Formation of the periodontium begins following onset of tooth-root formation in a coordinated manner after birth. Dental follicle progenitor cells are thought to form the cementum, alveolar bone and Sharpey＇s fibers of the periodontal ligament （PDL）. However, little is known about the regulatory morphogens that control differentiation and function of these progenitor cells, as well as the progenitor cells involved in crown and root formation. We investigated the role of bone morphogenetic protein-2 （Bmp2） in these processes by the conditional removal of the Bmp2 gene using the Sp7-Cre-EGFP mouse model. Sp7-Cre-EGFP first becomes active at E18 in the first molar, with robust Cre activity at postnatal day 0 （PO）, followed by Cre activity in the second molar, which occurs after P0. There is robust Cre activity in the periodontium and third molars by 2 weeks of age. When the Bmp2gene is removed from Sp7＋（Osterix＋） cells, major defects are noted in root, cellular cementum and periodontium formation. First, there are major cell autonomous defects in root-odontoblast terminal differentiation. Second, there are major alterations in formation of the PDLs and cellular cementum, correlated with decreased nuclear factor IC （Nfic）, periostin and α-SMA＋ cells. Third, there is a failure to produce vascular endothelial growth factor A （VEGF-A） in the periodontium and the pulp leading to decreased formation of the microvascular and associated candidate stem cells in the Bmp2-cKOsp7-cre＇EGFe. Fourth, ameloblast function and enamel formation are indirectly altered in the Bmp2-cKOsp7-cre＇EGFe. These data demonstrate that the Bmp2 gene has complex roles in postnatal tooth development and periodontium formation.

The superconvergence in the finite element method is a phenomenon in which the fi-nite element approximation converges to the exact solution at a rate higher than the optimal order error estimate. Wang proposed and analyzed superconvergence of the conforming finite element method by L2-projections. However, since the conforming finite element method (CFEM) requires a strong continuity, it is not easy to construct such finite elements for the complex partial differential equations. Thus, the nonconforming finite element method (NCFEM) is more appealing computationally due to better stability and flexibility properties compared to CFEM. The objective of this paper is to establish a general superconvergence result for the nonconforming finite element approximations for second-order elliptic problems by L2-projection methods by applying the idea presented in Wang. MATLAB codes are published at https://***/annaleeharris/Superconvergence-NCFEM for anyone to use and to study. The results of numerical experiments show great promise for the robustness, reliability, flexibility and accuracy of superconvergence in NCFEM by L2- projections.

Transgenic mice are widely used to delete or overexpress genes in a cell specific manner to advance knowledge of bone biology,function and *** numerous Cre models exist to target gene recombination in osteoblasts and osteoclasts,few target osteocytes specifically,particularly mature *** goal was to create a spatial and temporal conditional Cre model using tamoxifen to induce Cre activity in mature osteocytes using a Bac construct containing the 5’and 3’regions of the Sost gene(Sost ERT2 Cre).Four founder lines were crossed with the Ai9 Cre reporter *** founder line showed high and specific activity in mature *** and organs were imaged and fluorescent signal *** no activity was observed in 2 day old pups,by 2 months of age some osteocytes were positive as osteocyte Cre activity became spontaneous or‘leaky’with *** percentage of positive osteocytes increased following tamoxifen injection,especially in males,with 43%to 95%positive cells compared to 19%to 32%in *** signal was observed in any bone surface cell,bone marrow,nor in muscle with or without tamoxifen *** spontaneous signal was observed in any other ***,with tamoxifen injection,a few positive cells were observed in kidney,eye,lung,heart and *** other organs,28 in total,were negative with tamoxifen ***,with age,a muscle phenotype was apparent in the Sost-ERT2 Cre ***,although this mouse model may be useful for targeting gene deletion or expression to mature osteocytes,the muscle phenotype may restrict the use of this model to specific applications and should be considered when interpreting data.

Lithium(Li)penetration through solid electrolytes(SEs)induces short circuits in Li solid-state batteries(SSBs),which is a critical issue that hinders the development of high energy density *** cracking in ceramic SEs has been often shown to accompany Li penetration,the interplay between Li deposition and cracking remains ***,we constructed a mesoscale SSB inside a focused ion beam-scanning electron microscope(FIB-SEM)for in situ observation of Li deposition-induced cracking in SEs at nanometer *** results revealed that Li propagated predominantly along transgranular cracks in a garnet Li_(6.4)La_(3)Zr_(1.4)Ta_(0.6)O_(12)(LLZTO).Cracks appeared to initiate from the interior of LLZTO beneath the electrode surface and then propagated by curving toward the LLZTO *** resulting bowl-shaped cracks resemble those from hydraulic fracture caused by high fluid pressure on the surface of internal cracks,suggesting that the Li deposition-induced pressure is the major driving force of crack initiation and *** high pressure generated by Li deposition is further supported by in situ observation of the flow of filled Li between the crack flanks,causing crack widening and *** work unveils the dynamic interplay between Li deposition and cracking in SEs and provides insight into the mitigation of Li dendrite penetration in SSBs.

In order to improve the processibility in thermoplastic polyimides, a new method, termed the'reactive plasticizer' approach, has been proposed. This method uses a small amount (5～15 mol%) of a lessactivated, weak nucleophilic diamine co-monomer as a 'reactive plasticizer' to obtain copolyimide resinswhich possess relatively low viscosity at low temperatures and can be readily processed through the autoclavecycle at low pressures. During a high temperature treatment, the reactive plasticizers join the reaction to formhigh molecular weight copolyimides, and the preferred material properties are thus achieved. The mosteffective reactive plasticizer is aromatic heterocyclic diamines, such as 2, 6-diaminopyridine diamine (DAP),and the transimidization involved with a reactive plasticizer has been proposed to play a major role for thesuccess of this approach. In order to understand the transimidization mechanism, three steps have been takenin this research: first, a copolyimide system of 50% of DAP and 50% 1, 4 -bis [4-aminophenoxy] benzenediamine (DODA) with 100% of 2, 2'-bis[4-(3, 4-dicarboxyphenoxy)phenyl] propane dianhydride (Ultem^(R)DA) is prepared. Second, several specifically designed polyimide mixture systems were used, and they consistof two homopolyimides: one is Ultem^(R) DA-DODA, and the other is Ultem^(R) DA-DAP. The third step is toinvestigate two mixture systems in which Ultem^(R) DA-DODA is mixed with DAP monomer solution andUltem^(R) DA-DAP is mixed with DODA monomer solution. For all systems, with increasing degree oftransimidization upon heat-treatment, the chain structures of the mixtures and their thermal and dynamicmechanical transition behaviors are investigated via one-dimensional and two-dimensional nuclear magneticresonance, differential scanning calorimetry, and dynamic mechanical analysis experiments.
