The Immunome of Colon Cancer: Functional In Silico Analysis of Antigenic Proteins Deduced from IgG Microarray Pro?ling

Characterization of the colon cancer immunome and its autoantibody signature from differentially-reactive antigens （DIRAGs） could provide insights into aberrant cellular mechanisms or enriched networks associated with diseases. The purpose of this study was to characterize the antibody profile of plasma samples from 32 colorectal cancer （CRC） patients and 32 controls using proteins isolated from 15,417 human cDNA expression clones on microarrays. 671 unique DIRAGs were identified and 632 were more highly reactive in CRC samples. Bioinformatics analyses reveal that compared to control samples, the immunoproteomic IgG profiling of CRC samples is mainly associated with cell death, survival, and proliferation pathways, especially proteins involved in EIF2and mTOR signaling. Ribosomal proteins （e.g., RPL7, RPL22, and RPL27A） and CRC-related genes such as APC, AXIN1, E2F4, MSH2, PMS2, and TP53 were highly enriched. In addition, dif- ferential pathways were observed between the CRC and control samples. Furthermore, 103 DIR- AGs were reported in the SEREX antigen database, demonstrating our ability to identify known and new reactive antigens. We also found an overlap of 7 antigens with 48 ＂CRC genes.＂ These data indicate that immunomies profiling on protein mieroarrays is able to reveal the complexity of immune responses in cancerous diseases and faithfully reflects the underlying pathology.

Risk of recurrence of primary sclerosing cholangitis after liver transplantation is associated with de novo inflammatory bowel disease

AIM To evaluate risk factors for primary sclerosing cholangitis(PSC) recurrence(rPSC) after orthotopic liver transplantation(OLT) in patients with well-preserved colons. METHODS We retrospectively evaluated the medical records of all patients transplanted for PSC in our center between July 1994 and May 2015 and selected 47 with followup of at least 60 mo for further analysis based on strict inclusion and exclusion criteria. rPSC was confirmed by magnetic resonance or endoscopic retrograde cholangiopancreatography and liver biopsy. All patients were evaluated by protocolary pre-OLT colonoscopy with randomized mucosal biopsies. Colonoscopy was repeated annually after OLT. Both organ donors and recipients were human leukocyte antigen(HLA) typed by serological and/or DNA methods. All input data were thoroughly analyzed employing relevant statistical *** Altogether, 31 men and 16 women with a median(range) age of 36(15-68) years at the time of OLT and a median follow-up of 122(60-249) mo were included. rPSC was confirmed in 21/47(44.7%) of patients, a median 63(12-180) mo after transplantation. De novo colitis [rPSC in 11/12, P ≤ 0.05, hazard ratio(HR): 4.02, 95% confidence interval(CI): 1.58-10.98] and history of acute cellular rejection(rPSC in 14/25, P ≤ 0.05; HR: 2.66, 95%CI: 1.03-7.86) showed strong positive associations with rPSC. According to the univariate analysis, overlapping features of autoimmune hepatitis(r PSC in 5/5, P ≤ 0.05) and HLA-DRB1*07 in the donor(r PSC in 10/15, P ≤ 0.05) represent other potential risk factors for rPSC, while the HLA-DRB1*04(rPSC in 0/6, P ≤ 0.05), HLA-DQB1*03(rPSC in 1/11, P ≤ 0.05), and HLA-DQB1*07(rPSC in 0/7, P ≤ 0.05) recipient alleles may have protective *** De novo colitis and acute cellular rejection are clinical conditions significantly predisposed towards recurrence of PSC after liver transplantation.

Distinct gut microbiota profiles in patients with primary sclerosing cholangitis and ulcerative colitis

To characterize the gut bacterial microbiota of patients with primary sclerosing cholangitis (PSC) and ulcerative colitis (UC). METHODSStool samples were collected and relevant clinical data obtained from 106 study participants, 43 PSC patients with (n = 32) or without (n = 11) concomitant inflammatory bowel disease, 32 UC patients, and 31 healthy controls. The V3 and V4 regions of the 16S ribosomal RNA gene were sequenced on Illumina MiSeq platform to cover low taxonomic levels. Data were further processed in QIIME employing MaAsLin and LEfSe tools for analysis of the output data. RESULTSMicrobial profiles in both PSC and UC were characterized by low bacterial diversity and significant change in global microbial composition. Rothia, Enterococcus, Streptococcus, Veillonella, and three other genera were markedly overrepresented in PSC regardless of concomitant inflammatory bowel disease (IBD). Rothia, Veillonella and Streptococcus were tracked to the species level to identify Rothia mucilaginosa, Streptococcus infantus, S. alactolyticus, and S. equi along with Veillonella parvula and V. dispar. PSC was further characterized by decreased abundance of Adlercreutzia equolifaciens and Prevotella copri. Decrease in genus Phascolarctobacterium was linked to presence of colonic inflammation regardless of IBD phenotype. Akkermansia muciniphila, Butyricicoccus pullicaecorum and Clostridium colinum were decreased in UC along with genus Roseburia. Low levels of serum albumin were significantly correlated with enrichment of order Actinomycetales. CONCLUSIONPSC is associated with specific gut microbes independently of concomitant IBD and several bacterial taxa clearly distinguish IBD phenotypes (PSC-IBD and UC).

Increased Heart Rate Variability in Allergic and Highly Trait Anxious Individuals

Autonomic dysregulations were repeatedly observed in people with allergic reactions. Contradictory results of sympathetic over-activation on one hand and parasympathetic predominance on the other, were described for both atopic dermatitis and allergic rhinitis. Secondly, psychological traits of high anxiety and depression described in allergic patients are mostly associated with increased sympathetic and decreased vagal modulation. To address inconsistency in prior findings, we aim to study autonomic nervous system changes in allergic and highly anxious people via HR （heart rate） and HRV （heart rate variability） measures during stressful and relax days in day to day life. We further assess differences between allergic diagnoses and impact of acute symptom occurrence on HR and HRV. 61 subjects （26 allergic, 18 healthy high trait anxious and 17 healthy low trait anxious） took part in this study. They were repeatedly measured for HR by a portable electrocardiographic device, and assessed their subjectively experienced stress during everyday life. HRV was analyzed using time-domain, spectral and non-linear analyses. We observed increased HRV and decreased HR in allergic patients and also in healthy highly anxious subjects, when compared to controls. Both atopic dermatitis and allergic rhinitis patients exhibited parasympathetic predominance or sympathetic withdrawal, seen in increased HRV and lowered HR. Described effects of parasympathetic predominance were, however, not present during acute symptoms occurrence.

Chiral 1,1’-binaphthyl-linked diporphyrin‘tweezers’(R)-1/(S)-1 and the corresponding zinc(II)complexes(R)-2/(S)-2 were prepared as chiral host molecules,and their utility for chiral analyses(especially enantiomeric excess(ee)determinations)were ***(1-n-dodecyl)porphyrins were used for the first time as the interacting *** capabilities of the diporphyrin tweezers were investigated by titrations with(R,R)-and(S,S)-cyclohexane-1,2-diamine(CHDA).The host molecules could be used as multichannel probes of ee by using UVvis,circular dichroism(CD),fluorescence emission and 1H nuclear magnetic resonance(1 H-NMR)*** configurations could also be differentiated using CD or 1HNMR *** three optical techniques give good resolution of ee with reasonable sensitivity considering the low concentrations used(ca.10^-6 mol·L^-1).The ee determination of CHDA enantiomers using NMR spectroscopy is also possible because of the reasonably well separated resonances in the case of(R,R)-and(S,S)-***-metallated(R)-1/(S)-1 hosts could not be used to detect chiral information in a strongly acidic chiral *** work demonstrates the utility of 1,1’-binapthyllinked chiral hosts for chiral analysis of ditopically interacting enantiomers.

采用固态成形工艺生产的热塑性塑料零件,由于材料的粘弹性,与金属锻件相比,出模后尺寸收缩相当严重。特别是在生产象齿轮这类公差要求很严的产品时,模具型腔尺寸的设计就非常困难。但这一问题可以通过台式微型计算机,采用一套简单的人机交换程序进行辅助设计,可使设计工作容易得多。由于出模后塑料制件的尺寸稳定性受到多个工艺参数的制约,本文所介绍的技术则是以事先掌握了样件的尺寸收缩变化规律为基础的。这个样件与所研究的产品有相同的几何尺寸,并在相同的工艺条件下用价廉的初级模具锻出。计算机程序计算出需要的模具型腔形状,并在屏幕上显示,以得到具有正确的渐开线齿廓及尺寸合格的齿轮。计算程序输出的数据可以直接输入数控线切割机床,以便加工高精度的凹模及冲头。
