Proliferating cell nuclear antigen (PCNA) is the core component of replication complex in eukaryote. As a processive factor of DNA polymerase delta, PCNA coordinates the replication process by interacting with various replication proteins. PCNA appears to play an essential role in many cell events, such as DNA damage repair, cell cycle regulation, and apoptosis, through the coordination or organization of different partners. PCNA is an essential factor in cell proliferation, and has clinical significance in tumor research. In this article we review the functional structure of PCNA, which acts as a function switch in different cell events.

Medium spiny neurons(MSNs)in the striatum,which can be divided into D1 and D2.MSNs,originate from the lateral ganglionic eminence(LGE).Previously,we reported that Six3.is a downstream target of Sp8/Sp9 in the transcriptional regulatory cascade of D2.MSN development and that conditionally knocking out Six3.leads to a severe loss of D2.***,we showed that Six3.mainly functions in D2.MSN precursor cells and gradually loses its function as D2.MSNs *** deletion of Six3.had little effect on cell pro{2.ration but blocked the differentiation of D2.MSN precursor *** addition,conditional overexpression of Six3.promoted the differentiation of precursor cells in the *** measured an increase of apoptosis in the postnatal striatum of conditional Six3.knockout *** suggests that,in the absence of Six3.abnormally differentiated D2.MSNs are eliminated by programmed cell *** results further identify Six3.as an important regulatory element during D2.MSN differentiation.

Objective:The interrelationship between apoptosis and autophagy plays an important role in many pathophysiological processes,however,whether their interplay is involved in allergic contact dermatitis(ACD)has not yet been ***,we conducted this study to determine whether keratinocyte-specific autophagy-related gene 5(ATG5)deficiency can regulate apoptosis to inhibit skin damage in mice with 2.4-dinitrochlorobenzene(DNCB)-induced ***:This study involved keratinocyte-specificAtg5 conditional knockout(cKO)mice(Krt14cre/+-Atg5flox/flox)and control mice(Krt14+/+-Atg5flox/flox).We painted DNCB on the right ear of each mouse to induce *** scoring and measurements of ear weight and thickness were performed to evaluate inflammation *** immunohistochemical assay was performed to analyze immune cell *** study and TUNEL staining were performed to compare the differences in skin lesions betweenAtg5 cKO mice and control *** and western blotting were used to examine the levels of ATG5 and apoptosis-related *** results were statistically analyzed byt ***:After DNCB stimulation of mice ears,we observed a more severe phenotype inAtg5 cKO mice than in control mice(dermatitis score:7.500±2.588vs.3.2.0±0.82.,P=0.003..Further analysis of ATG5 protein confirmed keratinocyte-specific ablation ofAtg5 in cKO mice and showed that DNCB did not influence ATG5 *** assay revealed that the infiltrated immune cells were not involved in aggravation of the phenotype of DNCB-stimulatedAtg5 cKO ***,the histological study(P=0.02.),TUNEL staining(P=0.02.),immunofluorescence(P=0.03.),and western blotting showed that the increase in keratinocyte death,especially apoptosis,contributed to aggravation of the phenotype of DNCB-stimulatedAtg5 cKO ***:Deficiency ofAtg5 in keratinocytes increases apoptosis,aggravating skin damage in DNCB-induced ACD *** has no relation
