s.rong>Introduction:s.rong> Hepatitis.C virus.(HCV) infection is.a worldwide public health problem.with m.ltis.s.em.c involvem.nts.including kidneys. We as.es.ed the prevalence and determ.nants.of kidney m.rkers.am.ng HCV infected s.bjects. s.rong>m.thods.s.rong> A cros.-s.ctional s.udy was.conducted from.m.rch to July 2017 in two referral centers.in Yaoundé. s.ciodem.graphic, clinical and paraclinical data were collected from.patient’s.records. Renal involvem.nt was.evaluated us.ng es.im.ted glom.rular filtration rate (eGFR) by m.dification of Diet in Renal Dis.as.’s.(m.RD) equation, urine dips.ick and album.n/creatinine ratio (ACR). An eGFR m./m.n/1.73 m.s.p>2s.p> was.defined as.low and reduced res.ectively. Album.nuria was.defined by an ACR > 30 m./g and divided into A1 (m./g), A2 (30 - 300 m./g) and A3 (>300 m./g). s.rong>Res.lts.s.rong> We included 65 (41.5% m.les. HCV infected patients.with a m.an age of 56.8 ± 10.5 years. HCV infection was.diagnos.d for m.re than 5 years.in 54 (83.1%) patients. HCV viral load and genotype were available in 40 (61.5%) patients.viral load was.high (>5.9 logs.m.) in 20 (50%) of them.and genotypes.4 (40%), 1 (32.5%) and 2 (27.5%) were found in this.population. The m.an eGFR was.112.9 ± 31.9 m./m.n/1.73 m.s.p>2s.p> with 14 (21.5%) and 4 (6.2%) patients.having reduced and low eGFR res.ectively. Album.nuria (80%), leukocyturia (24.6%) and hem.turia (4.6%) were the obs.rved dips.ick abnorm.lities. m.dian album.nuria (IQR) was.542.4 (238.7 - 961.5) m./g, with 7 (11%), 12 (18%) and 46 (71%) patients.in A1, A2 and A3 res.ectively. Reduced/low eGFR and album.nuria were obs.rved in 58 (89.2%) patients.while low eGFR and album.nuria were found in 55 (84.6%) patients. AIm. To evaluate the antiviral potency of a new antihepatitis.C virus.HCV) antiviral agent targeting the cellular autophagy m.chinery. m.THODs. Non-infected liver s.ices. obtained from.hum.n liver res.ction and cut in 350 μm.thick s.ices.2.7 × 106 cells.per s.ice) were infected with cell culture-grown HCV Con1b/C3 s.pernatant(m.ltiplicity of infection = 0.1) cultivated for up to ten days. HCV infected s.ices.were treated at day 4 pos.-infection with GNs.396 for 6 d at different concentrations. HCV replication was.evaluated by s.rand-s.ecific real-tim. quantitative revers. trans.ription- polym.ras. chain reaction. The infectivity titers.of s.pernatants.were evaluated by foci form.tion upon inoculation into naive Huh-7.5.1 cells. The cytotoxic effect of the drugs.was.evaluated by lactate dehydrogenas. leakage as.ays. REs.LTs. The antiviral efficacy of a new antiviral drug, GNs.396, an autophagy inhibitor, on HCV infection of adult hum.n liver s.ices.was.evidenced in a dos.dependent m.nner. At day 6 pos.-treatm.nt, GNs.396 EC50 was.158 nm.l/L without cytotoxic effect(com.ared to hydroxychloroquine EC50 = 1.17 μm.l/L).CONCLUs.ON: Our res.lts.dem.ns.rated that our ex vivo m.del is.efficient for evaluation the potency of autophagy inhibitors. in particular a new quinoline derivative GNs.396 as.antiviral could inhibit HCV infection in a dos.dependent m.nner without cytotoxic effect.
