Antineoplastons are peptide and amino acid derivatives that occur naturally in the human body. They inhibit the growth of neoplastic cells without growth inhibition of normal cells. Phenylacetylglutaminate (PG) is an active ingredient of antineoplastons A10 and As2-1 (ANP) and is also a metabolic by-product of phenylbutyrate (PB). The formulation of antineoplaston As2-1 is a 4:1 mixture of phenylacetate (PN) and PG. Antineoplaston A10 is a 4:1 mixture of PG and isoPG. This study investigates the molecular mechanism of action of PG and PN. The Human U87 glioblastoma (GBM) cell line was used as the model system in this study. A total human gene array screen using the Affymetrix Human Genome plus 2.0 oligonucleotide arrays was performed using mRNA derived from U87 cells exposed to PG and PN. Pathway analysis was performed to allow the visualization of effect on metabolic pathways and gene interaction networks. Our preliminary results indicate that PG and PN interrupt signal transduction in RAs/MAPK/ERK and PI3K/AKT/PTEN pathways, interfere with cell cycle, decrease metabolism and promote apoptosis in human U87 GBM cells. The effect on multiple cellular pathways and targets, suggests that ANP and PB are promising candidates for clinical studies in GBM.

Mental health symptoms secondary to trauma exposure and substance use disorders(sUDs)co-occur frequently in both clinical and community *** possibility of a shared aetiology remains an important question in translational *** in genetics,basic science,and neuroimaging have led to an improved understanding of the neural basis of these disorders,their frequent comorbidity and high rates of relapse remain a clinical *** project aimed to conduct a review of the field’s current understanding regarding the neural circuitry underlying posttraumatic stress disorder and sUD.A comprehensive review was conducted of available published literature regarding the shared neurobiology of these disorders,and is summarized in detail,including evidence from both animal and clinical *** summarizing the relevant literature,this review puts forth a hypothesis related to their shared neurobiology within the context of fear processing and reward *** provides an overview of brain reward circuitry and its relation to the neurobiology,symptomology,and phenomenology of trauma and substance *** review provides clinical insights and implications of the proposed theory,including the potential development of novel pharmacological and therapeutic treatments to address this shared *** and extensions of this theory are discussed to provide future directions and insights for this shared phenomena.

Floods are one of nature's most destructive disasters because of the immense damage to land,buildings,and human *** is difficult to forecast the areas that are vulnerable to flash flooding due to the dynamic and complex nature of the flash ***,earlier identification of flash flood susceptible sites can be performed using advanced machine learning models for managing flood *** this study,we applied and assessed two new hybrid ensemble models,namely Dagging and Random subspace(Rs)coupled with Artificial Neural Network(ANN),Random Forest(RF),and support Vector Machine(sVM)which are the other three state-of-the-art machine learning models for modelling flood susceptibility maps at the Teesta River basin,the northern region of *** application of these models includes twelve flood influencing factors with 413 current and former flooding points,which were transferred in a GIs *** information gain ratio,the multicollinearity diagnostics tests were employed to determine the association between the occurrences and flood influential *** the validation and the comparison of these models,for the ability to predict the statistical appraisal measures such as Freidman,Wilcoxon signed-rank,and t-paired tests and Receiver Operating Characteristic Curve(ROC)were *** value of the Area Under the Curve(AUC)of ROC was above 0.80 for all *** flood susceptibility modelling,the Dagging model performs superior,followed by RF,the ANN,the sVM,and the Rs,then the several benchmark *** approach and solution-oriented outcomes outlined in this paper will assist state and local authorities as well as policy makers in reducing flood-related threats and will also assist in the implementation of effective mitigation strategies to mitigate future damage.

Light quality response is a vital environmental cue regulating plant development. Conifers, like angiosperms, respond to the changes in light quality including the level of red (R) and far-red (FR) light, which follows a latitudinal cline. R and FR wavelengths form a significant component of the entire plant life cycle, including the initial developmental stages such as seed germination, cotyledon expansion and hypocotyl elongation. With an aim to identify differentially expressed candidate genes, which would provide a clue regarding genes involved in the local adaptive response in scots pine (Pinus sylvestris) with reference to red/far-red light;we performed a global expression analysis of scots pine hypocotyls grown under two light treatments, continuous R (cR) and continuous FR (cFR) light;using Pinus taeda cDNA microarrays on bulked hypocotyl tissues from different individuals, which represented different genotypes. This experiment was performed with the seeds collected from northern part of sweden (Ylinen, 68?N). Interestingly, gene expression pattern with reference to cryptochrome1, a blue light photoreceptor, was relatively high under cFR as compared to cR light treatment. Additionally, the microarray data analysis also revealed expression of 405 genes which was enhanced under cR light treatment;while the expression of 239 genes was enhanced under the cFR light treatment. Differentially expressed genes were re-annotated using Blast2GO tool. These results indicated that cR light acts as promoting factor whereas cFR antagonises the effect in most of the processes like C/N metabolism, photosynthesis and cell wall metabolism which is in accordance with former findings in Arabidopsis. We propose cryptochrome1 as a strong candidate gene to study the adaptive cline response under R and FR light in scots pine as it shows a differential expression under the two light conditions.

AIM To assess overall confidence level of trainees in assessing and treating shock, we sought to improve awareness of recurrent biases in clinical decision-making to help address appropriate educational *** Pediatric trainees on a national listserv were offered the opportunity to complete an electronic survey anonymously. Four commonly occurring clinical scenarios were presented, and respondents were asked to choose whether or not they would give fluid, rank factors utilized in decision-making, and comment on confidence level in their decision. REsULTs Pediatric trainees have a very low confidence level for assessment and treatment of shock. Highest confidence level is for initial assessment and treatment of shock involving American College of Critical Care Medicine/Pediatric Advanced Life support recommendations. Children with preexisting cardiac comorbidities are at high risk ofunder-resuscitation. CONCLUsION Pediatric trainees nationwide have low confidence in managing various shock states, and would benefit from guidance and teaching around certain common clinical situations.

Bacterial Flora of Water and Rotifers in Outdoor Mass Culture Tanks Fed with Different Microalagal Diets

Investigations were carried out on the bacterial flora of water and the quality of rotifers in outdoor mass culture tanks. Brachionus plicatilis and B. rotundiformis were cultured in 10 numbers of 1 tonne tanks and the physico-chemical parameters and microbial loads of water, and total bacterial and Vibrio loads of microalgae and rotifers, during the period 2014-15, were studied. The study revealed a significant variation of the ammonia levels and total bacterial loads with different diets （P 〈 0.05）. A positive correlation was observed between the total bacterial loads of algal concentrations and loads of rotifers, but no correlation was observed between Vibrio loads of water and rotifers. The prevalence of Vibrio loads of rotifers was low in tanks fed with Nannochloropsis oculata and maximum Vibrio loads were recorded in Chaetoceros calcitrans fed rotifers. The Vibrio loads significantly varied with days of culture and also with the algal diets. A positive correlation was observed between the total Vibrio loads of water and rotifers.

successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MsCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MsCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the sirtuin-1 pathway are contributing factors to aging at the cellular level of both MsCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. Advanced age is the primary risk factor for a fracture, due to the low bone mass and inferior bone quality associated with aging;a better understanding of the dysfunctional behavior of the aging cell will provide a foundation for new treatments to decrease healing time and reduce the development of complications during the extended recovery from fracture healing in the elderly.
