative effects were studied in a yeast prolifer- ation model. Methotrexate was used as a standard anticancer ***： In the Allium assay, all concentrations of the extract （1, 2 and 3 mg/mL） and methotrexate （0.02 mg/mL） significantly inhibited the division ofA. cepa root cells, decreasing root growth and mitotic index compared to control; this effect was concentration-dependent for kanchnar guggulu extract. In the antiproliferative studies, treatment with the hydro-alcoholic extract of kanchnar guggulu （1, 5 and 10 mg/mL） and methotrexate （0.025, 0.05 and 0.1 mg/mL） resulted in marked reduction of dividing Saccharornyces cerevisiae cells and inhibition of cell viability compared to control. The cytotoxicity of the hydro-alcoholic extract of kanchnar guggulu, shown by its antimitotic and antiproliferative effects, may be due to the presence of flavonoids and ***： Kanchnar guggulu exhibited a cytotoxic effect by inhibiting cell division （antimitotic） and reducing cell proliferation. These results substantiate its potential for the treatment of cancer and support its traditional use in the treatment of cancer.

Vernacular settlements provide us an opportunity to learn how sustainable building was inherently practiced in various regions of the *** paper looks at how rituals that are a part of the Indra Jatra,a community festival,reinforces the ideals of sustainable spatial planning in the vernacular settlements of the Newars who are the indigenous community belonging to the Kathmandu Valley in *** settlement-pattern and vernacular dwellings are appraised,and the current scenario is analyzed so as to derive how the ritual spaces get activated during the festival(that have been spatially planned)and whether they guide decision-making and policy *** is essential to determine whether these spaces face any urban pressures and to further investigate how they can be mitigated.

Spectrum of fungal keratitis: clinicopathologic study of 44 cases

AIM: To determine the causative agents of fungal keratitis and study the predisposing factors over a period of ten years in a single tertiary care hospital. ·METHODS: A retrospective analysis of fungal corneal ulcers was done from 2003-2012. Patients’ clinical data were noted from the file records. Correlation of histopathological diagnosis was done with the report on fungal culture. · rESULTS: Mycotic keratitis was established in 44 cases by a positive fungal culture. Direct microscopic examination of potassium hydroxide(KOH) mounts revealed fungal elements in 39 cases while 40 cases showed fungus on Gram stained smears. Males(54.55%) were more commonly affected than the females(45.45%). The age ranged from 18 to 82 years. Most common age group to be involved was 41-60 years. Predisposing risk factors were seen in 34(77.27%) cases. Most common findings on clinical examination were anterior chamber reaction and conjunctival injection seen in all the cases. Other common findings were stromal infiltration and hypopyon seen in 20(45.45%) and 18(40.91%) cases respectively. On histopathological examination the fungus was typed,as aspergillus in 34 cases while no definite typing was possible in 10 cases. The predominant isolate was aspergillus flavus(59.09%) followed by fusarium(15.91%). Mixed fungal and bacterial infection was seen in 3(6.82%) cases. ·CONCLUSION: Although culture is the gold standard for definitive diagnosis of fungal keratitis,direct microscopic examination of corneal scrapings or histomorphological evaluation of biopsies allow a rapidpreliminary diagnosis. Early administration of antifungal treatment helps in preventing dreadful complications.
