miRNAs are non-coding RNAs that play a regulatory role in expression of.genes and are associated with diseases. Quantitatively measuring expression levels of.miRNAs can help understanding the mechanisms of.human diseases and discovering new drug targets. There are three major methods that have been used to measure the expression levels of.miRNAs: real-time reverse transcription PCR (qRT-PCR), microarray, and the newly introduced next-generation sequencing (NGS). NGS is not only suitable f.r prof.ling of.known miRNAs that qRT-PCR and microarray can do too but also able to detect unknown miRNAs that the other two methods are incapable. Prof.ling of.miRNAs by NGS has been progressed rapidly and is a promising f.eld f.r applications in drug development. This paper will review the technical advancement of.NGS f.r prof.ling miRNAs, including comparative analyses between dif.erent platf.rms and sof.ware packages f.r analyzing NGS data. Examples and f.ture perspectives of.applications of.NGS prof.ling miRNAs in drug development will be discussed.

Toxicity is the totality of.adverse ef.ects, which can be f.nctional and morphological lesions in a living organism, caused by a substance introduced in relatively high single doses or in small, repeated doses. The aim of.this study was to assess the OECD-recommended acute oral toxicity and anti-inf.ammatory activity of.ethanolic extract of.f.cus umbellata leaves. Animals were given a single oral dose of.1000, 3000 and 5000 mg/Kg body weight (BW) of.the extract. f.r the anti-inf.ammatory activity test, rats were given the ethanolic extract of.f. umbellata leaves at doses of.100, 300 and 500 mg/Kg or aspirin® at a concentration of.100 mg/Kg PC orally, one hour bef.re injection of.0.05 ml of.1% f.rmalin under the plantar f.scia of.the rat’s right hind paw. Paw volume measurements were taken one, two and three hours af.er f.rmalin injection, using an electronic caliper. Af.er 14 days of.observation, no deaths were observed in treated rats. The LD50 of.ethanolic extract of.f.cus umbellata leaf.powder is greater than 5000 mg/Kg body weight. This extract has no signif.cant ef.ects on hematological parameters and on the main markers of.nephrotoxicity and hepatotoxicity f.r a single dose of.less than 5000 mg/Kg PC. It reduces f.rmalin-induced edema. Evaluation of.the percentage inhibition showed that the extract had greater anti-inf.ammatory activity at 3 hours af.er the start of.the experiment. However, better inhibition of.inf.ammatory oedema of.the paw of.rats treated with 500 mg/Kg was observed at 5 hours af.er the start of.the experiment, with a percentage inhibition of.69.23 ± 1.02, compared with the ref.rence group treated with aspirin® 100 mg/Kg, which showed an inhibition of.63.50 ± 0.98. These results show that f. umbellata leaves possess anti-inf.ammatory activity, which would justif. their use in traditional Af.ican medicine to prevent or treat inf.ammation.

Landslides triggered by rainf.ll can possibly be f.reseen in real time by jointly using rainf.ll intensity-duration thresholds and inf.rmation related to land surf.ce susceptibility. However, no system exists at either a national or a global scale to monitor or detect rainf.ll conditions that may trigger landslides due to the lack of.suf.icient ground-based observing network in many parts of.the world. Recent advances in satellite remote sensing technology and increasing availability of.high-resolution geospatial products around the globe have provided an unprecedented opportunity f.r such a study. In this paper, a f.amework f.r developing a preliminary real-time prediction system to identif. where rainf.ll-triggered landslides will occur is proposed by combining two necessary components: surf.ce landslide susceptibility and a real-time space-based rainf.ll analysis system (http://***). f.rst, a global landslide susceptibility map is derived f.om a combination of.semi-static global surf.ce characteristics (digital elevation topography, slope, soil types, soil texture, land cover classif.cation, etc.) using a GIS weighted linear combination approach. Second, an adjusted empirical relationship between rainf.ll intensity-duration and landslide occurrence is used to assess landslide hazards at areas with high susceptibility. A major outcome of.this work is the availability f.r the f.rst time of.a global assessment of.landslide hazards, which is only possible because of.the utilization of.global satellite remote sensing products. This preliminary system can be updated continuously using the new satellite remote sensing products. This proposed system, if.pursued through wide interdisciplinary ef.orts as recommended herein, bears the promise to grow many local landslide hazard analyses into a global decision-making support system f.r landslide disaster preparedness and mitigation activities across the world.
