Genom.c analysis of a novel strain of Bacillus nealsonii, isolated from.Surti buffalo rum.n

Aim. Whole genom. sequencing and functional annotation of Bacillus nealsonii strain AAU1, an am.lolytic anaerobic spore form.ng isolate from.rum.nal contents of buffalo. m.thods and Results: m.rphologically, the strain was observed as slender Gram.ositive rods, occurring in pairs. Optim.l growth was observed at 40°C (range: 30°C to 45°C) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s m.dium.supplem.nted with starch. The m.croorganism.showed extracellular constitutive am.lolytic activity, proving to be capable of utilizing glucose, m.ltose, m.nnose, trehalose, dextrin and starch under anaerobic conditions. Sequence analysis revealed a GC content of 35.1 m.l%. Com.arison of housekeeping gene sequences for RNA polym.rase subunit B (rpoB) and gyrase A (gyrA) identified sequence sim.larity within the Bacillus genus, confirm.d by 16S rRNA gene sequence sim.larity which identified Bacillus nealsonii DSm.15077 as the closest publically available relative. Chem.taxonom.c analysis provided conflicting results with straight-chain saturated C16: 0/C16:0 aldehyde, C16:0 Dm., C14:0 and m.nounsaturated 16:1w7c and 16:1w9c the m.jor fatty acids in contrast to those reported for B. nealsonii DSm.5077. Further characterization using AN-Bilog and physiological param.ters provided genotypic and phenotypic support for taxonom.c classification of isolate AAU1 with published Bacillus species including B. licheniform.s, B. subtilis, B. circulans and B. nealsonii. Conclusion: Based on the data presented, isolate is likely to represent a new strain/subspecies, for which the identifier B. nealsonii AAU1 is proposed. Significance and Im.act of Study: The strict anaerobic conditions prevailing in the bovine rum.n from.where AAU1 was isolated m.y have resulted in genetic polym.rphism.influencing its m.tabolic characteristics.

The developm.nt of efficient single-atom.catalysts(SACs) for the oxygen reduction reaction(ORR)rem.ins a form.dable challenge,prim.rily due to the sym.etric charge distribution of m.tal singleatom.sites(m.N_(4)).To address such issue,herein,Fe-N_(x) sites coupled synergistic catalysts fabrication strategy is presented to break the uniform.electronic distribution,thus enhancing the intrinsic catalytic ***,atom.cally dispersed Fe-N_(x) sites supported on N/S-doped m.soporous carbon(NSC)coupled with FeS@C core-shell nanoparticles(FAS-NSC@950) is synthesized by a facile hydrotherm.l reaction and subsequent *** to the presence of an in situ-grown conductive graphitic layer(shell),the FeS nanoparticles(core) effectively adjust the electronic structure of single-atom.Fe sites and facilitate the ORR kinetics via short/long-range coupling ***,FAS-NSC@950displays a m.re positive half-wave potential(E_(1/2)) of 0.871 V with a significantly boosted ORR kinetics(Tafel slope=52.2 m. dec^(-1)),outpacing the com.ercial Pt/C(E_(1/2)=0.84 V and Tafel slope=54.6 m. dec^(-1)).As a bifunctional electrocatalyst,it displays a sm.ller bifunctional activity param.ter(ΔE) of 0.673 V,surpassing the Pt/C-RuO_(2) com.ination(ΔE=0.724 V).Besides,the FAS-NSC@950-based zincair battery(ZAB) displays superior power density,specific capacity,and long-term.cycling perform.nce to the Pt/C-Ir/C-based *** work significantly contributes to the field by offering a prom.sing strategy to enhance the catalytic activity of SACs for ORR,with potential im.lications for energy conversion and storage technologies.

Peristaltic transport of a m.D Carreau fluid in a tapered asym.etric channel with perm.able walls

The effect of perm.able walls and m.gnetic field on the peristaltic flow of a Carreau fluid in a tapered asym.etric channel is studied. The tapered asym.etric channel is norm.lly created due to the intra-uterine fluid flow induced by m.om.trial contractions and it was sim.lated by asym.etric peristaltic fluid flow in a two-dim.nsional infinite non-uniform.channel. The analysis has been perform.d under long wavelength and low- Reynolds num.er assum.tions to linearize the governing flow equations. A series solution in respect of a sm.ll Weissenberg num.er is obtained for the stream.function, axial pressure gradient and shear stress. Tim. average of pressure rise and frictional force on the upper wall has also been com.uted using num.rical integration. The results have been presented graphically for the various interested physical param.ters. It is observed that for Carreau fluids the peristalsis works as a pum. against a greater pressure rise com.ared with a Newtonian fluid, while there exists no significant difference in free pum.ing flux for Newtonian and Carreau fluids in the tapered asym.etric channel.

Effect of process param.ters on depth of penetration and topography of AZ91 m.gnesium.alloy in abrasive water jet cutting

In the present study,the influence of dynam.c process param.ters such as water pressure,traverse speed and abrasive m.ss flow rate on depth of penetration and surface topography in high strength AZ91 m.gnesium.alloy were investigated using Abrasive Water Jet(AWJ)cutting *** param.ters were varied at 3 levels and influences of each param.ter on penetration ability were identified using analysis of variance(ANOVA).Contribution of water pressure and traverse speed on jet penetration found higher com.ared to abrasive m.ss flow *** projector was used to m.asure depth of *** features and topography of cut surfaces were exam.ned using Scanning Electron m.croscopy(SEm..m.cro cutting and ploughing were observed on the top and bottom.portion of the cut which were sim.lar to that of m.des of deform.tion in other ductile m.terials like alum.nium.and *** roughness of cut surfaces was m.asured using Taylor Hobson surface roughness *** roughness found higher at higher traverse speeds and lower at lower traverse *** study also highlights the suitability of AWJ cutting technology for cutting m.gnesium.and its alloys.

In this paper, we have applied the Lie algebraic m.del to nano-bio m.lecules to determ.ne the vibrational spectra of different stretching and bending vibrational m.des. The determ.ned vibrational energy levels by the Lie algebraic m.del are com.ared with the experim.ntal data. The results from.the theoretical m.de[ are consistent with the experim.ntal data. The vibrational energy levels are clustering in the excited states.

A stability-indicating LC–m./m. m.thod for zidovudine: Identification,characterization and toxicity prediction of two m.jor acid degradation products

Zidvovudine(AZT) is a nucleoside analogue reverse transcriptase inhibitor(NRTI), a class of anti-retroviral drug. A stability-indicating assay m.thod for AZT was developed in line with ICH guideline. Successful separation of AZT and its degradation products was achieved by gradient elution m.de on reverse phase C_(18) colum. using 10 m. am.onium.acetate: acetonitrile as the m.bile phase at 0.8 m./m.n flow rate, 25 μL injection volum., 30 °C colum. tem.erature and 285 nm.detection wavelength. Two m.jor acid degradation products were identified and characterized by liquid chrom.tography–electrospray ionization m.ss spectrom.try(LC–ESI/m./m.) and accurate m.ss m.asurem.nts. The probable m.chanism. for the form.tion of degradation products were identified based on a com.arison of the fragm.ntation pattern of the [m.+ H]^+ions of AZT and its degradation products. One of the degradation products, DP-1, was isolated by sem.-preparative high perform.nce liquid chrom.tography(HPLC) using Waters XBridge Prep C_(18)(250 m.×10 m., 5 μm..Degradation products showed higher toxicity com.ared to the drug in som. m.dels assessed by TOPKAT software. The m.thod validation was perform.d with respect to robustness, specificity, linearity, precision and accuracy as per ICH guideline Q2(R1).

Ultra-High Strength Concrete m.xtures Using Local m.terials

This paper presents the developm.nt of ultra high strength concrete （UHSC） using local m.terials. UHSC m.xture proportions were developed using local m.terials so that UHSC m.y be m.de m.re affordable to a wider variety of applications. Specifically, local sand with a top size of 600 um. and locally available Type I/II cem.nt and silica fum. were used in this research. Each of these m.terial selections is seen as an im.rovem.nt in sustainability for UHSC. Two m.xtures （one with and one without fibers） were recom.ended as the UHSC m.xtures. The greatest com.ressive strengths obtained in this study were 165.6 m.a for UHSC with steel fibers and 161.9 m.a for UHSC without fibers. The com.ressive and flexural strengths obtained from.the UHSC m.xtures developed in this work are com.arable to UHSC strengths presented in the literature. Producing this innovative m.terial with local m.terials reduces the cost of the m.terial, im.roves sustainability, and produces m.chanical perform.nce sim.lar to prepackaged, com.ercially available products.
