Improved method of plasma 8-Isoprostane measurement and association analyses with habitual drinking and smoking

AIM: To develop a simple and accurate method for quantifying 8-isoprostane in plasma by employing a combination of two-step solid-phase extraction of samples and a commercially available ELISA kit, and by this method to examine the effects of drinking and smoking habits against the levels of plasma 8-isoprostane in healthy Japanese ***: Plasma 8-isoprostane was extracted with ODS gel suspension followed by NH2 Sep-Pak column. The 8-isoprostane fractions were assayed using a commercially available ELISA kit. We measured plasma 8-isoprostane levels in 157 healthy Japanese volunteers divided into three groups (64 non-habitual drinkers, 56 moderate drinkers and 37 habitual drinkers) according to their alcohol consumption per week. Genotypes of aldehyde dehydrogenase 2 (ALDH2) were also determined to investigate the plasma 8-isoprostane levels with reference to drinking habits. In addition, the plasma 8-isoprostane levels of 96 non-smokers and 61 smokers from the same subjects were ***: Our method fulfilled all the requirements for use in routine clinical assays with respect to sensitivity, intra- and inter-assay reproducibility, accuracy and dynamic assay range. Significant increases of plasma 8-isoprostane levels were observed in female habitual drinkers when compared with those of non-habitual drinkers (t = 5.494, P < 0.0001) as well as moderate drinkers (t = 3.542, P < 0.005), and 8-isoprostane levels were also significantly different between ALDH2*2/1 and ALDH2*1/1 in the female habitual drinkers (t = 6.930, P < 0.0001), suggesting that excessive drinking of alcohol may increase oxidization stress, especially in females. Prediction of Aerodynamic Noise in a Ring Fan Based on Wake Characteristics

A ring fan is a propeller fan that applies an axial-flow impeller with a ring-shaped shroud on the blade tip side. In this study, the entire flow field of the ring fan is simulated using computational fluid dynamics (CFD); the accuracy of the CFD is verified through a comparison with the aerodynamic characteristics of a propeller fan of current model. Moreover, the aerodynamic noise generated by the fan is predicted on the basis of the wake characteristics. The aerodynamic characteristic of the ring fan based on CFD can represent qualitatively the variation in the measured value. The main flow domain of the ring fan is formed at the tip side of the blade because blade tip vortex is not formed at that location. Therefore, the relative velocity of the ring fan is increased by the circumferential velocity. The sound pressure levels of the ring fan within the frequency band of less than 200 Hz are larger than that of the propeller fan. In the analysis of the wake characteristics, it revealed that Karman vortex shedding occurred in the main flow domain in the frequency domain lower than 200 Hz ; the aerodynamic noise of the ring fan in the vortex shedding frequency enlarges due to increase in the relative velocity and the velocity fluctuation.

Influence of Diffuser on Aerodynamic Noise of a Forward Curved Fan

In order to clarify the influence of a diffuser on the charactersfics of a forward curved fan, the influence of the bare ratio and the outlet angle on the characteristics of the fan were measured through an experiment performed with an actual fan as well as through a numerical simulation, respectively. The mechanism of the discrete fre- quency noise generated by the separated flow of the diffuser was analyzed. The optimized bare ratio was ap- proximately 17%. The flow separated inside of the diffuser generated discrete frequency noise owing to the inter- action between the reversed flow from the diffuser and the impeller rotating at the blade passing frequency. The diffuser outlet angle influenced the pressure ratio more than that by the bare ratio. Furthermore, it was confirmed that restraining the separation in the diffuser effectively decreases the fan noise.

In order to clarify the mechanism by which aerodynamic noise is generated from separated flow around an airfoil blade,the relation between the attack angle and the aerodynamic noise of the blade was analyzed using a wind tunnel experiment and a CFD *** the case of rear surface separation,the separated vortex which has a large-scale structure in the direction of the blade chord is transformed into a structure that concentrates at the trailing edge with an increase in the attack *** aerodynamic noise level then becomes small according to the vortex scale in the blade *** the flow is separated at the leading edge,a separated vortex of low pressure is formed at the vicinity of the trailing *** pressure fluctuations on the blade surface at the vicinity of the trailing edge become large due to the vortex in the *** is considered that the aerodynamic noise level increases when the flow is separated at the leading edge because the separated vortex is causing the fluctuations due to wake vortex shedding.

Extracapsular invasion as a risk factor for disease recurrence in colorectal cancer

AIM: To evaluate the presence of extracapsular invasion (ECI) in positive nodes as a predictor of disease recurrence disease in colorectal cancer. METHODS: Two hundred and twenty-eight consecutive patients who underwent colorectal resection were identified for inclusion in this study, of which 46 had positive lymph nodes. Among 46 cases with stage Ⅲcolorectal cancer, 16 had ECI at positive nodes and 8 had disease recurrence. The clinical and pathological features of these cases were reviewed. RESULTS: In the univariate analysis, the number of positive lymph nodes and depth of tumor invasion were significantly associated with the presence of ECI at positive nodes. Multivariate analysis demonstrated that only ECI was a predictor of recurrence. The recurrence-free interval differed significantly among patients with ECI at positive ***: Our results suggest that ECI at metastatic nodes can identify which cases are at high risk of short-term disease recurrence in colorectal cancer.

Effects on coagulation factor production following primary hepatomitogen-induced direct hyperplasia

AIM： To investigate the molecular mechanisms involved in coagulation factor expression and/or function during direct hyperplasia （DH）-mediated liver regeneration. METHODS： Direct hyperplasia-mediated liver regeneration was induced in female C57BL/6 mice by administering 1,4-bisr2-（3,5-dichloropyridyloxy）] benzene （TCPOBOP）, a representative hepatomitogen. Mice were weighed and sacrificed at various time points [Day 0 （D0： prior to injection）, 3 h, D1, D2, D3, and D10] after TCPOBOP administration to obtain liver and blood samples. Using the RNA samples extracted from the liver, a comprehensive analysis was performed on the hepatic gene expression profiling of coagulation-related factors by real-time RT-PCR （fibrinogen, prothrombin, factors Ⅴ, Ⅶ, Ⅷ, Ⅸ, Ⅹ, Ⅺ, Ⅻ, ⅩⅢβ , plasminogen, antithrombin, protein C, protein S, ADAMTS13, and VWF）. The corresponding plasma levels of coagulation factors （fibrinogen, prothrombin, factors Ⅴ, Ⅶ, Ⅷ, Ⅸ, Ⅹ, Ⅺ, Ⅻ, ⅩⅢ, and VWF） were also analyzed and compared with their mRNA levels. RESULTS： Gavage administration of TCPOBOP （3 mg/kg body weight） resulted in a marked and gradual increase in the weight of the mouse livers relative to the total body weight to 220% by D10 relative to the DO （control） ratios. At the peak of liver regeneration （D1 and D2）, the gene expression levels for most of the coagulationrelated factors （fibrinogen, prothrombin, factors Ⅴ, Ⅶ, Ⅷ, Ⅸ,Ⅺ, Ⅻ, ⅩⅢβ, plasminogen, antithrombin, protein C, ADAMTS13, VWF） were found to be downregulated in a time-dependent manner, and gradually recovered by D10 to the basal levels. Only mRNA levels of factor X and protein S failed to show any decrease during the regenerative phase. As for the plasma levels, 5 clotting factors （prothrombin, factors Ⅷ, Ⅸ, Ⅺ, and Ⅻ） demonstrated a significant decrease （P 〈 0.05） during the regeneration phase compared with DO.
