Polyfunctional T Cell and Neutralizing Antibody Responses to ACAM2000^TM Smallpox Vaccine Immunization in Primary-Vaccinated Individuals

Smallpox eradication was successful via prophylactic administration of live attenuated vaccinia virus. As a result of the discontinuation of the smallpox immunization program, many individuals are now susceptible to smallpox virus infection should it be used as a biological weapon. Presently, only individuals at high risk for exposure are required to receive smallpox vaccine, such as laboratory personnel that handle variola/vaccinia virus. This study endeavored to investigate a one-year period of vaccinia virus-specific T cell responses using polychromatic flow cytometry and neutralizing (Nt) antibody responses using plaque reduction neutralization test (PRNT) in individuals receiving primary immunization (n = 5) with ACAM2000TM smallpox vaccine. Functional and phenotypic profiles of vaccinia virus-specific T cell responses were characterized. Each single functional measurement {CD107a/b expression, production of interferon g (IFN-g), macrophage inflammatory protein 1b (MIP-1b), interleukin 2 (IL-2), and tumor necrosis factor a (TNF-a)} demonstrated that vaccinia virus-specific CD8+ T cells were functional at least one time point after vaccination (p ≤ 0.05). However, vaccinia virus-specific CD4+ T cells were functional only for MIP-1b production (p ≤ 0.05). Vaccinia virus-specific CD8+ T cells induced in these individuals showed increased polyfunctionality in at least 2 phenotypes relative to pre-vaccination (p ≤ 0.05). Although only three of five individuals (60%) showed positive Nt antibody (titer ≥ 20) at first month after vaccination, all five individuals (100%) demonstrated Nt antibody at 2 months, post-immunization. Interestingly, all vaccinees could retain the Nt antibody for 6 months after primary vaccination. In conclusion, ACAM2000TM smallpox vaccine induced both polyfunctional T cell-and Nt antibody-responses in primary immunized individuals.

1.Introduction Microbial biofilms cause microbiologically influenced corrosion(MIC),which is one of the operational risks that can lead to equipment failures in the oil and gas industry and other industries [1-4].Mitigation measures such as biocide treatment and corrosion monitoring programs are implemented to ensure vital equipment integrity [5,6].The mechanism investigations in MIC can assist case analyses,field detections and biocide treatment strategies [7-9].

No difference in high-magnification morphology and hyaluronic acid binding in the selection of euploid spermatozoa with intact DNA

In this study, we compared conventional sperm selection with high-magnification morphology based on the motile sperm organellar morphology examination （MSOME） criteria, and hyaluronic acid （HA） binding for sperm chromosome aneuploidy and DNA fragmentation rates. Semen from 50 severe male factor cases was processed through density gradient centrifugation, and subjected to sperm selection by using the conventional method （control）, high magnification at x6650 or HA binding. Aneuploidy was detected by fluorescence in situ hybridization with probes for chromosomes 13, 18, 21, X and Y, and DNA fragmentation by the terminal deoxynucleotidyl transferase dUTP nick end labelling （TUNEL） method. Spermatozoa selected under high-magnification had a lower DNA fragmentation rate （2.6% vs. 1.7%; P=0.032）, with no significant difference in aneuploidy rate （0.8% vs0.7%; P=0.583）, than those selected by the HA binding method. Spermatozoa selected by both methods had much lower aneuploidy and DNA fragmentation rate than the controls （7% aneuploidy and 26.8% DNA fragmentation rates, respectively）. In the high-magnification group, the aneuploidy rate was lower when the best spermatozoa were selected than when only the second-best spermatozoa were available for selection, but the DNA fragmentation rate was not different. In conclusion, sperm selection under high magnification was more effective than under HA binding in selecting spermatozoa with low DNA fragmentation rate, but the small difference （0.9%） might not be clinically meaningful. Both methods were better than the conventional method of sperm selection.

Effect of high-pressure homogenization on stability of emulsions containing zein and pectin

The aim of this study was to investigate the effect of high-pressure homogenization on the droplet size and physical stability of different formulations of pectin–zein stabilized rice bran oil emulsions. The obtained emulsions, both before and after passing through highpressure homogenizer, were subjected to stability test under environmental stress conditions,that is, temperature cycling at 4 °C/40 °C for 6 cycles and centrifugal test at 3000 rpm for 10 min. Applying high-pressure homogenization after mechanical homogenization caused only a small additional decrease in emulsion droplet size. The droplet size of emulsions was influenced by the type of pectin used;emulsions using high methoxy pectin(HMP) were smaller than that using low methoxy pectin(LMP). This is due to a greater emulsifying property of HMP than LMP. The emulsions stabilized by HMP–zein showed good physical stability with lower percent creaming index than those using LMP, both before and after passing through high-pressure homogenizer. The stability of emulsions after passing through high-pressure homogenizer was slightly higher when using higher zein concentration, resulting from stronger pectin–zein complexes that could rearrange and adsorb onto the emulsion droplets.

In the oil and gas industry,microbiologically influenced corrosion(MIC) is a major threat to hydrotest,a procedure which is required to certify whether a pipeline can be commissioned.Seawater is frequently used as a hydrotest fluid.In this bio film prevention lab study,an oilfield biofilm consortium was grown in an enriched artificial seawater anaerobically at 37℃ for 60 days.The combination of 100 ppm(w/w) 2,2-dibromo-3-nitrilopropionamide(DBNPA)+100 nM(180 ppb) Peptide A(a biofilm dispersal agent) led to extra SRB(sulfate reducing bacteria),APB(acid producing bacteria) and GHB(general heterotrophic bacteria) sessile cell count reductions of 0.9-log,0.8-log and 0.6-log,respectively,compared with the outcome obtained by using 100 ppm DBNPA only.The Peptide Aenhancement also led to extra reductions of 44 % in weight loss,43 % in maximum pit depth,and 54 % in corrosion current density.

The title compound, neogeodin hydrate （C17H14C1208, CAS： 94540-50-8）, was derived from marine fungus Aspergilhts terreus CRIM301. It was unequivocally characterized by IR, NMR spectroscopies, and single-crystal X-ray crystallography and tested for various biological activities. Neogeodin hydrate crystallizes in the triclinic space group P1 with a = 8.1159（5） A, b = 8.2472（4） A, c= 14.1278（7） A, a = 81.448（2）°, β = 84.860（2）°, γ= 70.400（2）°, V = 880.13（8） A3; Z = 2. It comprises a diphenyl ether, asterric acid skeleton and dichloro substituents. The methoxyphenoxy rings of the inversely related molecules form a ribbon-like structure that is stabilized by O-H...O hydrogen bonds through the doubly disordered carboxyl groups and by C-H...O interactions, generating the same R22（8） ring motif. The chlorinated methylbenzoate rings, making mostly a right angle, link the parallel upper and lower ribbons via bifurcated O-H...O and C-H...O hydrogen bonds, yielding endless channels. The channels formed are further sustained by C-H...O and π...π interactions Neogeodin hydrate exhibits inhibition against superoxide anion radical formation in the xanthine/xanthine oxidase （XXO） assay, but has no aromatase inhibitory activity.

A new self-emulsifying formulation of mefenamic acid with enhanced drug dissolution

To enhance the dissolution of poorly soluble mefenamic acid,self-emulsifying formulation(SEF),composing of oil,surfactant and co-surfactant,was formulated.Among the oils and surfactants studied,Imwitor■ 742,Tween■ 60,Cremophore■ EL and Transcutol■ HP were selected as they showed maximal solubility to mefenamic acid.The ternary phase diagram was constructed to find optimal concentration that provided the highest drug loading.The droplet size after dispersion and drug dissolution of selected formulations were investigated.The results showed that the formulation containing Imwitor■ 742,Tween■ 60 and Transcutol■ HP(10:30:60)can encapsulate high amount of mefenamic acid.The dissolution study demonstrated that,in the medium containing surfactant,nearly 100% of mefenamic acid were dissolved from SEF within 5 min while 80% of drugs were dissolved from the commercial product in 45 min.In phosphate buffer(without surfactant),80% of drug were dissolved from the developed SEF within 5 min while only about 13% of drug were dissolved in 45 min,from the commercial product.The results suggested that the SEF can enhance the dissolution of poorly soluble drug and has a potential to enhance drug absorption and improve bioavailability of drug.

A novel complex [Cd2(phen)4(fca)2](ClO4)2(H2O)2 (fca = dianion of 3-ferrocenyl- 2-crotonic acid, phen = 1,10-phenanthroline) has been synthesized and characterized by X-ray single-crystal structure analysis. It crystallizes in triclinic system, space group P?with a = 11.6297(5), b = 12.5165(5), c = 13.8986(6) ? a = 63.345(1), b = 87.747(1), g = 71.862(1), V = 1706.20(1) ?, Z = 1, Dc = 1.673 g/cm3, F(000) = 868 and m(MoKa) = 1.185 mm-1 (l = 0.71073 ). R = 0.0800 and wR = 0.1793 for 7653 observed reflections (I > 2s(I)). The structure consists of a dinuclear cation [Cd2(phen)4(fca)2]2+, two discrete perchlorate anions and two water molecules. The cationic part of the complex shows a tetrametallic core in which two fca groups act as the (O, O) bridging ligands between two cadmium(Ⅱ) ions with a distorted octahedral environment. The Cd…Cd intradimer distance is 4.128 ?

Improved Growth and Stress Tolerance in the Arabidopsis oxtl Mutant Triggered by Altered Adenine Metabolism

Plants perceive and respond to environmental stresses with complex mechanisms that are often associ- ated with the activation of antioxidant defenses. A genetic screen aimed at isolating oxidative stress-tolerant lines of Arabidopsis thaliana has identified oxtl, a line that exhibits improved tolerance to oxidative stress and elevated temperature but displays no apparent deleterious growth effects under non-stress conditions. Oxtl harbors a muta- tion that arises from the altered expression of a gene encoding adenine phosphoribosyltransferase （APT1）, an enzyme that converts adenine to adenosine monophosphate （AMP）, indicating a link between purine metabolism, whole-plant growth responses, and stress acclimation. The oxtl mutation results in decreased APT1 expression that leads to reduced enzymatic activity. Correspondingly, oxtl plants possess elevated levels of adenine. Decreased APT enzyme activity dir- ectly correlates with stress resistance in transgenic lines that ectopically express APT1. The metabolic alteration in oxtl plants also alters the expression of several antioxidant defense genes and the response of these genes to oxidative chal- lenge. Finally, it is shown that manipulation of adenine levels can induce stress tolerance to wild-type plants. Collectively, these results show that alterations in cellular adenine levels can trigger stress tolerance and improve growth, leading to increases in plant biomass. The results also suggest that adenine might play a part in the signals that modulate responses to abiotic stress and plant growth.

Two new complexes [Zn 2 (phen) 4 (FCA) 2 ](ClO 4 ) 2 ·(H 2 O) 2 ( 1 ) and [Co 2 (phen) 4 (FCA) 2 ](ClO 4 ) 2 ·(H 2 O) 2 (2) (FCA=anion of 3‐ferrocenyl‐2‐crotonic acid, phen=1,10‐phenanthroline) have been synthesized, and characterized by elemental analysis, IR, UV‐Vis spectra, thermal analyses, and single‐crystal X‐ray diffraction. Two M(II) (M=Zn or Co) ions are bridged by two FCA anions with syn‐anti bridging ligands, leading to dimeric cores, [M 2 (phen) 4 (FCA) 2 ] 2+ , and each M(II) ion is six‐coordinated in a distorted octahedral geometry by two chelate phen ligands and two μ 2 ‐carboxylate oxygen atoms from two FCA groups. The M(II)…M(II) intradimer distances are 0.4391 and 0.4462 nm in 1 and 2 , respectively. Electrochemical properties of the complexes have been discussed.
