Plant high-affinity K^(+) transporters(HKTs)mediate Na^(+) and K^(+) uptake,maintain Na^(+)/K^(+) homeostasis,and therefore play crucial roles in plant salt *** this study,we present cryoelectron microscopy structures of HKTs from two classes,classI HKT1;1 from Arabidopsis thaliana(AtHKT1;1)and class II HKT2;1 from Triticum aestivum(TaHKT2;1),in both Na^(+) -and K^(+) -bound states at 2.6-to 3.0-A ***1;1and TaHKT2;1function *** HKT subunit consists of four tan-dem domain units(D1-D4)with a repeated K^(+) -channel-like M-P-M *** each subunit,D1-D4 assemble into an ion conduction pore with a pseudo-four-fold *** both TaHKT2;1and AtHKT1;1 have only one putative Na^(+) ion bound in the selectivity filter with a similar coordination pattern,the two HKTs display different K^(+) binding modes in the ***2;1 has three K^(+) ions bound in the selec-tivity filter,but AtHKT1;1 has only two K^(+) ions bound in the filter,which has a narrowed external entrance due to the presence of a Ser residue in the first filter *** structures,along with compu-tational,mutational,and electrophysiological analyses,enable us to pinpoint key residues that are critical for the ion selectivity of *** findings provide new insights into the ion selectivity and ion transport mechanisms of plant HKTs and improve our understanding about how HKTs mediate plant salt tolerance and enhance crop growth.

WTV2.0:A high-coverage plant volatilomics method with a comprehensive selective ion monitoring acquisition mode

Volatilomics is essential for understanding the biological functions and fragrance contributions of plant ***,the annotation coverage achieved using current untargeted and widely targeted volatomics(WTV)methods has been limited by low sensitivity and/or low acquisition ***,we introduce WTV 2.0,which enabled the construction of a high-coverage library containing 2111 plant volatiles,and report the development of a comprehensive selective ion monitoring(cSIM)acquisition method,including the selection of characteristic qualitative ions with the minimal ion number for each compound and an optimized segmentation method,that can acquire the smallest but sufficient number of ions for most plant volatiles,as well as the automatic qualitative and semi-quantitative analysis of cSIM ***,the library and acquisition method we developed can be self-expanded by incorporating compounds not present in the library,utilizing the obtained cSIM *** showed that WTV 2.0 increases the median signal-to-noise ratio by 7.6-fold compared with the untargeted method,doubled the annotation coverage compared with the untargeted and WTV 1.0 methods in tomato fruit,and led to the discovery of menthofuran as a novel flavor compound in passion *** 2.0 is a Python library with a user-friendly interface and is applicable to profiling of volatiles and primary metabolites in any species.

In August 2023,Japan started to dump its 1.3 million tons of radioactive wastewater into the oceans[1,2].Most radioisotopes are claimed to be removed from the wastewater except tritium,which needs further dilution before ***,this dilution does not effectively remove any tritium from the wastewater but serves as a measure to bring its abnormally high concentration down to meet emission *** most worrisome situation is that many more bioaccumulative long-lived radioisotopes(BLLRs),such as carbon-14 and cobalt-60,slip through the treatment *** are attempts to reduce the concentrations of BLLRs via repurification to meet the regulatory standards[3];however,even low levels of these BLLRs can undergo biomagnification of up to 50,000 folds in marine fish species[4].

为准确预测锂离子电池剩余寿命、降低电池工作风险,提出一种新的锂离子电池剩余寿命在线预测模型。基于锂离子电池历史运行数据提取6种健康因子,用于表征电池的退化状态;采用随机森林(RF)算法完成健康因子的评价与筛选;利用经遗传算法优化的广义回归神经网络(GA-GRNN)完成锂离子电池剩余容量的估计。在此基础上,应用结合双向长短期记忆(Bi-LSTM)网络模型和非线性自回归(NAR)神经网络的混合模型(混合Bi-LSTM-NAR模型)预测锂电池剩余寿命。以NASA公开数据集为例完成案例研究,结果表明:通过因子筛选,可以为锂离子电池容量估计及剩余寿命预测的精度提供保障;与已有方法的预测结果相比,所提混合预测模型的预测精度显著提高。

Clinical observation and therapeutic evaluation of intravenous pump of recombinant human endostatin combined with TP regimen in treating patients with advanced ovarian cancer

Microbial community dynamics and flavor formation mechanisms during fermentation of fermented chilies from different regions based on macro-genome sequencing， SPME-GC-MS， electronic nose， and electronic tongue

Fermented chili products are popular traditional fermented foods around the world. However， differences in microbial communities in fermented chilies from different regions and how they affect the flavor compounds in chili fermentation have not been reported. In this study， the dynamics of flavor compounds and microbial communities in fermented chilies from Sichuan， Guizhou， and Hunan were systematically investigated by macro-genome sequencing， solid phase microextraction-gas chromatograph-mass spectrometry (SPME-GC-MS)， electronic nose， and electronic tongue techniques. Simultaneously， the microbial metabolic mechanisms and the relationship between flavor compounds and microbiome were unraveled through staged and simulated fermentation analysis. The results showed that 53 chemical odorants， including alcohols， esters， aldehydes， and acids， were identified as chemical markers to differentiate the regional samples. Twelve microbial species， including Staphylococcus xylosus， Unclassified staphylococcus species， Weissella confusus， Lactococcus cremoris， Lactococcus garvieae， Lactiplantibacillus sakei， Pediciococcus propionicigenes， Pediciococcus idahonensis， Pediciococcus aciditolerans， Nocardioides antri， Debaryomyces hansenii， and Colletotrichum scovillei， were identified as microbial markers to differentiate the regional samples. Inefficient drug penetration hurdled by the stroma in the tumor tissue leads to a diminished therapeutic effect for drugs and a reduced infiltration level of immune ***,we constructed a PEGylated dendritic epirubicin(Epi)prodrug(Epi-P4D)to regulate the metabolism of cancer-associated fibroblasts(CAFs),thus enhancing Epi penetration into both multicellular tumor spheroids(MTSs)and tumor tissues in mouse colon cancer(CT26),mouse breast cancer(4T1)and human breast cancer(MDA-MB-231)*** cytotoxicity against CT26 MTSs and remarkable antitumor efficacy of Epi-P4D were ascribed to reduced fibronectin,α-SMA,and collagen ***,thinning of the tumor tissue stroma and efficient eradication of tumor cells promoted the immunogenic cell death effect for dendritic cell(DC)maturation and subsequent immune activation,including elevating the CD4^(+)T cell population,reducing CD4^(+)and CD8^(+)T cell hyperactivation and exhaustion,and amplifying the natural killer(NK)cell proportion and effectively activating *** a result,this dendritic nanomedicine thinned the stroma of tumor tissues to enhance drug penetration and facilitate immune cell infiltration for elevated antitumor efficacy.

Bactericidal ability of target acidic phospholipids and phagocytosis of CDC42 GTPase-mediated cytoskeletal rearrangement underlie functional conservation of CXCL12 in vertebrates

Chemokine CXCL12 plays a crucial role in both direct bactericidal activity and phagocytosis in humans. However, the mechanisms and evolutionary functions of these processes in vertebrates remain largely unknown. In this study, we found that the direct bactericidal activity of CXCL12 is highly conserved across various vertebrate lineages, including Arctic lamprey (Lampetra japonica), Basking shark (Cetorhinus maximus), grass carp (Ctenopharyngodon idella), Western clawed frog (Xenopus tropicalis), Green anole (Anolis carolinensis), chicken(Gallus gallus), and human (Homo sapiens). CXCL12 also has been shown to promote phagocytosis in lower and higher vertebrates. We then employed C. idella CXCL12a (CiCXCL12a) as a model to further investigate its immune functions and underlying mechanisms. CiCXCL12a exerts direct broad-spectrum antibacterial activity by targeting bacterial acidic phospholipids, resulting in bacterial cell membrane perforation, and eventual lysis. Monocytes/macrophages are attracted to the infection sites for phagocytosis through the rapid production of CiCXCL12a during bacterial infection. CiCXCL12a induces CDC42 and CDC42 GTPase activation, which in turn mediates F-actin polymerization and cytoskeletal rearrangement. The interaction between F-actin and Aeromonas hydrophila facilitates bacterial internalization into monocytes/macrophages. Additionally, A. hydrophila is colocalized within early endosomes, late endosomes and lysosomes, ultimately degrading within phagolysosomes. CiCXCL12a also activates PI3K-AKT, JAK-STAT5 and MAPK-ERK signaling pathways. Notably, only the PI3K-AKT signaling pathway inhibits LPS-induced monocyte/macrophage apoptosis. Thus, CiCXCL12a plays key roles in reducing tissue bacterial loads, attenuating organ injury, and decreasing mortality rates. Altogether, our findings elucidate the conserved mechanisms underlying CXCL12-mediated bactericidal activity and phagocytosis, providing novel perspectives into the immune functions

Evolutionary mechanism and frequency response of graphite electrode at extreme temperatures

The battery management system is employed to monitor the external temperature of the lithium-ion battery in order to detect any potential ***,this outside–in detection method often suffers from a lag and is therefore unable to accurately predict the battery’s real-time ***,an inside–out frequency response approach is used to accurately monitor the battery’s state at various temperatures in real-time and correlate it with the solid electrolyte interphase(SEI)evolution of the graphite *** SEI evolution at temperatures of−15,25,60,and 90℃exhibits certain regular characteristics with temperature *** a temperature of−15℃,the Li^(+)-solvent interaction of lithium-ion slowed down,resulting in a significant reduction in *** 25℃,a LiF-rich inorganic SEI was identified as forming,which facilitated lithium-ion ***,high temperatures would induce decomposition of lithium hexafluorophosphate(LiPF_(6))and lithium-ion *** the extreme temperature of 90℃,the SEI would be organic-rich,and Li_(x)P_(y)F_(z),a decomposition product of lithium salts,was further oxidized to Li_(x)PO_(y)F_(z),which led to a surge in the charge-transfer resistance at SEI(R_(sei))and a reduction in Coulombic efficiency(CE).This changing relationship can be recorded in real time from the inside out by electrochemical impedance spectroscopy(EIS)*** provides a new theoretical basis for the structural evolution of lithium-ion batteries and the regular characterization of EIS.

Understanding the vertical distribution of ozone is crucial when assessing both its horizontal and vertical transport,as well as when analyzing the physical and chemical properties of the *** of the most effective ways to obtain high spatial resolution ozone profiles is through satellite *** Environmental Trace Gases Monitoring Instrument(EMI)deployed on the Gaofen-5 satellite is the first Chinese ultraviolet-visible hyperspectral ***,retrieving ozone profiles using backscattered radiance values measured by the EMI is challenging due to unavailable measurement errors and a low signal-to-noise *** algorithm developed for the Tropospheric Monitoring Instrument did not allow us to retrieve 87%of the EMI ***,we developed an algorithm specific to the characteristics of the *** fitting residuals are smaller than 0.3%in most *** retrieved ozone profiles were in good agreement with ozonesonde data,with maximum mean biases of 20%at five latitude *** applying EMI averaging kernels to the ozonesonde profiles,the integrated stratospheric column ozone and tropospheric column ozone also showed excellent agreement with ozonesonde data,The lower layers(0-7.5 km)of the EMI ozone profiles reflected the seasonal variation in surface ozone derived from the China National Environmental Monitoring Center(CNEMC).However,the upper layers(9.7-16.7 km)of the ozone profiles show different trends,with the ozone peak occurring at an altitude of 9.7-16.7 km in March,2019.A stratospheric intrusion event in central China from August 11 to 15,2019,is captured using the EMI ozone profiles,potential vorticity data,and relative humidity *** increase in the CNEMC ozone co ncentration showed that downward transport enhanced surface ozone pollution.
