Mucus-penetrating nonviral gene vaccine processed in the epithelium for inducing advanced vaginal mucosal immune responses

Establishment of vaginal immune defenses at the mucosal interface layer through gene vaccines promise to prevent infectious diseases among *** barriers composed of a flowing mucus hydrogel and tightly conjugated epithelial cells(ECs),which represent the main technical difficulties for vaccine development,reside in the harsh,acidic human vaginal *** from frequently employed viral vectors,two types of nonviral nanocarriers were designed to concurrently overcome the barriers and induce immune *** design concepts include the charge-reversal property(DRLS)to mimic a virus that uses any cells as factories,as well as the addition of a hyaluronic acid coating(HA/RLS)to directly target dendritic cells(DCs).With a suitable size and electrostatic neutrality,these two nanoparticles penetrate a mucus hydrogel with similar *** DRLS system expressed a higher level of the carried human papillomavirus type 16 L1 gene compared to HA/RLS in *** it induced more robust mucosal,cellular,and humoral immune ***,the DLRS applied to intravaginal immunization induced high IgA levels compared with intramuscularly injected DNA(naked),indicating timely protection against pathogens at the mucus *** findings also offer important approaches for the design and fabrication of nonviral gene vaccines in other mucosal systems.

Magnetic nanoparticles(MNPs)have become a research hotspot and widely used in the biomedical field in recent decades due to their unique magnetic *** minireview summarizes the specific gene transfection of magnetic particles(magnetofection)during eversy dynamic process of gene delivery(gene binding,cellular uptake,endosomal escape,intracellular trafficking and in vivo targeting).Meanwhile,the synergistic biomedical application of magnetofection and the effects of MNPs have also been discussed,including magnetic resonance imaging(MRI),magnetic mediated hyperthermia(MMH),Fenton reaction and ***,the clinical prospect of magnetofection was briefly expected.

Inhibition roles of molybdate and borate on Q235 steel corrosion in resistance reducing agent

The effects of Na_(2)MoO_(4) and Na_(2)B_(4)O_(7) on corrosion behavior of Q235 steel in resistance reducing agent(RRA)containing sodium bentonite were studied by mass loss,scanning electron microscopy and electrochemical *** results showed that both the independent and mixed additions of Na_(2)MoO_(4) and/or Na_(2)B_(4)O_(7),can reduce the corrosion rate of Q235 steel in RRA containing sodium *** the inhibition effect of Na_(2)MoO_(4) and/or Na_(2)B_(4)O_(7)increased with their dosage *** the same dosage,the inhibition efficiency of the mixed addition of Na_(2)MoO_(4) and Na_(2)B_(4)O_(7),was higher than that of their independent *** passivation effect of Q235 steel was easy to obtain in the RRA with the mixed addition of Na_(2)MoO_(4) and Na_(2)B_(4)O_(7).The optimized inhibitor for the RRA containing sodium bentonite was the mixture of Na_(2)B_(4)O_(7) and Na_(2)MoO_(4) with a total concentration of 1.5 wt.%.Furthermore,the increase in corrosion potential E_(corr) and the decrease in corrosion current density i_(cor) in carbon steel were one of the important criteria for the formation of passivation film.

Understanding the stability and reactivity of ultrathin tellurium nanowires in solution： An emerging platform for chemical transformation and material design

The stability and reactivity of nanomaterials are of crucial importance for their application, but the long-term effects of stability and reactivity of nanomaterials under practical conditions are still not well understood. In this study, we first established a comprehensive strategy to investigate the stability of a highly reactive nanomaterial from the viewpoint of reaction kinetics with ultrathin tellurium nanowires （TeNWs） as a model material in aqueous solution through an accelerated oxidation process. This allowed us to propose a new approach for the design and synthesis of other unique one-dimensional nanostructures by a chemical transformation process using the intermediate nanostructures ＂captured＂ during the dynamic oxidation process under different conditions. In essence, the oxidation of ultrathin TeNWs is a gas-solid reaction which involves liquid, gas and solid phases. It has been demonstrated that the oxidation process of ultrathin TeNWs in aqueous solution can be divided into three stages, namely oxygen limiting, ultrathin TeNWs limiting and mass transfer resistance limiting stages. The apparent oxidation kinetics for ultrathin TeNWs is approximately in accord with a first order reaction kinetics model and has an apparent activation energy as low as TeNWs are thermodynamically unstable 13.53 ***^-1, indicating that ultrathin However, the unstable nature of ultrathin TeNWs is actually an advantage since it can act as an excellent platform to help us synthesize and design one-dimensional functional nanomaterials--with special structures and distinctive properties--which are difficult to obtain by a direct synthesis method.

Third-order optical nonlinearities and dynamic responses of two imino squaramides under neutral and base conditions were studied using the femtosecond degenerate four-wave mixing technique at *** optical responses have been observed and the magnitude of the second-order hyperpolarizabilities of the squaramides has been measured to be as large as 10 31 esu. The absorption spectra,color of solution,and third-order optical nonlinearities of two imino squaramides change with the addition of sodium *** γ value under the base condition for each dye is approximately 1.25 times larger than that under neutral conditions.

Traditional Chinese Medicine comprehensive therapy for the improvement of motor function in spinal cord injury patients

OBJECTIVE: To study the effect of early comprehensive therapy of Traditional Chinese Medicine(TCM) on motor function of in patients with spinal cord ***: Fifty-one standard spinal cord injury patients with paraplegia were randomly assigned to an experimental or control group. The experimental group received TCM comprehensive therapy, and the control group received modern Western Medicine(WM) treatment for 4 weeks. The motor score(MS), Barthel Index(BI) and American Spinal Injury Association(ASIA) grading were measured in both groups before and after ***: After treatment, the MS and BI scores of the TCM comprehensive therapy group improved significantly(P 0.05). The differences between the experimental and control groups after treatment were not significant(P > 0.05).CONCLUSION: Early TCM comprehensive therapy is an effective method for improving motor function in patients with spinal cord injury.

Cloning and Functional Analysis of the Bifunctional Agglutinin/Trypsin Inhibitor from Helianthus tuberosus L.

In order to find new insect resistance genes, four homologous cDNAs, hta-a, hta-b, hta-c and hta-d with lengths of 775, 718, 784 and 752 bp, respectively （GenBank accession numbers AF477031-AF477034）, were isolated from a tuber cDNA expression library of Helianthus tuberosus L. Sequence analysis revealed that all four cDNAs contain an open reading frame of 444 bp, coding a polypeptide of 147 amino acid residues, and that the sequences of the cDNAs are very similar to those of the mannose-binding agglutinin genes of the jacalin-related family. In hemagglutination reactions and hapten inhibition assays, affinity-purified HTA （Helianthus tuberosus agglutinin） from induced Escherichia coli BL21（DE3） expressing GST-HTA shows hemagglutination ability and a higher carbohydrate-binding ability for mannose than other tested sugars. Trypsin inhibitory activity was detected in the crude extracts of induced E. coli BL21（DE3） expressing HTA, and was further verified by trypsin inhibitory activity staining on native polyacrylamide gel. The mechanism of interaction between HTA and trypsin was studied by molecular modeling. We found that plenty of hydrogen bonds and electrostatic interactions can be formed between the supposed binding sites of HTA-b and the active site of trypsin, and that a stable HTA/trypsin complex can be formed. The results above imply that HTA might be a bifunctional protein with carbohydrate-binding activity and trypsin inhibitory activity. Moreover, Northern blotting analysis demonstrated that hta is predominantly expressed in tubers of H. tuberosus, very weakly expressed in stems, but not expressed at all in other tissues. Southern blotting analysis indicated that hta is encoded by a multi-gene family. The insect resistance traits have been described in another paper.

Effect of Inoculation with Arbuscular Mycorrhizal Fungus on Nitrogen and Phosphorus Utilization in Upland Rice-Mungbean Intercropping System

The effect of arbuscular mycorrhiza fungi （AMF） on plant growth and nutrition utilization in upland rice and mungbean intercropping system was studied. A pot experiment was conducted in the greenhouse and AMF colonization rates of rice and mungbean roots, plant nutrient contents, the ability of nitrogen fixation, and nutrient contents changed in the soil were analyzed. The results were obtained as follows： the rates of AMF colonization of rice and mungbean roots were reached to 14.47 and 92.2% in intercopping system, and increased by 4.11 and 11.95% compared with that of in monocropping; the nirtrogen contents of mungbean and rice were increased by 83.72 and 64.83% in shoots, and 53.76 and 41.29% in roots, respectively, while the contents of iron in shoot and root of mungbean were increased by 223.08 and 60.19%, respectively. In the intercropping system with inoculation of AMF, the biomass of mungbean increased by 288.8%. However, the biomass of rice was not significantly changed among all treatments with or without inoculation of AMF recorded. The number and dry weight of nodules were significantly increased either when mungbean was intercropped with rice or inoculated with AMF. When compared with that of monocropping without AMF inoculation, the contents of nitrogen, phosphorus and iron in nodules of intercropping mungbean with inoculation increased by 80.14, 69.54 and 39.62%, respectively. Additionally, intercropping with AMF inoculation significantly increased soil nitrogen content, but reduced soil phosphorus content. We concluded that upland rice-mungbean intercropping system and inoculation with AMF improved the nutrient uptake, the ability of nitrogen fixation and the growth of mungbean.

Overlaying welding technology and performance of axle tube remanufacturing

Screw failure is one of the main scrap forms for rear axle tube, and thread turning after surfacing is a common means of remanufacturing. This paper takes rear axle tube as the main research topic, which is made of 40Mn and provided by an axle company. Manual electric arc welding, CO2 shielded arc welding and argon shielded arc welding are carried to overlay the damaged thread. The deposited metal consists of two welding material lays. Welding process and overlay properties are characterized by the test and analysis of microstructure, residual stress and hardness of surfacing layers. The results show that argon shielded arc welding is an effective method to repair the failure thread; its microstructure of fusion area is meticulous and uniform; its residual compressive stress （absolute value） of welding surface is the biggest among the three welding methods; its hardness curve is relatively fiat and appropriate for turning.

Preparation and Anti-ultraviolet Aging Performance of Organic Layered Double Hydroxides/Bitumen Composites

Layered double hydroxides(LDHs) with the physical property of high ultraviolet(UV)reflectance were used to enhance the anti-UV aging performance of bitumen. In view of the poor compatibility of LDHs with bitumen, three organic anions, namely, sodium dodecyl benzenesulfonate(SDBS), sodium dodecyl sulfate(SDS) and sodium dodecyl sulfonate(SDSO), were used as modifiers to prepare organic LDHs(OCLDHs) through regeneration process, and the crystal structure, chemical composition, morphological feature and UV shielding capability of synthesized OCLDHs were analyzed. Then the OCLDHs were added into bitumen to evaluate the storage stability and anti-UV aging property of OCLDHs/bitumen *** results show that the organic anions are inserted into the interlayers of LDHs, the intercalation expands the interlayer distance of LDHs, makes the particle shapes become more irregular and complicated, and enhances the absorption ability within the range from 200 to 300 nm while has little influence on the UV reflection *** of high temperature storage stability indicates the organic modification ameliorates the compatibility of LDHs with bitumen. Compared with LDHs, OCLDHs decrease the deteriorations of bitumen’s properties during UV exposure test, exhibiting better effect in enhancing anti-UV aging performance of bitumen. Furthermore,among the three OCLDHs, LDHs intercalated by SDBS exhibit the most effective improvement due to the best compatibility with bitumen and comparatively good UV shielding effect.
