Secretory Transactivating Transcription-apoptin fusion protein induces apoptosis in hepatocellular carcinoma HepG2 cells

AIM:To determine whether SP-TAT-apoptin induces apoptosis and also maintains its tumor cell specificity. METHODS:In this study,we designed a secretory protein by adding a secretory signal peptide(SP) to the N terminus of Transactivating Transcription(TAT)-apoptin(SP-TAT-apoptin),to test the hypothesis that it gains an additive bystander effect as an anti-cancer therapy. We used an artificial human secretory SP whose amino acid sequence and corresponding cDNA sequence were generated by the SP hidden Markov model. REsuLTS:In human liver carcinoma HepG2 cells,SP-TAT-apoptin expression showed a diffuse pattern in the early phase after transfection. After 48 h,however,it translocated into the nuclear compartment and caused massive apoptotic cell death,as determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay and annexin-V binding assay. SP-TAT-apoptin did not,however,cause any cell death in non-malignant human umbilical vein endothelial cells(HUVECs). Most importantly,the conditioned medium from Chinese hamster ovary(CHO) cells transfected with SP-TAT-apoptin also induced significant cell deathin HepG2 cells,but not in HUVECs. CONCLUSION:The data demonstrated that SP-TAT-apoptin induces apoptosis only in malignant cells,and its secretory property might greatly increase its potency once it is delivered in vivo for cancer therapy.

A polymeric gel is an aggregate of polymers and solvent molecules, which can retain its shape after a large deformation. The deformation behavior of polymeric gels was often described based on the Flory-Rehner free energy function without considering the influence of chain entanglements on the mechanical behavior of gels. In this paper,a new hybrid free energy function for gels is formulated by combining the EdwardsVilgis slip-link model and the Flory-Huggins mixing model to quantify the time-dependent concurrent process of large deformation and mass transport. The finite element method is developed to analyze examples of swelling-induced deformation. Simulation results are compared with available experimental data and show good agreement. We herein report the room temperature synthesis of colloidal SnO2 quantum dots and their application in non-fullerene organic solar cells as an excellent electron transport *** thiourea-assisted hydrolysis at room temperature affords the nanocrystalline SnO2 quantum dots with a diameter of 3-4 *** utilization of the SnO2 quantum dots as an electron transporting layer effectively reduces the interfacial trap density and charge recombination in the solar cell devices,leading to not only the reduced energy loss but also excellent photocurrent *** optimized organic solar cells employing SnO2 quantum dots with polyethylenimine ethoxylated achieves power conversion efficiencies up to 12.023%with a VOC,a JSC,and a FF of 0.89 V,18.89 mA cm^–2,and *** work suggest that the SnO2 quantum dot is a promising electron transporting material to construct efficient organic solar cells for practical *** work also demonstrates the key strategy for thiourea-assisted hydrolysis to synthesize fine and nanocrystalline SnO2 quantum dots.

Although all-inorganic perovskites possess suitable bandgap and thermal stability for photovoltaic ap-plications,the unstable film morphologies,and poor moisture stabilities lag behind the organic-inorganic hybrid ***,we demonstrate that high qualityα-CsPbI_(2 ) Br films with improved phase sta-bility,high crystallinity,and pinhole-free film morphology was achieved by employing tetramethylammo-nium chloride(TMACl)as an *** a result,the TMACl-added CsPbI_(2) Br films exhibited high power conversion efficiency of 14.12%with an open-circuit voltage of 1.19 V,a short-circuit current density of 15.08 mA/cm^(2),and a fill factor of *** importantly,the TMACl addition imparts hydrophobicity to the film surface of CsPbI_(2) Br,which delivered excellent stability up to 30 days in the ambient condition and thermal stability at 85℃ for 156 h,***,the TMACl-added device also exhibited excellent PCE of 27.16%under indoor light source(1000 lux),which presents a promising approach for designing stable but high performance all-inorganic perovskite solar cells.

The transmutation of long-lived fission products through spallation induced by light nuclides was investi-gated for the purpose of determining the feasibility of this approach for long-lived fission products,in both economic and environmental *** cross-section data were obtained from the TALYS Evaluated Nuclear Data Library(TENDL).A thick target model was used to study the consumption of the target isotopes in the transmutation *** transmutation yield was calculated using the highest beam intensity available with the China initiative accelerator-driven *** was found that the light nuclide-induced spallation reaction can significantly reduce the radio toxicity of the investigated long-lived fission *** the transmutation target made of elemental LLFP and the proton beam with an intensity of 5 mA,the consumption of 90 Sr,93 Zr,107 Pd,or 137 Cs can reach approximately 500 g per year.

A graph is said to be claw-free if it does not contain an induced subgraph isomorphic to K_（1,3）. Let s and k be two integers with 0≤s≤k and let G be a claw-free graph of order n. In this paper, we investigate clique partition problems in claw-free graphs. It is proved that if n≥3 s ＋4（k-s） and d（x）＋ d（y）≥n-2 s ＋2 k ＋1 for any pair of non-adjacent vertices x, y of G, then G contains s disjoint K3 s and k-s disjoint K4 s such that all of them are disjoint. Moreover, the degree condition is sharp in some cases.

All-trans retinoic acid（ATRA） inhibits matrix metalloproteinase（MMP）-2 and MMP-9 in synovial fibroblasts, skin fibroblasts,bronchoalveolar lavage cells and cancer cells, but activates MMP-9 in neuroblast and leukemia cells. Very little is known regarding whether ATRA can activate or inhibit MMPs in human dental pulp cells（HDPCs）. The purpose of this study was to determine the effects of ATRA on the production and secretion of MMP-2 and-9 in HDPCs. The productions and messenger RNA（mRNA） expressions of MMP-2 and-9 were accessed by gelatin zymography and real-time polymerase chain reaction（PCR）, respectively. ATRA was found to decrease MMP-2 level in a dose-dependent manner. Significant reduction in MMP-2 mRNA expression was also observed in HDPCs treated with 25 mmol？L21ATRA. However, HDPCs treated with ATRA had no effect on the pattern of MMP-9 produced or secreted in either cell extracts or conditioned medium fractions. Taken together, ATRA had an inhibitory effect on MMP-2 expression in HDPCs,which suggests that ATRA could be a candidate as a medicament which could control the inflammation of pulp tissue in vital pulp therapy and regenerative endodontics.
