Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(OC)-specific functions. Specifically, in OCs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in OCs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and mITF orchestrated a TF network essential for OC differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BRD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. In summary, our study demonstrates that combining GWASs with genome-wide binding studies and model organisms could decipher lineage-specific pathways contributing to complex disease states.

Study of plasma parameters of coaxial plasma source using triple Langmuir probe and Faraday cup diagnostics

Coaxial plasma guns are a type of plasma source that produces plasma which propagates radially and axially controlled by the shape of the ground electrode, which has attracted much interest in several applications. In this work, a 120° opening angle of CPG nozzle is used as a plasma gun configuration that operates at the energy of 150 J. The ionization of polyethylene insulator between the electrodes of the gun produces a cloud of hydrogen and carbon *** triple Langmuir probe and Faraday cup are used to measure plasma density and plasma temperature. These methods are used to measure the on-axis and off-axis plasma divergence of the coaxial plasma gun. The peak values of ion densities measured at a distance of 25 mm on-axis from the plasma gun are(1.6±0.5)×10^(19)m^(-3)and(2.8±0.6)×10^(19)m^(-3)for hydrogen and carbon plasma respectively and the peak temperature is 3.02±0.5 eV. The mean propagation velocity of plasma is calculated using the transit times of plasma at different distances from the plasma gun and is found to be 4.54±0.25 cm/μs and 1.81±0.18 cm/μs for hydrogen and carbon plasma respectively. The Debye radius is obtained from the measured experimental data that satisfies the thin sheath approximation. The shot-to-shot stability of plasma parameters facilitates the use of plasma guns in laboratory experiments. These types of plasma sources can be used in many applications like plasma opening switches, plasma devices, and as plasma sources.

Role of Social Capital in Determining Happiness and Life Satisfaction:mediation of Self-reported Health Using Path Analysis

This study links social capital with people’s health and well-being using data from the seventh wave of the World Value Survey,logistic regression,and path *** study’s findings show that happiness and life satisfaction are two different measures of the same construct,or *** determinants of each are also characterised differently:while life satisfaction is more of a stable and relative measure and is more strongly influenced by civic cooperation,social participation,and educational attainment,happiness is more of an unstable measure and is more strongly influenced by community belonging,trust and confidence aggregates,including employment and location of residence of an ***,freedom of choice,financial satisfaction or social comparison,and state of health were the most important factors influencing happiness and life *** findings also show that self-reported health(SRH)has a greater impact on happiness than a measure of life *** analysis shows out of the three other components of social capital—community belonging,civic engagement,and social participation—trust and confidence aggregates are the ones that have the biggest impact on increased social *** both well-being measures,SRH mediated the *** life satisfaction measure had a higher level of mediation of SRH than happiness,but social capital had a smaller direct impact on life satisfaction than on *** was found to be partially mediating this relationship for both happiness and life satisfaction measures,which means that people with higher social capital reported feeling better off.

During the decommissioning of the Fukushima Daiichi nuclear power plant,it is important to consider the retrieval of resolidified debris both in air and underwater *** the subsequent retrieval of debris from the reactor building,the resolidified debris must be cut into smaller pieces using various cutting *** the cutting process,aerosol particles are expected to be generated at the submicron *** has been noted that such aerosols sizing within the Greenfield gap(0.1-1μm)are difficult to remove effectively using traditional spraying ***,to improve the aerosol removal efficiency of the spray system,a new aerosol agglomeration method was recently proposed,which involves injecting water mist to enlarge the sizes of the aerosol particles before removing them using water *** this study,a series of experiments were performed to clarify the proper spray configurations for effective aerosol scavenging and to improve the performance of the water *** experimental results showed that the spray flow rate and droplet characteristics are important factors for the aerosol-scavenging efficiency and performance of the water *** results obtained from this study will be helpful for the optimization of the spray system design for effective aerosol scavenging during the decommissioning of the Fukushima Daiichi plant.

Variation in the Flaring Potential of Different Sunspot Groups During Different Phases of Solar Cycles 23 and 24

In this present study,we have analyzed different types of X-ray solar flares(C,m,and X classes)coming out from different classes of sunspot groups(SSGs).The data which we have taken under this study cover the duration of 24 yr from 1996 to *** this,we observed a total of 15015 flares(8417 in SC-23 and 6598 in SC-24)emitted from a total of 33780 active regions(21746 in SC-23 and 12034 in SC-24)with sunspot *** defined the flaring potential or flare-production potential as the ratio of the total number of flares produced from a particular type of SSG to the total number of the same-class SSGs observed on the solar *** we studied yearly changes in the flaring potential of different mcIntosh class groups of sunspots in different phases of SC-23 and *** addition,we investigated yearly variations in the potential of producing flares by different SSGs(A,B,C,D,E,F,and H)during different phases(ascending,maximum,descending,and minimum)of SC-23 and *** are our findings:(1)D,E,and F SSGs have the potential of producing flares≥8 times greater than A,B,C and H SSGs;(2)The larger and more complex D,E,and F SSGs produced nearly 80%of flares in SC-23 and 24;(3)The A,B,C and H SSGs,which are smaller and simpler,produced only 20%of flares in SC-23 and 24;(4)The biggest and most complex SSGs of F-class have flaring potential 1.996 and 3.443 per SSG in SC-23 and 24,respectively.(5)The potential for producing flares in each SSG is higher in SC-24 than in SC-23,although SC-24 is a weaker cycle than SC-23.(6)The alterations in the number of flares(C+m+X)show different time profiles than the alterations in sunspot numbers during SC-23 and 24,with several peaks.(7)The SSGs of C,D,E,and H-class have the highest flaring potential in the descending phase of both SC-23 and 24.(8)F-class SSGs have the highest flaring potential in the descending phase of SC-23 but also in the maximum phase of SC-24.

Prediction of rock fragmentation in a fiery seam of an open-pit coal mine in India

Spontaneous combustion of coal increases the temperature in adjoining overburden strata of coal seams and poses a challenge when loading blastholes. This condition, known as hot-hole blasting, is dangerous due to the increased possibility of premature explosions in loaded blastholes. Thus, it is crucial to load the blastholes with an appropriate amount of explosives within a short period to avoid premature detonation caused by high temperatures of blastholes. Additionally, it will help achieve the desired fragment size. This study tried to ascertain the most influencial variables of mean fragment size and their optimum values adopted for blasting in a fiery seam. Data on blast design, rock mass, and fragmentation of 100 blasts in fiery seams of a coal mine were collected and used to develop mean fragmentation prediction models using soft computational techniques. The coefficient of determination (R2), root mean square error (RmSE), mean absolute error (mAE), mean square error (mSE), variance account for (VAF) and coefficient of efficiency in percentage (CE) were calculated to validate the results. It indicates that the random forest algorithm (RFA) outperforms the artificial neural network (ANN), response surface method (RSm), and decision tree (DT). The values of R2, RmSE, mAE, mSE, VAF, and CE for RFA are 0.94, 0.034, 0.027, 0.001, 93.58, and 93.01, respectively. multiple parametric sensitivity analyses (mPSAs) of the input variables showed that the Schmidt hammer rebound number and spacing-to-burden ratio are the most influencial variables for the blast fragment size. The analysis was finally used to define the best blast design variables to achieve optimum fragment size from blasting. The optimum factor values for RFA of S/B, ld/B and ls/ld are 1.03, 1.85 and 0.7, respectively. © 2024 Institute of Rock and Soil mechanics, Chinese Academy of Sciences

山体滑坡是尼泊尔山区的主要灾害,主要是由于其脆弱的地貌、强烈的降雨、不当的土地管理和在修建山区道路过程中没有采取必要的稳定措施所导致的。由于频繁的山体滑坡,下游地区人类的生命财产遭受了巨大损失,并导致生态系统生产力和功能下降,河道泥沙淤积。土地利用与土地覆被、地形、道路、排水、地质环境和降雨等多种空间因素与滑坡的发生紧密相关,并决定了该地区滑坡的易感性。了解滑坡现象及其引发的相关因素,对于采取适当的预防、控制和恢复措施至关重要。本研究在尼泊尔桑科西河流域的新格地子流域进行,旨在了解滑坡危害、引发的相关因素以及为最大限度地减少可能的危害而采取的最佳预防策略。滑坡评估是通过使用时序谷歌影像和实地调查,对滑坡体进行识别和数字化来进行的。利用GIS软件对滑坡危险性进行评估,编制影响因子图,并对现场观测数据进行分析的基础上,计算了滑坡的规模。此外,在相关家庭的问卷调查(n=60)中,进行目的性抽样,探讨人们对山体滑坡、对生计资产的主要影响因素的看法。并采用Friedman非参数检验评估滑坡对当地人感知的最主要影响因素。根据现场调查,在研究区域的不同位置共调查了46处滑坡体,滑坡总面积为4.77km^(2)。通过使用Friedman检验的比较矩阵分析,本研究确定了滑坡发生的主要原因是农村道路、施工、边坡因素、降雨和土地利用与土地覆被。距离道路200-500 m造成最高比率的滑坡灾害,东南方向坡度30°-45°的山体对滑坡的易感性最高。与其他土地利用相比,因为在林内松软地区修建道路,茂密的森林更易受到山体滑坡的影响。比较道路建设前后滑坡的影响,约80%的受访者认为对滑坡有重要影响。
