Uncertainty analysis of hydrological modeling in a tropicalarea using different algorithms

Hydrological modeling outputs are subject to uncertainty resulting from different sources of errors (e.g., error in input data, model structure, and model parameters), making quantification of uncertainty in hydrological modeling imperative and meant to improve reliability of modeling results. The uncertainty analysis must solve difficulties in calibration of hydrological models, which further increase in areas with data scarcity. The purpose of this study is to apply four uncertainty analysis algorithms to a semi-distributed hydrological model, quantifying different source of uncertainties (especially parameter uncertainty) and evaluate their performance. In this study, the Soil and Water Assessment Tools (SWAT) eco-hydrological model was implemented for the watershed in the center of Vietnam. The sensitivity of parameters was analyzed, and the model was calibrated. The uncertainty analysis for the hydrological model was conducted based on four algorithms: Generalized Likelihood Uncertainty Estimation (GLUE), Sequential Uncertainty fitting (SUfI), Parameter Solution method (ParaSol) and Particle Swarm Optimization (PSO). The performance of the algorithms was compared using P-factor and Rfactor, coefficient of determination (R (2)), the Nash Sutcliffe coefficient of efficiency (NSE) and Percent bias (PbIAS). The results showed the high performance of SUfI and PSO with P-factor > 0.83, R-factor 0.91, NSE > 0.89, and 0.18 bIAS fI-2 initially to set the parameter ranges, and further use PSO for final analysis. Indeed, the uncertainty analysis must be accounted when the outcomes of the model use for policy or management decisions.

Evolving ethical issues in third party reproduction:Local and global considerations

There continues to be an increase in utilization of assisted reproductive technology(ART), including the use of third party gametes. Specifically, the use of third party oocytes, most recently reported in 2010 by the United States(US) Center for Disease Control and Society of Reproductive Medicine, accounted for 15 504 cycles and 7334 live births. This translates into approximately 11% of all the in vitro fertilization cases performed in the US. As utilization increases and the technological tools advance, they have created underappreciated and unforeseen ethical quandaries. As such, many practitioners think they "have heard it all". However, each ART scenario is novel with the potential to pose complex unforeseen issues, potentially creating global challenges that could impact broad social and legal questions and test the moral consciousness' of practitioners, policymakers and patients. While there are published US national guidelines to assist practitioners, we have identified new complex issues in assisted reproduction that present unique challenges, and we give a perspective from oureyes in the Western Hemisphere looking out to a global level. Specifically, this review focuses on some of the more recent and evolving issues that currently are and will be confronting us in the upcoming years. Particular attention focuses on discrepancies between third party legal contracts and ART consents regarding level of information sharing, and oocyte and embryo directives and management; dilemmas and obligations surrounding disclosure of medical outcomes especially in the context of growing access to Direct to Consumer genetic testing and Reproductive Tourism-Exile. Given the complexity of these and other ethical questions, finding answers may be achieved by ending the isolation of reproductive professionals and instead promoting increased and consistent communication among physicians, embryologists, therapists and reproductive attorneys to confront these evolving ethical qu

AIM： To ascertain whether constitutive androstane receptor （CAR） activation by 1,4-bis-[2-（3,5,- dichloropyridyloxy）] benzene （TCPObOP） modulates steatohepatitis in the methionine choline-deficient （MCD） diet-fed ***： C57/bL6 wild-type mice were fed the MCD or standard diet for 2 wk and were treated with either the CAR agonist, TCPObOP, or the CAR inverse agonist, ***： Expression of CYP2b10 and CYP3A11, known CAR target genes, increased 30-fold and 45-fold, respectively, in TCPObOP-treated mice fed the MCD diet. TCPObOP treatment reduced hepatic steatosis （44.6 ＋ 5.4% vs 30.4 ＋ 4.5%, P 〈 0.05） and serum triglyceride levels （48 ＋ 8 vs 20 ＋ 1 mg/dL, P 〈 0.05） in MCD diet- fed mice as compared with the standard diet-fed mice. This reduction in hepatic steatosis was accompanied by an increase in enzymes involved in fatty acid microsomal co-oxidation and peroxisomal p-oxidation, namely CYP4A10, LPbE, and 3-ketoacyI-CoA thiolase. The reduction in steatosis was also accompanied by a reduction in liver cell apoptosis and inflammation. In contrast, androstanol was without effect on any of the above ***： CAR activation stimulates induction of genes involved in fatty acid oxidation, and ameliorates hepatic steatosis, apoptosis and inflammation.

Decreased Door to balloon Time: better Outcome for the Patient?

Recently the American College of Cardiology and the American Heart Association instituted DTb of 90 minutes or less as a class I recommendation. Since 2006 the percentage of patient meeting this metric has substantially increased, although research has demonstrated discrepancies in whether or not this objective is associated with better patient outcome. Here, we reviewed seven studies in effort to investigate the validity of the 90 minute or less door to balloon time. Our findings suggest that patient outcome in the setting of acute myocardial infarction is multifactorial, and while the door to balloon time metric likely has played a role in better patient outcomes it is not the sole factor in improving mortality rates.

AIM To critically evaluate previous scientific evidence on fusobacterium's role in colorectal neoplasia *** Two independent investigators systematically reviewed all original scientific articles published between January,2000,and July,2017,using Pub Med,EMbASE,and MEDLINE. A total of 355 articles were screened at the abstract level. Of these,only original scientific human,animal,and in vitro studies investigating fusobacterium and its relationship with colorectal cancer(CRC) were included in the analysis. Abstracts,review articles,studies investigating other colonic diseases,and studies written in other languages than English were excluded from our analysis. Ninety articles were included after removing duplicates,resolving disagreements between the two reviewers,and applying the above *** Studies have consistently identified positive associations between fusobacterium,especially fusobacterium nucleatum(f. nucleatum),and CRC. Stronger associations were seen in CRCs proximal to the splenic flexure and Cp G island methylator phenotype(CIMP)-high CRCs. There was evidence of temporality and a biological gradient,with increased f. nucleatum DNA detection and quantity along the traditional adenoma-carcinoma sequence and in CIMP-high CRC precursors. Diet may have a differential impact on colonic f. nucleatum enrichment;evidence suggests that high fiber diet may reduce the risk of a subset of CRCs that are f. nucleatum DNA-positive. Data also suggest shorter CRC and disease-specific survival with increased amount of f. nucleatum DNA in CRC tissue. The pathophysiology of enrichment of f. nucleatum and other fusobacterium species in colonic tissue is unclear;however,the virulence factors and changes to the local colonic environment with disruption of the protective mucus layer may contribute. Next generation sequencing for profiling expression of miRNAs: technical progress and applications in drug development

miRNAs are non-coding RNAs that play a regulatory role in expression of genes and are associated with diseases. Quantitatively measuring expression levels of miRNAs can help understanding the mechanisms of human diseases and discovering new drug targets. There are three major methods that have been used to measure the expression levels of miRNAs: real-time reverse transcription PCR (qRT-PCR), microarray, and the newly introduced next-generation sequencing (NGS). NGS is not only suitable for profiling of known miRNAs that qRT-PCR and microarray can do too but also able to detect unknown miRNAs that the other two methods are incapable. Profiling of miRNAs by NGS has been progressed rapidly and is a promising field for applications in drug development. This paper will review the technical advancement of NGS for profiling miRNAs, including comparative analyses between different platforms and software packages for analyzing NGS data. Examples and future perspectives of applications of NGS profiling miRNAs in drug development will be discussed.
