Expression profiling of transgenes(Cry1Ac and Cry2A) in cotton genotypes under different genetic backgrounds

Transgenic cotton carrying the CrylAc gene has revolutionized insect pest control since its adoption,although the development of resistance in insect pests has reduced its *** 10 years of cultivating Bacillus thuringiensis(Bt)cotton with a single Cry1 Ac gene,growers are on the verge of adopting Bt cotton that carries the double gene(Cry1 Ac+Cry2 A)due to its better effectiveness against insect ***,the current study was designed to evaluate the role of each gene in the effectiveness of Bt cotton carrying the double *** expression levels of the Cry1 Ac and Cry2 A genes were evaluated in the leaves of 10 genotypes(2 parents and 8 Fhybrids)at 30 days after sowing(DAS),while samples of leaves,bolls and flowers were taken from the upper and lower canopies at 70 and 110 *** Fhybrids were developed through reciprocal crosses between two Bt(CKC-1,CKC-2)and two non-Bt(MNH-786,FH-942)*** differential expression of transgenes was evaluated through Enzyme Linked Immuno-Sorbent Assay(ELISA).The results showed that the MNH786 xCKC-1 hybrid had the highest concentrations of Cry1 Ac gene at30 DAS(3.08μg g^(-1))and 110 DAS(1.01μg g^(-1))in *** contrast,the CKC-2 xMNH-786 hybrid showed the lowest concentrations of Cry1 Ac gene at 30 DAS(2.30μg g^(-1))and 110 DAS(0.86μg g^(-1)).The Fhybrid FH-942×CKC-2 showed the highest concentrations of Cry2 A gene at 30 DAS(8.39μg g^(-1))and 110 DAS(7.74μg g^(-1))in leaves,while the CKC-1 xMNH-786 hybrid expressed the lowest concentrations of Cry2 A gene at 30 DAS(7.10μg g^(-1))and 110 DAS(8.31μg g^(-1)).A comparison between the two stages of plant growth showed that leaves had the highest concentrations at 30 DAS,whereas the lowest concentrations were observed at 110 DAS for both genes in *** the expression pattern was compared between various plant parts in genotype CKC-2,it was found that leaves had higher concentrations of Cry1 Ac(3.12μg g^(-1))and Cry2 A(8.31μg g^(-1))at 70 DAS,followed by bolls

In this research study, we have synthesized the bio-capped ZnO/g-C_(3)N_(4) nanocomposites by employing lemon juice(Citrus limon) as a stabilizer and mediator. Fruitfully, lemon juice which contains various acidic functional groups and citric acid has the capability to block the surface of g-C_(3)N_(4) from chemical reactivity and activated the surface of g-C_(3)N_(4) for various reactions. Consequently, the agglomeration behavior and controlled shape of g-C_(3)N_(4) has also been achieved. Our experimental results i.e. XRD,TEM, HRTEM, PL, FS, XPS, and PEC have confirmed that the lemon juice mediated and green g-C_(3)N_(4)(L-CN) have good performances and remarkable visible light photocatalytic activities as compared to the chemically synthesized g-C_(3)N_(4)(CN). Furthermore, the small surface area and low charge separation of g-C_(3)N_(4) is upgraded by coupling with Zn O nanoparticles. It is proved that the coupling of Zn O worked as a facilitator and photoelectron modulator to enhance the charge separation of g-C_(3)N_(4). Compared to pristine lemon-mediated green g-C_(3)N_(4)(L-CN), the most active sample 5Zn O/L-CN showed ~ 5-fold improvement in activities for ciprofloxacin(CIP) and methylene blue(MB) degradation. More specifically,the mineralization process and degradation pathways, and the mineralization process of ciprofloxacin(CIP) and methylene blue(MB) are suggested. Finally, our present novel research work will provide new access to synthesize the eco-friendly and bio-caped green g-C_(3)N_(4)nanomaterials and their employment for pollutants degradation and environmental purification.

Randomly amplified polymorphic DNA(RAPD)is a tremendously convenient approach used to discriminate between Brassica species owing to its accuracy and *** primers generate adequate genetic information that can be used in the primer-marker *** this work,twenty RAPD-PCR based markers were executed to generate polymorphic data,like polymorphic information content(PIC),mean resolving power(MRP),resolving power(RP),effective multiplex ratio(EMR),and marker index(MI)for the first time and genetic distance among and between six Brassica species were *** results indicated that 20 primers produced a total of 231 scored band and generated 87%polymorphic *** PIC,MRP,RP,MI,and EMR values were 0.088,0.65,6.7,0.78,and 8.9,*** showed an overall negative correlation with MRP,RP,MI,and EMR,whereas MRP,RP,and EMR,were positively correlated with each *** identities ranged from 41.99%(between Brassica napus and Brassica oleracea)to 68.83%(between Brassica campestris and Brassica oleracea).Dendrogram results showed no clustering between species except between Brassica campestris and Brassica ***,these results will be helpful to acquire useful information about the markers and their use to determine the genomic structures of Brassica ***,based on genetic distance and polymorphic information,new hybrids can be developed for effective oilseed production.

With the rapid development of urban power grids and the large-scale integration of renewable energy, traditional power grid fault diagnosis techniques struggle to address the complexities of diagnosing faults in intricate power grid systems. Although artificial intelligence technologies offer new solutions for power grid fault diagnosis, the difficulty in acquiring labeled grid data limits the development of AI technologies in this area. In response to these challenges, this study proposes a semi-supervised learning framework with self-supervised and adaptive threshold (SAT-SSL) for fault detection and classification in power grids. Compared to other methods, our method reduces the dependence on labeling data while maintaining high recognition accuracy. First, we utilize frequency domain analysis on power grid data to filter abnormal events, then classify and label these events based on visual features, to creating a power grid dataset. Subsequently, we employ the Yule–Walker algorithm extract features from the power grid data. Then we construct a semi-supervised learning framework, incorporating self-supervised loss and dynamic threshold to enhance information extraction capabilities and adaptability across different scenarios of the model. Finally, the power grid dataset along with two benchmark datasets are used to validate the model’s functionality. The results indicate that our model achieves a low error rate across various scenarios and different amounts of labels. In power grid dataset, When retaining just 5% of the labels, the error rate is only 6.15%, which proves that this method can achieve accurate grid fault detection and classification with a limited amount of labeled data.

This study pertains to the evaluation of shale gas and rock physics properties of this area with respect to its total organic content of Sember Formation, Khiproarea, Pakistan. We use well logs data for this study. The Khipro area is prominent in the Lower Indus Basin for its hydrocarbon (oil and gas) structural traps. In shale gas evaluation, TOC of Sember Formation is estimated. The analysis has been done with the help of the wire line data of the well Bilal North-01. The presence of shale gas in the study area is analyzed with the help of different techniques. Rock physics and petrophysical analysis have been done in order to get the properties of the area related to the shale gas evaluation.

Multi-energy synergy systems integrating high-penetration large-scale plug-in electric vehicles, distributed renewable energy generations, and battery energy storage systems have great potential to reduce the reliance of the grid on traditional fossil fuels. However, the random charging characteristics of plug-in electric vehicles and the uncertainty of photovoltaics may impose an additional burden on the grid and affect the supply–demand equilibrium. To address this issue, judicious scheduling optimization offers an effective solution. In this study, considering charge and discharge management of plug-in electric vehicles and intermittent photovoltaics, a novel Multi-energy synergy systems scheduling framework is developed for solving grid instability and unreliability issues. This formulates a large-scale mixed-integer problem, which calls for a powerful and effective optimizer. The new binary level-based learning optimization algorithm is proposed to address nonlinear large-scale high-coupling unit commitment problems. To investigate the feasibility of the proposed scheme, numerical experiments have been carried out considering multiple scales of unit numbers and various scenarios. Finally, the results confirm that the proposed scheduling framework is reasonable and effective in solving unit commitment problems, can achieve 3.3% cost reduction and demonstrates superior performance in handling large-scale energy optimization problems. The integration of plug-in electric vehicles, distributed renewable energy generations, and battery energy storage systems is verified to reduce the output power of 192.72 MW units during peak periods to improve grid stability. Therefore, optimizing energy utilization and distribution will become an indispensable part of future power systems.

Balkassar is an old field with considerable remaining potential. The 2d, 3d seismic and earlier vintages show that Balkassar is composed of two folds that impart heart shaped geometry. It appears likely the early Eocene age Bhadrar formation may provide commercial production with lower water cuts from the eastern lobe (yet untapped) of the structure that may have at least 30 million barrels of unrecovered oil. Horizontal drilling may have promise as an optimum technique for recovery of oil from Paleogene reservoirs. Encouraging oil indications have also been recorded from the early Permian age tobra formation from Balkassar oxy-1 that was not tested by Oxy. Tobra sandstone reservoir can have a matrix porosity approaching 8%. When fractured recoveries from it can be relatively better than the Paleogene reservoirs. The 2D and 3D seismic acquisition has confirmed that the eastern lobe (yet untapped) of the Balkassar field is structurally higher and steeper than the Western lobe which has thus far produced over 30 million barrels. The Eastern lobe thus offers good potential for recovery of oil from the Bhadrar reservoir. The entire field is likely to have potential for recovery of oil from the early Eocene aged Tobra formation. 2-d and 3-D Seismic data interpretation, attribute analysis and visualization for deeper prospect carried in Balksasar field. Tobra and Khewra formation studied for deeper potential drilling. Time contour and depth contour map shows potential for deeper prospects. Also attribute analysis and 3d visualization show good results for deeper potential of Tobra and Khewar formations. Seismic amplitude, Reflection strength, Apparent polarity attribute are visualized and interpreted to find the potential for Tobra and Khewra formation. 3-D visualization also showed positive results for Tobra and Khewra formations.

The main purpose of this research is to estimate the structural analysis and hydrocarbon potential of Miano Block by using seismic and well log techniques. Miano area hosts a number of gas fields with structural and stratigraphic traps. The area is located in Central Indus Basin which is a part of an extensional regime exhibiting normal faulting due to the split of the Indian Plate firstly from Africa and then from Madagascar and Seychelles. Miano area recognized as a proven petroleum province which has complex tectonic history of Cretaceous extensional and overprints of Tertiary strike-slip tectonics. The area has prospect with accumulation of hydrocarbons in structural and stratigraphic traps including pinchouts. NW-SE oriented Khairpur and Mari Highs are main structural features with impact on the fault system. The sands of Lower Goru of Lower Cretaceous age are acting as a reservoir in the area. The area has great potential of hydrocarbons for which more exploratory wells are required to be drilled with better insight of structural and stratigraphic traps.
