Long-term antifibrotic action of interferon-γ treatment in patients with chronic hepatitis B virus infection

BAcKGROUND:The first priority in treating fibrosis is to eliminate the causes that result in liver injury,e.g.,hepatitis B and c ***,in many liver diseases the cause is either unknown or *** present study was designed to investigate the long-term antifibrotic effect of interferon-gamma(IFN-γ)treatment in patients chronically infected with hepatitis B virus. METHODS:A total of 42 patients,30 treated with IFN-γand 12 controls,were enrolled from an original clinical trial(clin Gastroenterol Hepatol 2005;3:819.).Three serial liver biopsies that were obtained at the initiation and end of IFN-γtreatment as well as 4 to 6 years after treatment discontinuation were assessed according to the modified chevallier scoring system. RESULTS:Twenty-five out of 30 IFN-γ-treated patients were followed up until 4 to 6 years after the treatment was stopped. However,all controls were excluded from follow-up due to death,loss and elevated virus level within 2 ***-five IFN-γ-treated patients had stable serum liver function and liver fibrosis indices without any further antiviral or anti-fibrotic *** inflammatory and fibrotic scores were found after nine months of IFN-γtreatment according to the modified chevallier scoring system(inflammation:11.8±6.5 at the beginning of IFN-γtreatment vs.9.2±4.1 after 9 months, Pccepted a third serial liver biopsy 4 to 6 years after treatment discontinuation,and the fibrotic score was increased(14.2±8.3 vs.11.9±7.6 after 9 months, PcONcLUSIONS:Nine-month IFN-γtreatment significantly improves the fibrosis score in patients with chronic HBV *** majority of patients demonstrate stable serum biochemical indices and quality of ***,they do not show a long-term benefit according to histological criteria. The interleukin-17 (IL-17) family of cytokines and receptors play central roles in host defense against infection, and development of inflammatory diseases1. The compositions and structures of functional IL-17 family ligand-receptor signaling assemblies remain unclear. Interleukin-17E (IL-17E or IL-25) is a key regulator of Th2 immune responses and driver of inflammatory diseases such as allergic asthma and requires both IL-17 receptor A (IL-17RA) and IL-17RB to elicit functional responses2. Here, we studied IL-25-IL-17RB binary and IL-25-IL-17RB-IL-17RA ternary complexes using a combination of cryo-electron microscopy (cryo-EM), single-molecule imaging, and cell-based signaling approaches. The IL-25-IL-17RB-IL-17RA ternary signaling assembly is a c2-symmetric complex in which the IL-25-IL-17RB homodimer is flanked by two "wing-like" IL-17RA co-receptors through a "tip-to-tip" geometry that is the key receptor-receptor interaction required for initiation of signal transduction.

A Field Observation and Significant Range Extension of Manouria impressa in Myanmar

Few records of the impressed tortoise （Manouria impressa） are available from Myanmar and most are based on historic accounts, old museum specimens, and shells obtained from villagers and wildlife traffickers. We herein report the observation of a living M. impressa in the Hukaung Valley Wildlife Sanctuary of northern Myanmar. Our observation extends the known distribution of M. impressa in Myanmar approximately 850 km northwards from previously reported populations in the Rakhine Hills of Magway Region.

The VEGF signaling pathway in cancer: the road ahead

The vascular endothelial growth factor (VEGF) family of soluble protein growth factors consists of key mediators of angiogenesis and lymphangiogenesis in the context of tumor biology. The members of the family, VEGF-A (also known as VEGF), VEGF-B, VEGF-c, VEGF-D, and placenta growth factor (PlGF), play important roles in vascular biology in both normal physiology and pathology. The generation of a humanized neutralizing antibody to VEGF-A (bevacizumab, also known as Avastin) and the demonstration of its benefit in numerous human cancers have confirmed the merit of an anti-angiogenesis approach to cancer treatment and have validated the VEGF-A signaling pathway as a therapeutic target. Other members of the VEGF family are now being targeted, and their relevance to human cancer and the development of resistance to anti-VEGF-A treatment are being evaluated in the clinic. Here, we discuss the potential of targeting VEGF family members in the diagnosis and treatment of cancer.

High-energy sodium-ion hybrid capacitors through nanograin-boundary-induced pseudocapacitance of co_(3)O_(4) nanorods

Sodium-ion hybrid capacitors (SIcs) have been proposed to bridge performance gaps between batteries and supercapacitors,and thus realize both high energy density and power density in a single ***,applications of SIcs are severely restricted by their insufficient energy densities (cs imbalance between cathodes and ***,we report a nanograin-boundary-rich hierarchical co_(3)O_(4) nanorod anode composed of~20 nm *** pseudocapacitance (up to 72%@1.0 mV/s) is achieved through nanograin-boundary-induced pseudocapacitive-type Na^(+) storage ***_(3)O_(4) nanorod anode delivers in this case highly reversible capacity (810 mAh/g@0.025 A/g),excellent rate capability (335 mAh/g@5.0 A/g),and improved cycle stability (100 cycles@1.0 A/g with negligible capacity degradation).The outstanding performance can be credited to the hierarchical morphology of co_(3)O_(4) nanorods and the well-designed nanograinboundaries between nanocrystallites that avoid particle agglomeration,induce pseudocapacitive-type Na^(+) storage,and accommodate volume variation during sodiation-desodiation *** of the co_(3)O_(4) nanorods not only generates defects for extra surficial Na^(+) storage but also increases the electronic conductivity for efficient charge separation and lowers energy barrier for Na^(+) *** of conventional reaction mechanism and pseudocapacitive-type Na^(+) storage enables high specific capacity,rapid Na^(+) diffusion,and improved structural stability of the co_(3)O_(4) nanorod *** SIc integrating this highly pseudocapacitive anode and activated carbon cathode delivers exceptional energy density (175 Wh/kg@40 W/kg),power density (6632 W/kg@37 Wh/kg),cycle life (6000 cycles@1.0 A/g with a capacity retention of 81%),and coulombic efficiency (~100%).

A simple umbelliferone based fluorescent probe for the detection of nitroreductase

A simple nitrobenzyl-umbelliferone （NcOU1） was synthesised containing a nitroreductase （NTR） trigger moiety. The presence of NTR, resulted in the fragmentation of the parent molecule and release of the highly emissive fluorophore umbelliferone via an NTR-catalyzed reduction of the nitro group. In the presence of the NTR enzyme, NcOU1 gave rise to a 5-fold increase in fluorescence intensity at 455 nm and was selective for NTR over other reductive enzymes. These results indicate that NcOU1 can be used as a simple assay for the detection of NTR.

Osteoclasts, the bone-resorbing cells, play a pivotal role in skeletal development and adult bone remodeling. They also participate in the pathogenesis of various bone disorders. Osteoclasts differentiate from cells of the monocyte/macrophage lineage upon stimulation of two essential factors, the monocyte/ macrophage colony stimulating factor （M-cSF） and receptor activation of NF-κB ligand （RANKL）. M-cSF binds to its receptor c-Fms to activate distinct signaling pathways to stimulate the proliferation and survival of osteoclast precursors and the mature cell. RANKL, however, is the primary osteoclast differentiation factor, and promotes osteoclast differentiation mainly through controlling gene expression by activating its receptor, RANK. Osteoclast function depends on polarization of the cell, induced by integrin avβ3, to form the resorptive machinery characterized by the attachment to the bone matrix and the formation of the bone-apposed ruffled border. Recent studies have provided new insights into the mechanism of osteoclast differentiation and bone resorption. In particular, c-Fms and RANK signaling have been shown to regulate bone resorption by cross-talking with those activated by integrin avβ3. This review discusses new advances in the understanding of the mechanisms of osteoclast differentiation and function.

Prophylactic fixation of the unaffected contralateral side in children with slipped capital femoral epiphysis seems favorable:A systematic review

Volterra filter modeling of a nonlinear discrete-time system based on a ranked differential evolution algorithm

This paper presents a ranked differential evolution(RDE) algorithm for solving the identification problem of nonlinear discrete-time systems based on a Volterra filter model. In the improved method, a scale factor, generated by combining a sine function and randomness, effectively keeps a balance between the global search and the local search. Also, the mutation operation is modified after ranking all candidate solutions of the population to help avoid the occurrence of premature convergence. Finally, two examples including a highly nonlinear discrete-time rational system and a real heat exchanger are used to evaluate the performance of the RDE algorithm and five other approaches. Numerical experiments and comparisons demonstrate that the RDE algorithm performs better than the other approaches in most cases.
