Cyclooxygenase-2 expression is dependent upon epidermal growth factor receptor expression or activation in androgen independent prostate cancer

Aim： To investigate the expression of cyclooxygenase-2 （COX-2） and epidermal growth factor receptor （EGFR） and the possible mechanism in the development in androgen independent prostate cancer （AIPC）. Methods： Immunohistochemistry was performed on paraffin-embedded sections with goat polyclonal against COX-2 and mouse monoclonal antibody against EGFR in 30 AIPC and 18 androgen dependent prostate cancer （ADPC） specimens. The effect of epidermal growth factor （EGF） treatments on the expression of COX-2 and signal pathway in PC-3 and DU-145 cells was studied using reverse transcription-polymerase chain reaction （RT-PCR） and Western blot analysis. ELISA was used to measure prostaglandin E2 （PGE2） levels in the media of PC-3 and DU-145 incubated with EGF for 24 h. Results： COX-2 was positively expressed in AIPC and ADPC, which were predominantly in endochylema of prostate cancer （PCa） cells. Intense staining was seen in AIPC （80%） and in ADPC （55.5%）, but there was no significant association between the two groups. EGFR expression was also positive in the two groups （61.8% in ADPC and 90% in AIPC, P 〈 0.01）. A significant association was found between EGFR expression and a higher Gleason score （P 〈 0.05） or tumor stage （P 〈 0.05）. The expression of PGE2 was increased in PC-3 and DU-145 cells after being incubated with EGF. Both p38MAPK and PI-3K pathway were involved in the PC-3 cell COX-2 upregulation course. In DU- 145, only p38MAPK pathway was associated with COX-2 upregulation. Conclusion： EGFR activation induces COX-2 expression through PI-3K and/or p38MAPK pathways. COX-2 and EGFR inhibitors might have a cooperative anti-tumor effect in PCa.

Discovery of the eclogite and its petrography in the Northern Dabie Mountain

Eclogite from the Northern Dabie Mountain is a new finding by the authors. These eclogites in foliated perdotite are enveloped by banded gneiss and occur in the mafic-ultramafic rock belt. They are mainly composed of omphacite, garnet, diopside, orthopyroxene, amphibole, plagioclase and magnetite, and a small amount of rutile, spinel, olivin and 鏾rundum. The mineral association of peak metamorphism of the eclogite is omphacite+garnet+rutile. The existence of eclogite in the Northern Dabie Mountain implies that there was an eclogitic metamorphism prior to the granulitic facies one in the mafic-ultramafic rock belt.

Background:Non-alcoholic fatty liver disease(NAFLD)is a liver disorder characterized by the accumulation and degeneration of fat in the liver cells,a condition that may further deteriorate and lead to cirrhosis and liver *** studies showed that metabolic dysfunction can promote NAFLD *** Zhugan Decoction(LGZGD)has therapeutic effects on *** mechanism of LGZGD still remains *** study was to examine the impact of LGZGD on the metabolic processes involved in the development of ***:A mice model of NAFLD was treated with *** therapeutic potential of LGZGD was evaluated by assessing the activity of transaminases,lipids levels of blood,and pathological changes in the liver of the mice model of ***,this study also evaluated the influence of LGZGD on liver inflammation and oxidative ***:The results of untargeted metabolomics analysis showed that LGZGD reduced the disordered lipid metabolism in NAFLD *** improved the oxidative stress and also reduced the levels of pro-inflammatory cytokines in the *** metabolomics analysis of liver samples revealed that LGZGD treatment improved metabolic disorders,including alanine,aspartate,glutamate,glycerophospholipid metabolism,and citrate *** RT-qPCR and Western blot results showed that LGZGD could regulate the expression of key enzymes in the metabolic pathway of the citrate cycle,including ATP-citrate lyase(ACLY),alanine-glyoxylate aminotransferase-2(AGXT2),phosphatidylethanolamine N-methyltransferase(PEMT),and succinate dehydrogenase(SDH).Conclusion:We found that LGZGD can treat NAFLD by reducing inflammatory responses,inhibiting oxidative stress,regulating alanine,aspartate,glutamate,and glycerophospholipid metabolism,and citrate cycle pathways.

Nonlinear output-feedback tracking in multiagent systems with an unknown leader and directed communication

This paper addresses cooperative global robust output regulation for heterogeneous and uncertain multiagent nonlinear systems in the output-feedback normal form. Specifically, we develop a Lyapunov-based dynamic output-feedback law using a nonlinear internal model approach. We show that an effective control law can be constructed under general(static) directed communication topologies even when the leader is unknown. Hence, the present study offers a more general investigation of the problem in comparison with the developments in the recent literature.

Meca,et al.(2004)studied a class of inventory games which arise when a group of retailers who observe demand for a common good decide to cooperate and make joint *** this paper,the authors extend their model to the situation where the manufacturer provides the retailers with a price discount on purchases in excess of a certain order *** authors define the corresponding inventory game with quantity discount,and show that the inventory game has a nonempty ***,the authors propose a core allocation rule,the demand-proportionality rule,and characterize it by introducing efficiency,the retailer splitting property and continuity.

Event-triggered tracking control for a class of nonholonomic systems in chained form

In this study, the event-triggered asymptotic tracking control problem is considered for a class of nonholonomic systems in chained form for the time-varying reference input. First, to eliminate the ripple phenomenon caused by the imprecise compensation of the time-varying reference input, a novel time-varying event-triggered piecewise continuous control law and a triggering mechanism with a time-varying triggering function are developed. Second, an explicit integral input-to-state stable Lyapunov function is constructed for the time-varying closed-loop system regarding the sampling error as the external input. The origin of the closed-loop system is shown to be uniformly globally asymptotically stable for any global exponential decaying threshold signals, which in turn rules out the Zeno behavior. Moreover, infinitely fast sampling can be avoided by appropriately tuning the exponential convergence rate of the threshold signal. A numerical simulation example is provided to illustrate the proposed control approach.

Effect of male HBV infection on the outcomes of IVF/ICSI cycles:a retrospective cohort study based on propensity score matching

This study aimed to investigate the effects of male hepatitis B virus(HBV)infection on male fertility,embryonic development,and in vitro fertilization/intracytoplasmic sperm injection(IVF/ICSI)*** performed a retrospective cohort study that included 3965 infertile couples who received fresh embryo transfer cycles for the first time at the Fujian Maternity and Child Health Hospital(Fuzhou,China)from January 2018 to January *** couples were categorized based on their HBV infection status into the HBV group(HBV-positive men and HBV-negative women)and the control group(HBV-negative couples).A 1:1 propensity score matching was performed with relatively balanced *** characteristics,semen parameters,laboratory outcomes,clinical outcomes,and obstetric and neonatal outcomes were compared between *** propensity score matching,821 couples were included in each *** groups had similar semen parameters and obstetric and neonatal *** HBV group showed a significantly lower live birth rate than the control group(P<0.05).The HBV group had a significantly higher abortion rate than the control group(P<0.05).The rates of high-quality embryos and blastocyst formation were significantly lower in the HBV group than those in the control group(both P<0.05).In conclusion,in couples who undergo IVF/ICSI,male HBV infection reduces the live birth rate and increases the risk of ***,the incidence of low birth weight in women with IVF/ICSI does not increase with male HBV infection.

Peptidomimetics and metalloprotease inhibitors as anticancer drugs

Peptidomimetics with three types, as the structural or functional mimetics of natural active peptides, can preserve the bioactivity and improve the bioavailability and the specificity towards the targets of the lead peptides. Peptidomimetics of high bioactivity can be designed through various ways including conformation restriction, modification and non-peptide design. Recently the concentration on the de-velopment of cancer chemotherapeutic drugs was transferred from cytotoxic drugs to target-based drugs, and many proteases and peptidases that play key roles in the process of tumor genesis and development was discovered, which means that peptidomimetics as potential cancer chemotherapeu-tic drugs should be paid close attention to. Our laboratory has focused on the development of small-molecule peptidomimetic inhibitors of APN, MMPs and HDACs as target-based anticancer agents. These three zinc-dependent metalloproteinases play very important roles in the process of tumor genesis, invasion, metastasis, angiogenesis and matrix degradation, so small-molecule peptidomimetic inhibitors based on them would be quite potential in the development of chemotherapeutic drugs with high selectivity.
