Long-term culture-induced phenotypic difference and efficient cryopreservation of small intestinal organoids by treatment timing of Rho kinase inhibitor

To investigate a suitable long-term culture system and optimal cryopreservation of intestinal organoid to improve organoid-based therapy by acquiring large numbers of *** were isolated from jejunum of C57BL/6 mouse. Two hundred crypts were cultured in organoid medium with either epidermal growth factor/Noggin/R-spondin1 (ENR) or ENR/CHIR99021/VPA (ENR-CV). For subculture, organoids cultured on day 7 were passaged using enzyme-free cell dissociation buffer (STEMCELL Technologies). The passage was performed once per week until indicated passage. For cryopreservation, undissociated and dissociated organoids were resuspended in freezing medium with or without Rho kinase inhibitor subjected to different treatment times. The characteristics of intestinal organoids upon extended passage and freeze-thaw were analyzed using EdU staining, methyl thiazolyl tetrazolium assay, qPCR and time-lapse live cell *** established a three-dimensional culture system for murine small intestinal organoids using ENR and ENR-CV media. Both conditions yielded organoids with a crypt-villus architecture exhibiting Lgr5+ cells and differentiated intestinal epithelial cells as shown by morphological and biochemical analysis. However, during extended passage (more than 3 mo), a comparative analysis revealed that continuous passaging under ENR-CV conditions, but not ENR conditions induced phenotypic changes as observed by morphological transition, reduced numbers of Lgr5+ cells and inconsistent expression of markers for differentiated intestinal epithelial cell types. We also found that recovery of long-term cryopreserved organoids was significantly affected by the organoid state, i.e., whether dissociation was applied, and the timing of treatment with the Rho-kinase inhibitor Y-27632. Furthermore, the retention of typical morphological characteristics of intestinal organoids such as the crypt-villus structure from freeze-thawed cells was observed

目的:了解中韩两国居家老年人抑郁水平和生活满意度的差异性及相关因素。方法:采用自填式问卷,调查中国厦门市201名与韩国大田市206名60岁以上的居家老年人,问卷包括一般人口学资料、抑郁量表(GDS)及生活满意度量表(LSI-A)。应用两个独立样本t检验分析中韩老年人的GDS和LSI-A是否存在差异;采用多元逐步回归分析分别探索影响中韩老年人GDS及LSI-A的相关因素。结果:大田市老年人的LSI-A得分高于厦门市老年人(P=0.008);集体活动(β=0.37)、居住方式(β=-0.30)、受教育程度(β=0.16)与中国老年人GDS相关,集体活动(β=-0.36)、居住方式(β=0.17)与LSI-A相关;健康状态(β=-0.33)、集体活动(β=0.24)、居住方式(β=-0.13)与韩国老年人GDS相关,健康状态(β=0.32)、集体活动(β=-0.15)、居住方式(β=0.16)与LSI-A相关。结论:韩国大田市老年人生活满意度优于中国厦门市老年人。

A case of colon perforation due to enteropathy-associated T-cell lymphoma

Enteropathy-associated T-cell lymphoma (EATL) is an extremely rare disease, which is often related to gluten-sensitive enteropathy. It is an uncommon intestinal lymphoma with very poor prognosis and high mortality rate. In the absence of specific symptoms or radiological findings, it is difficult to diagnose early. Major complications of EATL have been known as intestinal perforation or obstruction, and only 5 cases of EATL are reported in South Korea. In this study, we report a case of 71-year-old male with symptoms of diarrhea, which later it progressed into cancer perforation of the colon. The initial colonoscopic findings were normal and computed tomography scan demonstrated a segmental wall thickening of the distal ascending colon with nonspecific multiple small lymphnodes, along the ileocolic vessels, but no signs of mass or obstruction. The histologic findings of resected specimen confirmed EATL type II. Patient expired two weeks after the operation. Therefore, we emphasize the need of random biopsy in the presence of normal mucosa appearance on colonoscopy for the early diagnosis of EATL.

Dear Editor,Plant tissue culture involves callus formation and de novo shoot ***,explants from differentiated tissues are used to generate a pluripotent cell mass,called callus,on auxin-rich callus-inducing medium(CIM),followed by shoot regeneration on cytokinin-rich shoot-inducing medium(SiM).Callus results from division of pericycle-like cells(Atta et al.,2009;Sugimoto et al.,2010);its cellular identity resembles that of lateral root primordia(Atta et al.,2009;Sugimoto et al.,2010).Callus acquires cellular pluripotency by forming root stem cell niches on CIM(Sugimoto et al.,2010)。

Distant skeletal muscle metastasis from intrahepatic cholangiocarcinoma presenting as Budd-Chiari syndrome

Intrahepatic cholangiocarcinoma is a malignant neoplasm arising from the biliary epithelium, which frequently invades adjacent organs or metastasizes to other visceral organs such as the lungs, bones, adrenals, and brain. However, distant skeletal muscle metastasis of cholangiocarcinoma has never been described before to the best of our knowledge and, furthermore, Budd-Chiari syndrome secondary to intrahepatic cholangiocarcinoma is also extremely rare. Here we present the first case overall of distant muscle metastasis from intrahepatic cholangiocarcinoma presenting as Budd-Chiari syndrome. A 44-year-old man admitted to the hospital with complaints of abdominal distension, edema of both legs, back pain and anorexia of 30 d' duration. Computed tomography and ultrasonography-guided percutaneous muscle biopsy established intrahepatic cholangiocarcinoma with disseminated thrombosis from inferior vena cava to bilateral iliac and femoral veins, and multiple skeletal muscle metastases in bilateral buttock and erector spinal muscle.

Cornual patency and integrity following laparoscopic cornuotomy for interstitial pregnancy

Objective: To evaluate cornual patency and integrity following laparoscopic cornuotomy. Study Design: This is a prospective cohort study on seven women who underwent laparoscopic cornuotomy for interstitial pregnancy. The cornual patency and integrity were evaluated using hysterosalpingography (HSG) and magnetic resonance imaging (MRI). On MRI, the entire cornual wall thickness were measured bilaterally at 5-mm intervals. The thickness of the affected and unaffected cornua matched at the corresponding contralateral point in each of the women. Statistically, all possible pairwise 28 comparisons were compared using paired t-tests. Results: Among six eligible women, four women had excellent cornual patency on the affected side. Among seven women (i.e., 28 pairs), no significant difference was observed in cornual thickness compared to the unaffected cornu and no remarkable defects were seen in the endometrial and cornual contour. But there is no statistical significance. Conclusion: Laparoscopic cornuotomy seems to have advantage to preserve the cornu in interstitial pregnancy.

Comparison of double pants with single pants on satisfaction with colonoscopy

AIM: To increase satisfaction and diminish anxiety and shame during colonoscopy, we developed novel double pants (NDP) which consist of doubled fabrics with an inner hole. The aim of study was to compare satisfaction, anxiety and shame between NDP and conventional single pants (CSP). METHODS: Total 160 consecutive examinees were randomly divided into NDP and CSP group. Before colonoscopy, questionnaires identifying state and trait anxiety were completed. After colonoscopy, questionnaires for overall satisfaction (Group Health Association of America 9) and pants-specific satisfaction (5-20), state anxiety (20-80), and shame (6-24) were interviewed. RESULTS: Pants-specific satisfaction scores regarding willingness to repeat colonoscopy using same pants (3.3 ± 0.8 vs 2.1 ± 0.9, P < 0.001) and recommendation of same pants to other people (3.3 ± 0.7 vs 2.0 ± 1.0, P < 0.001) were significantly higher in NDP than CSP groups. State anxiety (33.0 ± 7.0 vs 35.4 ± 6.9, P = 0.028) and shame (6.6 ± 1.5 vs 8.1 ± 3.2, P = 0.001) after colonoscopy was lower in NDP group compared with CSP group. CONCLUSION: The NDP contribute to increase satisfaction and decrease anxiety and shame after colonoscopy.

AIM:Smallanthus sonchifolius(Yacon)has been used as a food and folk medicine in the Andes *** was screened the pharmacological effects of a 50% ethanol ex- tract of Yacon on spermatogenesis in *** study is carded out to evaluate the possibility of using Yacon tubers extracts as a fertility agent to treat infertility caused by spermatogenic disorders. METHODS:The dried,pulverized Yacon tubers were extracted with 50% ethanol at 50℃ for 5 h under sonica- *** Yacon extracts were analyzed by using HPLC system on a Kromasil ClS column(250mm×4.6 mm). The Yacon tubers extracts,chlorogenic acid and ferulic acid were administrated to rats once daily by oral gavage. The number of sperm heads in the testis was assessed by counting the number of sperm heads using slight modifica- tion of the procedure reported by Toth et al(1989). RESULTS:After administration of chlorogenic acid and Yacon tuber extracts to rat for 5 weeks,the number of sperm in epididymis were increased by 20.33% and 34.48%,*** femlic acid which was report- ed as a metabolite of chlorogenic acid and constituent of Yacon to estimate the spermatogenic activity was also ad- *** administration of ferulic acid and Yacon tubers extracts,the sperm number were increased by 37.87% and 43.33%,*** ef- fects of chlorogenic acid and ferulic acid were 0.59 times and 0.87 times of Yacon tubers extracts,respectively. CONCLUSION:The spermatogenic effect of Yacon tu- bers extracts may result from the combined effect of chloro- genic acid or its analogues and their metabolites.

Monsoonal precipitation variation in the East Asia since A.D. 1840 ——Tree-ring evidences from China and Korea

Three tree-ring rainfall reconstructions from China and Korea are used in this paper to investigate the East Asian summer monsoon-related precipitation variation over the past 160 years. Statistically, there is no linear correlation on a year-by-year basis between Chinese and Korean monsoon rainfall, but region-wide synchronous variation on a decadal-scale was observed. More rainfall intervals were 1860-1890, 1910-1925, and 1940-1960, and dry or even drought peri-ods were 1890-1910, 1925-1940, and 1960-present. Reconstructions also display that the East Asian summer monsoon precipitation suddenly changed from more into less around mid-1920. These tree-ring precipitation records were also confirmed by Chinese historical dry-ness/wetness index and Korean historical rain gauge data.

Limitations of urease test in diagnosis of pediatric Helicobacter pylori infection

The diagnosis of Helicobacter pylori(H. pylori) infection is usually based on the results of urease test and histology. The urease test known as a simple and cheap method does not need special skills to perform or to read the result. The time needed for the test to turn positive depends on the concentration of bacteria, and the accuracy is up to the density of H. pylori density in the biopsy sample, which is generally lower in children than adolescents and adults. Therefore, there are debates about the sensitivity of the urease test in children. The reason for lower sensitivity of the urease test in children was not identified, but might be related to the low density and patchy distribution of bacteria. In this review, we discuss the limitations of the urease test in children according to age, histology, number of biopsy samples, and biopsy site. In children under 5 years old, the differences in positivity rate when the urease test used one or three biopsy samples, and samples from the antrum or the gastric body, were larger than those in children aged 5-15 years. Thus, three or more biopsy samples from both the antrum and body would improve the sensitivity of H. pylori infection diagnosis in children under 5 years old.
