目的:研究美洲远志(Polygala senega)根的乙醇提取物对苯并芘致肺癌小鼠的抗肿瘤作用。方法:瑞士白化小鼠被分为5组,每组6只。组1为对照组,予口服橄榄油;组2予苯并芘(50mg/kg体质量,溶于橄榄油中),每周2次,连续4周;组3除予与组2相同的苯并芘外,再予48%乙醇;组4予与组2相同的苯并芘,并同时给予美洲远志的乙醇提取物,每日1次,连续16周;组5仅给予美洲远志的乙醇提取物,每日1次,连续16周。16周后,处死所有小鼠并检测和评价以下指标:2,2-二苯基-1-苦基肼(2,2-diphenyl-1-picrylhydrazyl,DPPH)法测定美洲远志乙醇提取物的抗氧化功效;分别称取各组小鼠的肺组织质量及体质量;彗星实验及酶联免疫吸附法检测DNA损伤情况;分别测定过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶、谷胱甘肽还原酶、脂质过氧化反应及总含硫化合物含量。结果:给予美洲远志乙醇提取物治疗的组4小鼠与组3模型组小鼠相比,体质量明显增加而肺组织质量明显降低(P<0.01)。彗星实验结果显示组4小鼠的DNA损伤较组3小鼠明显减轻(P<0.01)。酶联免疫吸附法测定p53蛋白水平,组4明显高于组3(P<0.01)。组2和组3的模型小鼠与组1比较,脂质过氧化反应水平明显升高,而抗氧化标志物的水平明显降低(P<0.05),组4小鼠的这些指标被明显逆转(P<0.01)。结论:美洲远志对于化学制剂致小鼠肺癌有明确的治疗效果。

目的:东莨菪亭(7-羟基-6-甲氧基香豆素)是从常绿钩吻中提取的有效成分,前期体外研究证实了其抗肿瘤潜能,本研究评价其在小鼠体内的抗肿瘤作用。方法:30只健康小鼠随机分为正常对照组、模型组、溶剂对照组和低、高剂量东莨菪亭组,每组6只。正常对照组小鼠不接受任何干预,其余小鼠背部涂抹100μg 7 ,12-二甲基苯并蒽(7 ,12-di methylbenz[a]anthra-cene ,DMBA)(每周一次)和1 %巴豆油(每周2次)诱导皮肤乳头状瘤,共24周。溶剂组小鼠在造模基础上每天予2 %酒精口服,低剂量(50 mg/kg)和高剂量(100 mg/kg)东莨菪亭组小鼠每天予东莨菪亭治疗。治疗24周后,检测碱性磷酸酶、超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶和谷胱甘肽S-转移酶活性;信号蛋白及其受体,包括芳香烃受体(Aryl hydrocarbon receptor , AhR)、p53、细胞色素P450亚酶1A1(cytochrome P450 1A1 , CYP1A1)、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、信号转导和转录激活因子3(signal transducer and activator of transcription-3 ,Stat-3)、存活素、金属基质蛋白酶2、cyclin D1、c-myc、金属蛋白酶组织抑制因子2和半胱氨酸蛋白酶3(caspase-3) ,采用逆转录聚合酶链反应、蛋白印迹法或免疫沉淀法检测。结果:致癌物DMBA和巴豆油可以诱导毒性反应,生化指标中相关酶活性升高,AhR、CYP1A1、PCNA、Stat-3、存活素、MMP-2、cyclin D1和c-myc表达上调,p53、caspase-3和TI MP-2表达下调。荷瘤小鼠采用东莨菪亭治疗后,生化指标中相关酶的活性下降,蛋白表达和毒性生物标志物恢复正常。结论:东莨菪亭可能通过下调AhR表达来下调一些重要信号蛋白的表达,其作用机制可能是通过竞争性抑制存活素的表达。分裂原活化蛋白激酶可能也起到关键作用。东莨菪亭或许可作为化疗的替代药物用于治疗肿瘤。

Productive Performance and Response of Green Chilli （Capsicum annum L.） to Drip Irrigation Schedules under Water Limited Conditions

Drip irrigation is often preferred under water limited conditions over the other irrigation methods because of its high water application efficiency on account of reduced surface evaporation and percolation losses. The major limitation of drip irrigation is its initial investment. Therefore, there is a need to reduce the cost of drip installation by altering the planting geometry besides enhancing the productivity and moisture use. Results of the two years field study conducted on sandy loam soils revealed that drip irrigation at 1.0 Epan with normal row system （60 cm×30 cm） registered significantly higher yield of green chilli（ 14.08 t ha^-1） which was on par with 0.8 Epan two rows paired system （90-45-90 cm×30 cm：13.45 t ha^-1） and significantly lowest was noticed with 0.6 Epan four rows paired system（45-45-45-120 cm×30 cm ）（5.50 t ha^-1）. The quality parameters viz; TSS and Ascorbic acid were significantly higher with drip irrigation at 1.0 Epan with normal row system and lowest was observed with 0.6 Epan four rows paired system. Total water used was maximum in 1.0 Epan （691.7 mm） followed by 0.8 Epan irrigation （590.2 mm） and the lowest was recorded in 0.6 Epan irrigation （488.8 mm）. There was a saving of water by 14.68 percent and 29.34 percent under 0.8 and 0.6 Epan drip irrigation schedules respectively over 1.0 Epan. Indications of less leaf water potential in 1.0 Epan drip irrigation （-0.58 to-0.67 MPa） followed by 0.8 Epan （-0.61 to-0.68 MPa） and more negative leaf water potential in 0.6 Epan （-0.71 to-0.88 MPa） in relation to water applied were observed. Water use efficiency recorded was maximum in 0.6 Epan normal row systems （24.1 Kg ha-mm^-1） and the least was recorded in 0.6 Epan four rows paired system （11.3 Kg ha-mm^-1）. Morbidity Profile and Causes of Mortality in Type 2 Diabetes Patients: Data from a Tertiary Teaching Hospital from Eastern India

Background: Data regarding the prevalence of morbidity and mortality in patients of Type 2 Diabetes Mellitus (T2DM) is scanty in India. Objectives: To determine the prevalence of micro and macro vascular complications, acute metabolic complications, infections, Non Alcoholic Fatty Liver Disease (NAFLD) and cause of mortality in T2DM patients admitted to a tertiary care teaching hospital in Eastern India. Material and Methods: This was a hospital-based prospective study evaluating 150 T2DM patients admitted to a tertiary care institution in Eastern India. Diagnosis of micro and macro vascular complications, infections and NAFLD was made using standard protocols. In case of death, the most probable cause was noted. Results: Out of 150 patients, 14.7% of patients were newly diagnosed T2DM and out of them 41% of patients had vascular complications and 54.5% had infections. Of the total patients, 56% had nephropathy, 20% neuropathy, 17.3% retinopathy, 31.3% CVD, 11.3% CAD, 4.6% acute metabolic complications, 44% infections and 16.6% had NAFLD respectively. Macrovascular events occured earlier than microvascular complications. Multiple logistic regression analysis showed strong association of age, duration of diabetes, serum cholesterol, triglyceride, LDL-C with retinopathy (Regression coefficient β: -0.1086807, 0.4127152, -0.0513393, 0.0146429, 0.0587475;p < 0.05, < 0.001, < 0.05, < 0.05, < 0.05 respectively), while only duration of diabetes was strongly associated with nephropathy and neuropathy (Regression coefficient β: 0.2538751, 0.2261636;p < 0.001 for each). Increasing age was associated with CAD (Regression coefficient β: 0.055392;p β: 0.0055014;p 18.6% patients died due to diabetes related complications. Cardiovascular (CV)-related deaths (CVD+CAD) were most common cause (51.5%: CVD 36.4%, CAD 15.1%) to be followed by infections (27.3%) and then chronic kidney disease (12.1%). Doxorubicin is associated with adverse effects on organs like liver. This study was done to find the effects of doxorubicin on liver of wistar albino rats. Sixty healthy wistar albino rats were taken for the study. The rats were randomly divided in to 4 groups （2 experimental groups; group AI and A2, and 2 control groups;group Bland B2 each containing 16 rats）. The experimental groups were given a single dose of doxorubicin i.e. 10 mg/kg body weight intraperitoneally and sacrificed after 7 days and 14 days for each group. Rats under control groups were given a single intraperitoneal dose of 2.5 ml/kg body weight normal saline and sacrificed with their respective experimental groups. A significant difference （p 〈 0.001） and （p = 0.005） in final body weight was observed among group A1 experimental and BI control rats, and among group A2 experimental and B2 control rats. As compared to group B2 control, group A2 experimental rats had significantly （p = 0.043） lower liver weight. Diameter ofhepatocyte （p 〈 0.001 for both groups） and nucleus （p=0.004 for group A1 and control BI, and p 〈 0.001 for group A2 and control B2） was significantly higher in experimental rats as compared to their respective controls. Cross-sections of the liver of both control groups had normal architecture. However, there was progressive destruction of liver architecture across the experimental groups. Doxorubicin causes the disruption of normal architecture of liver in rats. Thus care needs to be taken during doxorubicin chemotherapy to minimize effects on river.

The surface energy characteristics of uncoated (clean) and coated stainless steel with UHT milk at various contact time (5 min, 30 min, 1 hours, 3 hours, 6 hours, 24 hours) were determined using contact angle measurement. Whatever the contact time, the clean stainless steel coupons became more hydrophobic and more electron acceptor when they are coated by milk. Inversely, the electron donor character seems to decreasing in this condition. The calculated surface energy component of coated stainless steel was found to vary with contact time. Its hydrophobicity and its electron acceptor were minimal after 3 hours of contact, but its electron donor was minimal after 1 hours of contact. Adhesion experiments of Staphylococcus aureus were carried out on uncoated and coated stainless steels at various contact times. For all contact times, the adhesion results show that milk reduce S. aureus adhesion, and the level of this reduction depend on contact time. This reduction was lower and higher after 1 hours, 5 min and 30 min of contact respectively.

Oxygen Concentration during Reperfusion in Mitral Valve Surgery

Background: By lowering the oxygen fraction of the reperfusate, the reactive oxygen-derived free radicals can be reduced thus facilitating myocardial recovery during weaning from cardiopulmonary bypass and after surgery. Materials & Methods: Thirty patients undergoing mitral valve replacement were randomly exposed to an oxygen fraction of 0.7 (hyperoxic, n = 15) or 0.5 (normoxic, n = 15) during reperfusion. Hemodynamic variables, number of patients requiring additional inotropes and who developed new arrhythmia, duration of ventilation and intensive care unit stay, arterial blood gas and renal function were measured. Results: The demographic data, duration of cardiopulmonary bypass, aortic cross clamp time, duration of mechanical ventilation, intensive care unit stay, additional inotropes, arrhythmia after reperfusion and renal function were similar in both groups. Arterial blood gas analysis was not significantly different, except for the low oxygen partial pressure in the normoxic group during reperfusion. With regard to hemodynamic variables, mean arterial pressure of the hyperoxic group was higher one hour after the cross clamp release. Hemodynamic variables were comparable in all other time periods. Conclusion: By reducing the oxygen concentration during reperfusion, the clinical outcomes in terms of inotropes usage, new arrhythmia after reperfusion, renal function, duration of ventilation and intensive care unit stay were not significantly altered.
