＜正＞Background:The cellular functions of DNA repair and programmed cell death are believed to play an important role in controlling the fate of tumor cells in response to chemotherapy or *** has been speculated that single nucleotide polymorphisms （SNPs） in related genes may alter their expression or activity,affect the functions of DNA repair and/or apoptosis,and in turn influence the effects of cancer treatment and *** assess this possibility,we genotyped four single nucleotide polymorphisms （SNPs） in ATM （A60G）,ERCCl （Asn118Asn）,APEl （Asn148Glu）,XRCCl（Arg194Trp）,XRCCl （Arg399Gln） and iASPP （A67T）,and examined their associations with treatment response and overall survival among patients with advanced non-small cell lung cancer （NSCLC）. Methods:Included in the study were 230 patients diagnosed with inoperable advanced *** these patients,76 received platinum-based chemotherapy,125 received chemotherapy plus radiation,and 29 received radiotherapy *** SNPs were genotyped using the TaqMan methods. Associations between clinical variables,treatment response and genotype were analyzed using the Chi-square *** unconditional logistic regression model was used to analyze the association between genotype and treatment response while adjusting for *** analysis was performed for overall survival using Kaplan-Meier survival curves and Cox proportional hazards regression. Results:No significant correlation between genotype and overall survival was found,but ERCCl （Asn118Asn） and iASPP （A67T） genotype was significantly associated with treatment response separately among the patients who received chemotherapy only and the patients who were treated with both chemotherapy and *** with either one or two T alleles （T/T+C/T） at Asn118Asn of ERCCl were more likely not to respond to platinum-based chemotherapy compared to those without the T allele （OR=4.10,95% CI:1.31-12.85）.Patients carrying an A allele （A/T+A/A） at A67T o

Colorectal cancer remains the third most common cancer in both women and men *** modifiable dietary factors is crucial in developing primary prevention *** B6 is involved in more than 100 coenzyme reactions,and may influence colorectal cancer risk in multiple ways including through its role in one-carbon metabolism related DNA synthesis and methylation and by reducing inflammation,cell proliferation,and oxidative *** studies of dietary or dietary plus supplementary intake of vitamin B6 and colorectal cancer risk have been inconsistent with most studies reporting nonsignificant positive or inverse ***,published studies of plasma pyridoxal 5'-phosphate(the active form of vitamin B6) levels consistently support an approximately 30%-50% reduction in risk of colorectal cancer comparing high with low *** reasons for the discrepancy in the results between dietary-based and plasma-based studies remain *** unresolved questions include the effects of vitamin B6 intake in early life(i.e.,childhood or adolescence) and of suboptimal vitamin B6 status on colorectal cancer risk,whether the associations with vitamin B6 differ across molecular subtypes of colorectal cancer,and whether the vitamin B6-colorectal cancer association is modified by genetic variants of one-carbon metabolism.

Monkeypox(mpox),is a disease from the Poxviridae family that can cause several serious medical *** mini-review sought to analyze the existing literature regarding the current mpox outbreak with a focus on the prevalence,diagnostics,and containment *** cases have been reported to World Health Organization(WHO)from 85 Member States in all six WHO regions during the period of January 1,2022,through August 3,*** or optimized guidelines for the clinical care of patients with mpox are limited,particularly in low-resource *** an effort to achieve guidance and meet standards,special attention should be paid to this outbreak in order to eradicate such a rare infectious disease by analyzing prevention and control *** outcomes may also be poor,and their illnesses may last for a long *** spectrum of clinical symptoms,including complications and sequelae,as well as aspects of the illness may be indicators of sickness severity and complications;therefore,its clinical presentation must be better understood to improve containment *** addition,it is important to create and evaluate a standard of care that takes a variety of parameters into account,including antiviral,immune therapies,and clinical metrics that are particular to *** global emergence of mpox has presented new challenges for public health and has called for further investigation into its epidemiological profile across international contexts.

Serine integrases have been shown to be efficient tools for metabolic pathway *** further improve the flexibility and efficiency of pathway engineering via serine integrases,we explored how multiple orthogonally active serine integrases can be applied for use in vitro for the heterologous expression of complex biosynthesis pathways in Streptomyces spp.,the major producers of useful bioactive natural *** results show that multiple orthogonal serine integrases efficiently assemble the genes from a complex biosynthesis pathway in a single in vitro recombination reaction,potentially permitting a versatile combinatorial assembly ***,the assembly strategy also permitted the incorporation of a well-characterised promoter upstream of each gene for expression in a heterologous *** results demonstrate how site-specific recombination based on orthogonal serine integrases can be applied in Streptomyces spp.

Multidisciplinary approach for multifocal, bilobar hepatocellular carcinoma: A case report and literature review

BACKGROUND Hepatocellular carcinoma(HCC) is the second most lethal malignancy worldwide. There has been virtually no change in the survivability of HCC in spite of improvement in therapies. Surgery is considered the ideal first, curative intervention, however most patients present in advanced stages with unresectable disease. Therefore, systemic and liver-directed non-operative therapies are initially offered to downstage the disease. To ensure optimal management, a multidisciplinary team approach is often warranted. Our case highlights the benefits of a multidisciplinary approach in a young woman with multifocal, bilobar *** SUMMARY A 36-year-old Chinese woman with untreated hepatitis B was found to have large bilobar HCC during work up for abdominal pain. Her initial serum alphafetoprotein was significantly elevated to 311136 ng/mL. Computed tomography scan demonstrated bulky bilobar liver masses, consistent with intermediate stage HCC, Barcelona Clinic Liver Cancer Stage B. Her case was discussed and a personalized care plan was developed at the Multidisciplinary Center for Advanced Minimally Invasive Liver Oncologic Therapies at the University of Washington. She initially underwent bilobar transarterial chemoembolization with partial response of the left lobar tumor. Salvage hypofractionated proton beam radiation therapy was delivered to the right lobe followed by two additional transarterial chemoembolizations to the left lobe with good ***, to remove left lobar residual disease, she was taken to the operating roomfor a left hepatectomy eleven months after her initial presentation. She continues to be without evidence of *** Coordinating the multiple HCC treatment modalities is complex and our case highlights the benefits of a multidisciplinary approach.

In this paper we review the design and development of a 100 J, 10 Hz nanosecond pulsed laser, codenamed DiPOLE100 X,being built at the Central Laser Facility(CLF). This 1 kW average power diode-pumped solid-state laser(DPSSL) is based on a master oscillator power amplifier(MOPA) design, which includes two cryogenic gas cooled amplifier stages based on DiPOLE multi-slab ceramic Yb:YAG amplifier technology developed at the CLF. The laser will produce pulses between 2 and 15 ns in duration with precise, arbitrarily selectable shapes, at pulse repetition rates up to 10 Hz, allowing real-time shape optimization for compression experiments. Once completed, the laser will be delivered to the European X-ray Free Electron Laser(XFEL) facility in Germany as a UK-funded contribution in kind, where it will be used to study extreme states of matter at the High Energy Density(HED) instrument.

Activated Carbon and Clay Minerals for the Sorptive Removal of Denatonium Ions from Denatonium Benzoate Solutions

This study assessed the feasibility of utilizing activated carbon and clay minerals for treating water impacted with the bittering agent denatonium benzoate (DB). Our specific study objectives were to 1) evaluate denatonium ion sorption to smectite clay minerals (bentonite and hectorite) and activated carbon (powdered and granular) at constant pH and ionic strength and 2) examine the impact of pH on denatonium ion sorption to each solid material. The experimental results indicated that high doses (33,000 mg/L) of as-received granular activated carbon and as-received clay minerals completely removed denatonium from aqueous solutions containing 100 - 1000 mg/L denatonium benzoate. Powdered activated carbon at doses of 5 - 100 mg/L exhibited favorable monolayer sorption of denatonium ions from a pH 6.95, 70 mg/L aqueous denatonium benzoate solution with a Langmuir separation factor (r) of 0.481, a maximum sorption capacity (Sm) of 74 mg/g, and a Langmuir constant of 15.3 L/g. A maximum removal of 23% of denatonium was achieved at the highest powdered activated carbon dosage employed. Denatonium ion removal with peroxide treated bentonite and peroxide treated hectorite did not result in complete removal of the ion and exhibited favorable sorption as evidenced by Freundlich 1/n values ranging from 0.803 to 1.194;Freundlich constants (Kf) ranged from 8 ng/L to 575 ng/L. Denatonium ion sorption to peroxide treated bentonite appeared to depend on pH while hectorite sorption of denatonium ions was independent of hydrogen ion concentrations. For powdered activated carbon adsorption, as pH increased denatonium ion sorption decreased. Overall, the work demonstrates that denatonium can be effectively removed from water via activated carbon or clay mineral sorption.
