Droplet digital microfluidic system for screening filamentous fungi based on enzymatic activity

Fungal cell-wall-degrading enzymes have great utility in the agricultural and food *** cell-wall-degrading enzymes are known to have functions that can help defend against pathogenic *** existing methods used to discover these enzymes are not well adapted to fungi culture and morphology,which prevents the proper evaluation of these *** report the first droplet-based microfluidic method capable of long-term incubation and low-voltage conditions to sort filamentous fungi inside nanoliter-sized *** new method was characterized and validated in solid-phase media based on colloidal chitin such that the incubation of single spores in droplets was possible over multiple days(2–4 days)and could be sorted without droplet *** long-term culture,we examined the activity of cell-wall-degrading enzymes produced by fungi during solid-state droplet fermentation using three highly sensitive fluorescein-based *** also used the low-voltage droplet sorter to select clones with highly active cell-wall-degrading enzymes,such as chitinases,β-glucanases,and β-Nacetylgalactosaminidases,from a filamentous fungi droplet library that had been incubated for>4 *** new system is portable,affordable for any laboratory,and user-friendly compared to classical droplet-based microfluidic *** propose that this system will be useful for the growing number of scientists interested in fungal microbiology who are seeking high-throughput methods to incubate and sort a large library of fungal cells.

How can Practitioners Support citizen Volunteers in Disaster Risk Reduction?Insight from“Good and Ready”in Aotearoa New Zealand

Global and national policy frameworks emphasize the importance of people's participation and volunteers'role in disaster risk *** research has extensively focused on volunteers in disaster response and recovery,less attention has been paid on how organizations involved in disaster risk management can support volunteers in leading and coordinating communitybased disaster risk *** 2019,the New Zealand Red cross piloted the Good and Ready initiative in Auckland,Aotearoa New Zealand,with the objective to empower local people in resilience building with a focus on volunteers and community *** research examined the positive and negative outcomes of Good and Ready and investigated volunteers'experiences in the disaster resilience *** involved the codesign of a questionnaire-based survey using participatory methods with Good and Ready volunteers,the dissemination of the survey to gather volunteers'viewpoints,and a focus group discussion with participatory activities with Red cross *** findings highlight that a key challenge lies in finding a balance between a program that provides flexibility to address contextual issues and fosters communities'ownership,versus a prescriptive and standardized approach that leaves little room for creativity and *** pinpoints that supporting volunteers with technical training is critical but that soft skills training such as coordinating,communicating,or facilitating activities at the local level are *** concludes that the sustainability of Good and Ready requires understanding and meeting volunteers'motivations and expectations and that enhancing partnerships with local emergency management agencies would strengthen the program.

Measuring the Student Group capacity for Obtaining Geometric Information in the van Hiele Developmental Thought Process:A Fuzzy Approach

The student group possibilistic uncertainty in the van Hiele process of geometric reasoning is considered as the student group capacity for obtaining geometric information. Hence, the student group capacity for obtaining geometric information can be measured by the total possibilistic uncertainty T(r) of an ordered possibility distribution. The capacities of three student groups are found by calculating the total possibilistic uncertainties of these groups. These capacity values set an expectation of future group performance. Future research indications of how the total possibilistic uncertainty may be applied to other models of learning and development will help to establish it as a viable measure in educational psychology.

Development of a new humane toxin for predator control in New Zealand

The endemic fauna of New Zealand evolved in the absence of mammalian predators and their introduction has been responsible for many extinctions and *** species including possums(Trichosurus vulpecula Kerr),ship rats(Rattus rattus L.)and stoats(Mustela erminea L.)are targeted to protect native *** methodologies currently rely largely on labor-intensive trapping or the use of increasingly unpopular poisons,or poisons that are linked with low welfare ***,the development of safer humane predator toxins and delivery systems is highly ***-aminopropiophenone(PAPP)is being developed as a toxin for feral cats(Felis catus L.)and *** appear to be much more susceptible to PAPP than birds,so it potentially has high target specificity,at least in New *** trials with 20 feral cats and 15 stoats have been undertaken using meat baits containing a proprietary formulation of PAPP.A PAPP dose of 20-34 mg kg^(-1)was lethal for feral cats and 37-95 mg kg^(-1)was lethal for *** assessments suggest that PAPP,for the control of feral cats and stoats,is a humane and effective *** causes methaemoglobinaemia,resulting in central nervous system anoxia,lethargy and death.

Diversity in global gene expression and morphology across a watercress (Nasturtium officinale R. Br.) germplasm collection: first steps to breeding

Watercress(Nasturtium officinale ***.)is a nutrient intense,leafy crop that is consumed raw or in soups across the globe,but for which,currently no genomic resources or breeding programme *** morphological,biochemical and functional genomic variation was identified for the first time in a newly established watercress germplasm collection,consisting of 48 watercress accessions sourced from contrasting global *** length,stem diameter and anti-oxidant(AO)potential varied across the *** variation was used to identify three extreme contrasting accessions for further *** in global gene expression was investigated using an Affymetrix Arabidopsis ATH1 microarray gene chip,using the commercial control(c),an accession selected for dwarf phenotype with a high AO potential(dwarfAO,called‘Boldrewood’)and one with high AO potential alone.A set of transcripts significantly differentially expressed between these three accessions,were identified,including transcripts involved in the regulation of growth and development and those involved in secondary *** particular,when differential gene expression was compared between c and dwarfAO,the dwarfAO was characterised by increased expression of genes encoding glucosinolates,which are known precursors of phenethyl isothiocyanate,linked to the anti-carcinogenic effects well-documented in *** study provides the first analysis of natural variation across the watercress genome and has identified important underpinning information for future breeding for enhanced anti-carcinogenic properties and morphology traits in this nutrient-intense crop.

Fabrication of all-transparent polymer-based and encapsulated nanofluidic devices using nano-indentation lithography

In this paper,we describe a novel and simple process for the fabrication of all-transparent and encapsulated polymeric nanofluidic devices using nano-indentation ***,a nanomechanical probe is used to‘scratch’nanoscale channels on polymethylmethacrylate(PMMA)substrates with sufficiently high ***,polydimethylsiloxane(PDMS)is used twice to duplicate the nanochannels onto PDMS substrates from the‘nano-scratched’PMMA substrates.A number of experiments are conducted to explore the relationships between the nano-indentation parameters and the nanochannel dimensions and to control the aspect ratio of the fabricated *** addition,traditional photolithography combined with soft lithography is employed to fabricate microchannels on another PDMS‘cap’*** manually aligning the substrates,all uncovered channels on two separate PDMS substrates are bonded to achieve a sealed and transparent nanofluidic device,which makes the dimensional transition from microscale to nanoscale *** smallest dimensions of the achievable nanochannels that we have demonstrated thus far are of~20 nm depth and~800 nm width,with lengths extendable beyond 100μ*** flow experiments are performed to verify the reliability of the *** types of colloidal solution are used to visualize the fluid flow through the nanochannels,that is,ethanol is mixed with gold colloid or fluorescent dye(fluorescein isothiocyanate),and the flow rate and filling time of liquid in the nanochannels are estimated based on time-lapsed image *** simplicity of the fabrication process,bio-compatibility of the polymer substrates,and optical transparency of the nanochannels for flow visualization are key characteristics of this approach that will be very useful for nanofluidic and biomolecular research applications in the future.

The impact of alloantibodies directed against the second donor on long-term outcomes of repeat liver transplantation

Background:Despite reports that associate donor specific antibody(DSA)with rejection after liver transplantation,grafts are still allocated according to blood group(ABO)but not human leukocyte antigen(HLA)compatibility,possibly due to the absence of an easily discernible clinical association between adverse recipient outcome and ***-transplantation provides a test environment where the presence of preformed DSA is prevalent and its effect on outcome should be ***:All patients undergoing a second liver transplantation with available pre-operative serum were included with the exception of ABO incompatible or multiple organ *** sera were tested for anti-HLA antibodies with Luminex-based solid phase ***-HLA antibodies to the second donor(D2SA)were determined using antibodies specificity and HLA typing of 2nd liver ***:Preformed HLA antibodies directed to second liver transplantation(D2SA)were found in 31(39%)of the 79 patients that were included in the *** and re-transplantation characteristics were similar in both subgroups except first graft survival which was significantly shorter in recipients who are negative for *** survival of the second graft was similar in D2SA+and D2SA?cohorts[8.55(range,0.01–24.74)vs.7.56(range,0–23.53)years respectively,P=0.574].Mean patient survival after 2nd liver transplantation was similar in D2SA+and D2SA?cohorts[9.11(range,0.01–24.74)vs.8.10(range,0–23.53)years respectively,P=0.504].Subgroup univariate analysis demonstrated no detrimental effect of class,locus,or strength of D2SA on survival of the second liver *** multivariate cox regression model,neither class I D2DSA(HR=1.101,P=0.92)nor class II D2SA(HR=1.74,P=0.359)were significant risks of graft ***:Presence of D2SA was not found to be associated with inferior outcomes in this retrospective cohort study of liver re-transplantation suggesting that changes to the allocation system are not requ
