Integrated genomic analysis for prediction of survival for patients with liver cancer using The Cancer Genome Atlas

AIM To evaluate the prognostic power of different molecular data in liver *** Cox regression screen and least absolute shrinkage and selection operator were performed to select significant prognostic variables. Then the concordance index was calculated to evaluate the prognostic power. For the combination data, based on the clinical cox model, molecular features that better fit the model were combined to calculate the concordance index. Prognostic models were built based on the arithmetic summation of the significant variables. Kaplan-Meier survival curve and log-rank test were performed to compare the survival difference. Then a heatmap was constructed and gene set enrichment analysis was performed for pathway *** The m RNA data were the most informative prognostic variables in all kinds of omics data in liver cancer, with the highest concordance index(C-index) of 0.61. For the copy number variation, methylation and mi RNA data, the combination of molecular data with clinical data could significantly boost the prediction accuracy of the molecular data alone(P < 0.05). On the other hand, the combination of clinical data with methylation, mi RNA and m RNA data could significantly boost the prediction accuracy of the clinical data itself(P < 0.05). Based on the significant prognostic variables, different prognostic models were built. In addition, the heatmap analysis, survival analysis, and gene set enrichment analysis validated the practicability of the prognostic *** In all kinds of omics data in liver cancer, the m RNA data might be the most informative prognostic variable. The combination of clinical data with molecular data might be the future direction for cancer prognosis and prediction.

Individualized Chemotherapy in Advanced NSCLC Patients Based on mRNA Levels of BRCA1 and RRM1

Objective： Experimental evidence suggests that the overexpression of breast cancer-specific tumor suppressor protein 1 （BRCA1） gene enhances sensitivity to docetaxel and resistance to cisplatin and ribonucleotide reductase M1 （RRM1） gene overexpression enhances resistance to gemcitabine. To further examine the effect of BRCA1 and RRM1 mRNA levels on outcome in advanced non-small cell lung cancer （NSCLC）, we performed this non-randomized phase II clinical trial which tested the hypothesis that customized therapy would confer improved outcome over non-customized therapy. Methods： RNA was isolated from fresh tumor tissue. Patients received chemotherapy regimen based on their BRCA1 and RRM1 mRNA levels： both low-cisplatin plus gemcitabine （GP）; both high-vinorelbine plus cisplatin （NP）; BRCA1 low and RRM1 high-cisplatin plus docetaxel （TP）; BRCA1 high and RRM1 low-vinorelbine plus gemcitabine （GN）. Results： From Dec 2005 to Nov 2008, 94 metastatic and locally advanced NSCLC patients from our institute were enrolled in this study. The median age was 58 years old. Among them, 21 patients received GP, 30 patients received TP and 43 patients received NP chemotherapy. GP group had a higher response rate, and longer median time to progression （TTP） and median overall survival （OS） time than the other 2 groups. The response rates in the GP, TP and NP groups were 42.9%, 36.7% and 27.9%, respectively （P=0.568）. The median TTP was 5.6, 5.0, 4.8 months （P=0.975）, respectively, and the median OS time was 12.5, 11.0, 9.7 months （P=0.808）, respectively. Conclusion： Chemotherapy customized according to BRCA1 and RRM1 expression levels is associated with higher response rate and longer TTP and OS time in the GP group. This suggests that BRCA1 and RRM1 mRNA levels could be used as biomarkers in individual therapy in NSCLC.

Helicobacter Pylori and Gastric Cancer： Clinical Aspects

Objective： Although Helicobacterpylori （H. pylori） is considered as the main etiological factor for gastric cancer, the strategy of screening and treating the oncogenic bacterium is still controversial. The objective was to evaluate the status and progress of the cognition about the relationship between 11. pylori infection and gastric cancer from a clinical aspect. Data Sourees： The data used in this review were mainly from the PubMed articles published in English from 1984 to 2015. Study Selection： Clinical research articles were selected mainly according to their level of relevance to this topic. Results： Gastric cancer is the fifth most common malignancy and the third leading cause of cancer deaths worldwide. The main etiological factor for gastric cancer is H. pylori infection. About 74.7-89.0% gastric cancer was related to H. pylori infection. Up to date, some regional gastric cancer prevention programs including the detection and treatment of H. pylori infection are under way. Current data obtained from the randomized controlled trials suggest that population-based H. pylori screening and treatment is feasible and cost-effective in preventing gastric cancer; however, a population-based H. pylori eradication campaign would potentially lead to bacterial resistance to the corresponding antibiotics, as well as a negative impact on the normal flora. Conclusions： The important questions of feasibility, program costs, appropriate target groups for intervention, and the potential harm of mass therapy with antibiotics must first be answered before implementing any large-scale program.

Application of DNA barcoding to the identification of Hymenoptera parasitoids from the soybean aphid （Aphis glycines） in China

Aphis glycines Matsumura is an important pest of soybean in Asia and North America. Hymenoptera parasitoids play a key role in the control of the soybean aphid. Thecorrect identification of parasitoids is a critical step that precedes the assessment of their potential biological control agents. Accurate identification of the majority of the speciesattacking the soybean aphid often requires elaborate specimen preparation and expert taxonomic knowledge. In this study, we facilitated the identification of soybean aphidparasitoids by applying a DNA barcoding approach following a preliminary morphological identification. We generated DNA sequence data from the mitochondrial COI gene andthe D2 region of 28S rDNA to assess the genetic variation within and between parasitoid species emerging from the soybean aphid in China. Fifteen Hymenoptera parasitoid speciesbelonging to 10 genera of five families were identified with little intra-specific variation （0.09% ± 0.06% for 28S and 0.36% ± 0. 18% for COI） and large inter-specific divergence（30.46% ± 3.42% for 28S and 20.4% ± 1.20% for COI）.

The lattice parameters, elastic constants, cohesive energy, structural energy differences, as well as the properties of point defects and planar defects of hexagonal closepacked yttrium （hcpY） have been studied with ab initio density functional theory for constructing an ex tensive database. Based on an analytical bondorder poial scheme, empirical manybody interatomic potential for hcpY has been developed. The model is fitted to some properties of Y, e.g., the lattice parameters, elastic constants, bulk modulus, cohesive energy, vacancy formation energy, and the structural energy differences. The present potential has ability to reproduce defect properties including the selfinterstitial atoms formation energies, vacancy formation energy, divacancy binding energy, as well as the bulk properties and the thermal dynamic properties.

Assessment on the Vulnerability of Agricultural Eco-environment of Hilly Basin Area in the Middle of Hunan Province——A Case Study of Hengyang Basin

[Objective] The study aimed to assess the vulnerability of agricultural eco-environment of hilly basin area in the middle of Hunan Prov- ince. [ Method] Taking Hengyang basin as an example, we firstly chose 13 indicators from natural, social and economic subsystem to assess the vulnerability of agricultural eco-environment in hilly basin area in the middle of Hunan Province. Afterwards, by using principal component analysis, we calculated the weight of each indicator and the vulnerability of agricultural eco-environment of seven regions, finally analyzed the spatial distribu- tion of agricultural eco-envimnment vulnerability. [ Result] The weight of farmers＇ net income per capita was up to 0.140, followed by forest coverage （0.137）. Among the seven counties, the vulnerability of agricultural eco-environment was primarily moderate, while Hengnan County had the highest vulnerability, followed by Hengyang County, and their vulnerability was extremely serious. In addition, Leiyang City was the minimum fragile region, namely slightly fragile region. In general, the vulnerability of peripheral counties was lower than that of central counties. [ Conclusion] The research could provide scientific references for the restoration and reconstruction of agro-ecological environment as well as the establishment of agri- cultural Dreduction decisions.

Effects of helium irradiation dose and temperature on the damage evolution of Ti3SiC2 ceramic

The effects of 400 keV helium ion irradiation dose and temperature on the microstructure of the Ti3SiC2 ceramic were systematically investigated by grazing incidence x-ray diffraction, scanning electron microscopy, and transmission electron *** helium irradiation experiments were performed at both room temperature(RT) and 500℃ with a fluence up to 2.0 × 1017 He+/cm2 that resulted in a maximum damage of 9.6 displacements per *** results demonstrate that He irradiations produce a large number of nanometer defects in Ti3SiC2 lattice and then cause the dissociation of Ti3SiC2 to TiC nano-grains with the increasing He *** induced cell volume swelling of Ti3SiC2 at RT is slightly higher than that at 500℃, suggesting that Ti3SiC2 is more suitable for use in a high temperature *** temperature dependence of cell parameter evolution and the aggregation of He bubbles in Ti3SiC2 are different from those in *** formation of defects and He bubbles at the projected depth would induce the degradation of mechanical performance.

A new method of quantitative pre-corrosion damage of aviation aluminium(Al-Cu-Mg)alloy was proposed,whichregarded corrosion pits as equivalent semi-elliptical surface *** analytical model was formulated to describe the entire regionof fatigue crack propagation(FCP).The relationship between the model parameters and the fatigue testing data obtained in thepre-corroded experiments,crack propagation experiments and S-N fatigue experiments was *** equivalent crack sizesand the FCP equation were used to calculate the fatigue life through numerical integration based on MATLAB/*** resultsconfirm that the sigmoidal curve fitted by the FCP model expresses the whole change from Region I to Region *** addition,thepredicted curves indicate the actual trend of fatigue life and the conservative result of fatigue ***,the new analytical methodcan estimate the residual life of pre-corroded Al-Cu-Mg alloy,especially smooth specimens.
