Effect of recombinant and native buffalo OVGP1 on sperm functions and in vitro embryo development: a comparative study

Background： An oviduct-specific glycoprotein, OVGP1, is synthesized and secreted by non-ciliated epithelial cells of the mammalian oviduct which provides an essential milieu for reproductive functions. The present study reports the effects of recombinant buffalo OVGP1 that lacks post-translational modifications, and native Buffalo OVGP1 isolated from oviductal tissue, on frozen-thawed sperm functions and in vitro embryo ***： The proportion of viable sperms was greater（P 〈 0.05） in the recombinant OVGP1-treated group compared to the native OVGP1-treated group at 2 h, 3 h, and 4 h of incubation. The proportion of motile sperms at3 h and 4 h of incubation; and membrane-intact sperms at 4 h was greater（P 〈 0.05） in the native OVGP1-treated group compared to the control and recombinant OVGP1-treated groups. The proportion of capacitated and acrosome-reacted sperms was greater（P 〈 0.05） in the native OVGP1-treated group compared to the recombinant OVGP1 group at 4 h. The rates of cleavage of embryos and their development to the blastocyst stage were greater（P 〈 0.05） in the presence of either native or recombinant OVGP1 in comparison to control at 10 μg/mL concentration as compared to 5 or 20 μg/***： The study suggests that both native and recombinant OVGP1 impart a positive effect on various sperm features and in vitro embryo development. However, native OVGP1 was found to have a more pronounced effect in comparison to recombinant non-glycosylated OVGP1 on various sperm functions except viability. Hence,our current findings infer that glycosylation of OVGP1 might be essential in sustaining the sperm functions but not the in vitro embryo development.

In this study, a series of Gd^(3+)-doped mixed Ni-Cu-Zn ferrites with composition of Zn_(0.5)Ni_(0.3)Cu_(0.2)Fe_(2-x)Gd_(x)O_(4)(x = 0, 0.025, 0.05, 0.075, 0.1) was prepared using self-ignition sol-gel method. The prepared nanoparticles with an average size ranging from 22 to 26 nm show a single-phase cubic structure belonging to the spinel matrix. A rise in the Gd^(3+)concentration leads to an increase in crystallite size and lattice parameter. In Fourier transform infrared spectra, two main absorption bands belonging to the spinel structure are observed. The high-frequency bands(v_(1)) represent the tetrahedral complex, while the lowfrequency bands(v_(2)) signify the octahedral complex. The optical bandgap of the nanoferrites is found within the range of 2.91 to 2.41 eV, depending on their size. The magnetic characteristics of the material,such as saturation magnetization and coercivity are significantly altered with the concentration of Gd^(3+)in the solution. Using Rhodamine B(RhB) as a model organic pollutant, an in-depth investigation of the photocatalytic activity of the compounds was carried out. The present outcomes show that adding an adequate amount of Gd^(3+)significantly enhances the number of hydroxyl radicals produced by the ferrite,in turn, increasing the photocatalytic activity of the material. Mechanism elucidated by scavenger studies reveals that ·OH and holes are the primary reactive radicals responsible for the degradation *** photocatalysts show an insignificant performance loss in five consecutive cycles. Thus, it is concluded that these photocatalysts are highly suitable for the remediation of dye-contaminated wastewater.

A set of 45 diverse Asian and African origin/bred populations of pearl millet were investigated multi-locationally for morphological traits including grain yield and assessed for their fertility restoration ability on three cytoplasmic male sterility(CMS) systems in two contrasting seasons. Significant genetic variation was found for all the yield linked traits. Multivariate analysis for grain yield and its component traits grouped these populations into five clusters. Most of the Asian origin populations grouped into single cluster(Cluster III) and separated from the rest of the African origin populations indicating the presence of correspondence between clustering pattern and geographical origin of the populations. Clusters dominated with Asian populations(As-As) had linkages with early flowering, short plant height, more number of tillers, small and thin panicles, small seeded and low grain yield compared to the clusters dominated by African populations(Af-Af) or African origin and Asian bred(Af-As) *** related populations having common parentage were found grouped in same *** restoration/maintainer frequency of 45 populations on three diverse CMS systems revealed that overall fertility restoration frequency was highest for A1(86%) followed by A4(37%) and for A5(7%) CMS system. Five populations were identified as potential sources for developing maintainer lines for all three CMS systems and eight populations were identified specifically for A;and A;CMS systems. A set of 11 and four populations were identified for restorer line development exclusively for A;and A;CMS system,respectively. Six populations were identified for the development of dual restorers for both A;and A;CMS system.

The Deccan Syneclise is considered to have significant hydrocarbon potential. However, significant hydrocarbon discoveries, particularly for Mesozoic sequences, have not been established through conventional exploration due to the thick basalt cover over Mesozoic sedimentary rocks. In this study, near-surface geochemical data are used to understand the petroleum system and also investigate type of source for hydrocarbons generation of the study area. Soil samples were collected from favorable areas identified by integrated geophysical studies. The compositional and isotopic signatures of adsorbed gaseous hydrocarbons （methane through butane） were used as surface indicators of pe- troleum micro-seepages. An analysis of 75 near-surface soil-gas samples was carried out for light hydrocarbons （C_1-C_4） and their carbon isotopes from the western part of Tapti graben, Deccan Syneclise, India. The geochemical results reveal sites or clusters of sites containing anomalously high concentrations of light hydrocarbon gases. High concentrations of adsorbed thermogenic methane （C1 518 ppb） and ethane plus higher hydrocarbons （∑C2＋ = 977 ppb） were observed. Statistical analysis shows that samples from 13% of the samples contain anomalously high concentrations of light hydrocarbons in the soil-gas constituents. This seepage suggests largest magnitude of soil gas anomalies might be generated/source from Mesozoic sedimentary rocks, beneath Deccan Traps. The carbon isotopic composition of methane, ethane and propane ranges are from 22.5‰to -30.2‰ PDB, -18.0‰ to 27.1‰,, PDB and 16.9‰-32.1‰ PDB respectively, which are in thermogenic source. Surface soil sample represents the intersection of a migration conduit from the deep subsurface to the surface connected to sub-trappean Mesozoic sedimentary rocks. Prominent hydrocarbon concentra- tions were associated with dykes, lineaments and presented on thinner basaltic cover in the study area, which probably acts as channel for the micro-seep

Nitrogen fractionation of certain conventional-and lesser-known by-products for ruminants

Dietary proteins for ruminants are fractionated according to solubility, degradability and digestibility. In the present experiment, 11 vegetable protein meals and cakes used in ruminant nutrition were included with a main focus on determining various nitrogen(N) fractions in vitro. Total N(N×6.25) content varied from 22.98%(mahua cake) to 65.16%(maize gluten meal), respectively. Guar meal korma contained the lowest and rice gluten meal had the highest acid detergent insoluble nitrogen(ADIN; N×6.25). Boratephosphate insoluble N(BIN, N×6.25) and Streptomyces griseus protease insoluble N(PIN; N×6.25) were higher(P<0.01) in maize gluten meal than in other feeds, whereas groundnut cake and sunflower cake had lower(P < 0.01) BIN, and PIN, respectively. Available N, calculated with the assumption that ADIN is indigestible, was maximum in guar meal korma and minimum in rice gluten meal. Furthermore, rapid and slowly degradable N(N × 6.25) was found to be higher(P < 0.01) in groundnut cake and coconut cake, respectively. Intestinal digestion of rumen undegradable protein, expressed as percent of PIN, was maximum in guar meal korma and minimum in rice gluten meal. It was concluded that vegetable protein meals differed considerably in N fractions, and therefore, a selective inclusion of particular ingredient is needed to achieve desired level of N fractions to aid precision N rationing for an improved production performance of ruminants.

Antibiotics are routinely used in intensive animal agriculture operations collectively known as Concentrated Animal Feed Operations （CAFO） which include dairy, poultry and swine farms. Wastewater generated by CAFOs often contains low levels of antibiotics and is typically managed in an anaerobic lagoon. The objective of this research is to investigate the uptake and fate of aqueous sulfamethazine （SMN） antibiotic by alfalfa （Medicago satiua） grass grown under hydroponic conditions. Uptake studies were conducted using hydroponically grown alfalfa in a commercially available nutrient solution supplemented with 10 mg/L of SMN antibiotic. Analysis of alfalfa sap, root zone, middle one-third, and top portion of the foliage showed varying uptake rate and translocation of SMN. The highest average amount of SMN （8.58 μg/kg） was detected in the root zone, followed by the top portion （1.89μg/kg）, middle one-third （1.30 μg/kg）, and sap （0.38 μg/kg） samples, indicating a clear distribution of SMN within the sampled regions. The ultraviolet （UV） spectra of parent SMN and translocated SMN identified in different parts of the plant present the possibility of metabolization during the uptake process. Uptake of SMN using alfalfa grown under hydroponic conditions has potential as a promising remediation technology for removal of similar antibiotics from wastewater lagoons.

Water quality impact due to excessive nutrients has been extensively studied. In recent years, however, micro-pollutants such as pharmaceuticals and hormonal products used in animal agriculture have added an additional impact to overall water quality. Pharmaceuticals used in the poultry, swine, beef, and dairy industries have been detected in various environmental matrices such as, soil, groundwater and surface water. In this study, 26 surface water samples were collected throughout the Bosque River Watershed （BRW） with samples representing a range of land use conditions and locations of major dairy operations. Samples were analyzed using commercially available Enzyme-Linked Immunosorbent Assay test. Of the 26 samples, three samples consistently tested positive for monensin antibiotic with concentration ranging from 0.30 to 3.41 μg/L. These three samples were collected from sites that received varying amount of agriculture wastes （11.7% to 31.3%） and located downstream from sites associated with moderate levels of animal agriculture. The preliminary results suggest that there is a potential for monensin occurrence in the BRW, although initial findings indicate only very low levels.
