The HIF-1α/PLOD2 axis integrates extracellular matrix organization and cell metabolism leading to aberrant musculoskeletal repair

While hypoxic signaling has been shown to play a role in many cellular processes,its role in metabolism-linked extracellular matrix(ECM)organization and downstream processes of cell fate after musculoskeletal injury remains to be ***(HO)is a debilitating condition where abnormal bone formation occurs within extra-skeletal *** andhypoxia-inducible factor 1α(HIF-1α)activation have been shown to promote ***,the underlying molecular mechanisms bywhich the HIF-1αpathway in mesenchymal progenitor cells(MPCs)contributes to pathologic bone formation remain to ***,we used a proven mouse injury-induced HO model to investigate the role of HIF-1αon aberrant cell ***-cell RNA sequencing(scRNA-seq)and spatial transcriptomics analyses of the HO site,we found that collagen ECM organizationis the most highly up-regulated biological process in *** mesenchymal cell-specific deletion of Hif1α(Hoxa11-CreER^(T2);Hif1a^(fl/fl))significantly mitigated HO in ***-seq analysis of these Hoxa11-CreER^(T2);Hif1a^(fl/fl)mice identified the PLOD2/LOXpathway for collagen cross-linking as downstream of the HIF-1αregulation of ***,our scRNA-seq data and mechanisticstudies further uncovered that glucose metabolism in MPCs is most highly impacted by HIF-1α*** a translational aspect,a pan-LOX inhibitor significantly decreased HO.A newly screened compound revealed that the inhibition of PLOD2 activity in MPCssignificantly decreased osteogenic differentiation and glycolytic *** suggests that the HIF-1α/PLOD2/LOX axis linked tometabolism regulates HO-forming MPC *** results suggest that the HIF-1α/PLOD2/LOX pathway represents a promisingstrategy to mitigate HO formation.

Integrins and their potential roles in mammalian pregnancy

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(ECM) interactions leading to assembly of integrin adhesion complexes(IACs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. A Composite Index-Based Approach for Mapping Ecosystem Service Production Hotspots and Coldspots for Priority Setting in Integrated Watershed Management Programs

Despite the potential synergism, integrated watershed management and ecosystem services frameworks are rarely used jointly to address the myriad of current water-related issues. The two frameworks are used in this study to spatially identify ecosystem hotspots and coldspots for priority setting in natural resource management programs. Inferred proxies of carbon storage, groundwater supply, surface water supply, and soil retention ecosystem service production potentials were quantified for Texas, U.S., using two complimentary hydro-ecological models, and valued using a non-monetary multi-criteria valuation approach. Maps of individual and composite ecosystem service values showed that several services were co-located and unevenly distributed with most of the high-value hotspots clustered in the eastern part of the state. Individual impacts of land use, climatic and soil properties on the distribution and value of ecosystem services across space were discernable. The study underscored the need for holistic management of landscapes to take advantage of the multiplicity of benefits provided by nature. The approach can readily be incorporated into resource management programs to identify high-value ecosystem service production areas that need conservation, low-value areas that may need restoration, and anthropogenic activities influencing the distribution of ecosystem services.

Evaluating Biomass of Juniper Trees (Juniperus pinchotii) from Imagery-Derived Canopy Area Using the Support Vector Machine Classifier

Both temporal and spatial magnitude, structure, and distribution of rangeland aboveground biomass (AGB) are important inputs for many necessities, in particular for estimating terrestrial carbon amount, ecosystem productivity, climate change studies, and potential bioenergy uses. Much of the remote sensing research previously completed has focused on determining carbon stocks in forested ecosystems with little attention directed to estimate AGB amount in rangelands. Our objectives were to: 1) identify and delineate individual redberry juniper (Juniperus pinchotii) plants from surrounding live vegetation using the support vector machine method for classifying two-dimensional (2D) geospatial imagery with a 1-m spatial resolution at two sites;and 2) develop regression models relating imagery-derived and fieldmeasured single tree canopy area and diameter for dry AGB estimation. The regression results show that there were very close and significant relationships between field measured juniper plant AGB and canopy area derived from the image classification with r2 > 0.90. These results suggest that spectral reflectance recorded on 2D high resolution imagery is capable to assess and quantify AGB as a quick, repeatable, and unbiased method over large land areas.

This paper celebrates Professor Yongqi GAO's significant achievement in the field of interdisciplinary studies within the context of his final research project Arctic Climate Predictions: Pathways to Resilient Sustainable Societies-ARCPATH(https://***/en/projects/finished-projects/arcpath). The disciplines represented in the project are related to climatology, anthropology, marine biology, economics, and the broad spectrum of social-ecological studies. Team members were drawn from the Nordic countries, Russia, China, the United States, and Canada. The project was transdisciplinary as well as interdisciplinary as it included collaboration with local knowledge holders. ARCPATH made significant contributions to Arctic research through an improved understanding of the mechanisms that drive climate variability in the Arctic. In tandem with this research, a combination of historical investigations and social, economic, and marine biological fieldwork was carried out for the project study areas of Iceland, Greenland, Norway, and the surrounding seas, with a focus on the joint use of ocean and sea-ice data as well as social-ecological drivers. ARCPATH was able to provide an improved framework for predicting the near-term variation of Arctic climate on spatial scales relevant to society, as well as evaluating possible related changes in socioeconomic realms. In summary, through the integration of information from several different disciplines and research approaches, ARCPATH served to create new and valuable knowledge on crucial issues, thus providing new pathways to action for Arctic communities.

We discuss the crystal,electronic,and magnetic structures of La_(2−x)Sr_(x)CuO_(4)(LSCO)for x=0.0 and x=0.25 employing 13 density functional approximations,representing the local,semi-local,and hybrid exchange-correlation approximations within the Perdew–Schmidt *** meta-generalized gradient approximation(meta-GGA)class of functionals is found to perform well in capturing the key properties of LSCO,a prototypical high-temperature cuprate *** contrast,the localspin-density approximation,GGA,and the hybrid density functional fail to capture the metal-insulator transition under doping.

Scale Effects of STATSGO and SSURGO on Flow and Water Quality Predictions

Soil information is one of the crucial inputs needed to assess the impacts of existing and alternative agricultural management practices on water quality. Therefore, it is important to understand the effects of spatial scale at which soil databases are developed on water quality evaluations. In the United States, STATSGO (State Soils Geographic) and SSURGO (Soil Survey Geographic) are the most commonly available soil databases. The purpose of this paper was to quantify the effect of scale by employing STATSGO (1:250,000) and SSURGO (1:24,000) soil databases in predicting and comparing flow, sediment, nitrate and phosphorus losses for High Island Creek. This watershed is predominately agricultural and located in south-central Minnesota. The ADAPT (Agricultural Drainage and Pesticide Transport), model was calibrated for flow, sediment, nitrate and phosphorus losses over two years (2001-2002) using STATSGO and SSURGO soil databases. Then the calibrated model was used to evaluate alternative tillage and fertilizer management practices such as adoption of conservation tillage, and rate, timing and method of N- and P-fertilizer applications. Statistical comparison of calibration results with observed data indicated excellent agreement for both soil databases (STATSGO with r2 of 0.95, 0.97, 0.77 and 0.92 and SSURGO with r2 of 0.90, 0.97, 0.82 and 0.99 for flow, sediment, nitrate and phosphorus losses, respectively). However, STATSGO based predictions of annual nitrate-N losses were consistently greater than those with SSURGO database and vice-versa for predicted annual phosphorus losses for the alternative management practice that were evaluated.

Co-Gasification of Mesquite and Coal Blend in an Updraft Fixed Bed Gasifier

In order to reduce the emission of NOx, SOx, and CO2 and mitigate the dependence on the fossil fuel, the use of renewable energy, especially the biomass energy, has been explored. Since most biomass fuels are hard to grind to the very fine size, gasification is the preferred technology of energy conversion. Updraft fixed bed gasification using partial oxidation process is adopted where air less than the stoichiometric quantity is admitted to oxidize the char to CO. The temperature profile within the bed reveals a characteristic temperature peak. The results reveal a correlation between the higher heating value (HHV) of producer gas and the peak gasification temperature (Tpeak). Coal with higher char content (~45%) is blended with biomass of low char content (~20%) in order to produce high quality gas. In this study, the Texas-based mesquite fuel was blended with Wyoming Power River Basin (PRB) coal with mass ratios of 100:0, 90:10, and 80:20 and fired downward into the gasifier. It was found that at a given mesquite to the coal mass ratio, the peak gasification temperature decreased with the increase in ER. With the increase of the coal ratio in the mesquite: coal blend and the peak temperature increased significantly;more combustible gases such as CO, CH4 were generated at the end of product gas, and the HHV of the product gas increased by 10%-20%.
