Efficient and safe cell engineering by transfection of nucleic acids remains one of the long-standing hurdles for fundamental biomedical research and many new therapeutic applications,such as CAR T cell-based *** has recently gained increasing attention as a more safe and versatile alternative tool over viral-or DNA transposon-based approaches for the generation of adoptive T ***,limitations associated with existing nonviral mRNA delivery approaches hamper progress on genetic engineering of these hard-to-transfect immune *** this study,we demonstrate that gold nanoparticle-mediated vapor nanobubble(VNB)photoporation is a promising upcoming physical transfection method capable of delivering mRNA in both adherent and suspension *** transfection experiments on HeLa cells showed the importance of transfection buffer and cargo concentration,while the technology was furthermore shown to be effective for mRNA delivery in Jurkat T cells with transfection efficiencies up to 45%.Importantly,compared to electroporation,which is the reference technology for nonviral transfection of T cells,a fivefold increase in the number of transfected viable Jurkat T cells was ***,our results point toward the use of VNB photoporation as a more gentle and efficient technology for intracellular mRNA delivery in adherent and suspension cells,with promising potential for the future engineering of cells in therapeutic and fundamental research applications.

We demonstrate a Ⅲ-Ⅴ-on-silicon-nitride mode-locked laser through the heterogeneous integration of a semiconductor optical amplifier on a passive silicon-nitride cavity using the technique of micro-transfer printing. In the initial phase of our study, we focus on optimizing the lasing wavelength to be centered at 1550 nm. This optimization is achieved by conducting experiments with 27 mode-locked lasers, each incorporating optical amplifiers featuring distinct multiple-quantum-well photoluminescence values. Subsequently we present a comprehensive study investigating the behavior of the mode-locking regime when the electrical driving parameters are varied. Specifically, we explore the impact of the gain voltage and saturable absorber current on the locking stability of a tunable mode-locked laser. By manipulating these parameters, we demonstrate the precise control of the optical spectrum across a wide range of wavelengths spanning from 1530 to 1580 nm. Furthermore, we implement an optimization approach based on a Monte Carlo analysis aimed at enhancing the mode overlap within the gain region. This adjustment enables the achievement of a laser emitting a 23-nm-wide spectrum while maintaining a defined 10 dB bandwidth for a pulse repetition rate of 3 GHz.

Background:The AbSeS-classification defines specific phenotypes of patients with intra-abdominal infection based on the(1)setting of infection onset(community-acquired,early onset,or late-onset hospital-acquired),(2)presence or absence of either localized or diffuse peritonitis,and(3)severity of disease expression(infection,sepsis,or septic shock).This classification system demonstrated reliable risk stratification in intensive care unit(ICU)patients with intra-abdominal *** study aimed to describe the epidemiology of ICU patients with pancreatic infection and assess the relationship between the components of the AbSeS-classification and ***:This was a secondary analysis of an international observational study(“AbSeS”)investigating ICU patients with intra-abdominal *** patients with pancreatic infection were included in this analysis(n=165).Mortality was defined as ICU mortality within 28 days of observation for patients discharged earlier from the *** with mortality were assessed using logistic regression analysis and reported as odds ratio(OR)and 95%confidence interval(CI).Results:The overall mortality was 35.2%(n=58).The independent risk factors for mortality included older age(OR=1.03,95%CI:1.0 to 1.1 P=0.023),localized peritonitis(OR=4.4,95%CI:1.4 to 13.9 P=0.011),and persistent signs of inflammation at day 7(OR=9.5,95%CI:3.8 to 23.9,P<0.001)or after the implementation of additional source control interventions within the first week(OR=4.0,95%CI:1.3 to 12.2,P=0.013).Gramnegative bacteria were most frequently isolated(n=58,49.2%)without clinically relevant differences in microbial etiology between survivors and ***:In pancreatic infection,a challenging source/damage control and ongoing pancreatic inflammation appear to be the strongest contributors to an unfavorable short-term *** this limited series,essentials of the AbSeS-classification,such as the setting of infection onset,diffuse

Exhuming the Meso-Cenozoic Kyrgyz Tianshan and Siberian Altai-Sayan:A review based on low-temperature thermochronology

Thermochronological datasets for the Kyrgyz Tianshan and Siberian Altai-Sayan within Central Asia reveal a punctuated exhumation history during the Meso-Cenozoic. In this paper, the datasets for both regions are collectively reviewed in order to speculate on the links between the Meso-Cenozoic exhumation of the continental Eurasian interior and the prevailing tectonic processes at the plate margins. Whereas most of the thermochronological data across both regions document late Jurassic -Cretaceous regional basement cooling, older landscape relics and dissecting fault zones throughout both regions preserve Triassic and Cenozoic events of rapid cooling, respectively. Triassic cooling is thought to reflect the Qiangtang-Eurasia collision and/or rifting/subsidence in the West Siberian basin. Alternatively, this cooling signal could be related with the terminal terrane-amalgamation of the Central Asian Orogenic Belt. For the Kygyz Tianshan, late Jurassic-Cretaceous regional exhumation and Cenozoic fault reactivations can be linked with specific tectonic events during the closure of the Palaeo-Tethys and Neo-Tethys Oceans, respectively. The effect of the progressive consumption of these oceans and the associated collisions of Cimmeria and India with Eurasia probably only had a minor effect on the exhumation of the Siberian Altai-Sayan. More likely, tectonic forces from the east （present-day co- ordinates） as a result of the building and collapse of the Mongol-Okhotsk orogen and rifting in the Baikal region shaped the current Siberian Altai-Sayan topography. Although many of these hypothesised links need to be tested further, they allow a first-order insight into the dynamic response and the stress propagation pathways from the Eurasian margin into the continental interior.

Analogies between density Functional Theory Response Kernels and derivatives of Thermodynamic State Functions

In view of its use as reactivity theory,Conceptual density Functional Theory(DFT),introduced by Parr et al.,has mainly concentrated up to now on the E = E[N,v] ***,different ensemble representations can be used involving other variables also,such as ρ and μ.In this study,these different ensemble representations(E,?,F,and R) are briefly *** attention is then given to the corresponding second-order(functional) derivatives,and their analogieswith the second-order derivatives of thermodynamic state functions U,F,H,and G,which are related to each other via Legendre transformations,just as the DFT functionals(Nalewajski and Parr,1982).Starting from an analysis of the convexity/concavity of the DFT functionals,for which explicit proofs are discussed for some cases,the positive/negative definiteness of the associated kernels is derived and a detailed comparison is made with the thermodynamic *** stability conditions in thermodynamics are similar in structure to the convexity/concavity conditions for the DFT ***,the DFT functionals are scrutinized based on the convexity/concavity of their two variables,to yield the possibility of establishing a relationship between the three second-order reactivity descriptors derived from the considered *** two ensemble representations,F and ?,F is eliminated as it has two dependent(extensive)variables,N and ρ.For ?,on the other hand,which is concave for both of its intensive variables(μ and υ),an inequality is derived from its three second-order(functional) derivatives:the global softness,the local softness,and the softness *** with the negative value of the diagonal element of the linear response function,this inequality is shown to be compatible with the Berkowitz-Parr relationship,which relates the functional derivatives of ρ with υ,at constant N and μ.This was recently at stake upon quantifying Kohn's Nearsightedness of Electronic *** analogy

Cenozoic low temperature cooling history of the eastern Lhasa terrane:Implications for high-relief topography of external drainage area in the southern Tibetan Plateau
