XBAT35, a Novel Arabidopsis RING E3 Ligase Exhibiting Dual Targeting of Its Splice Isoform., Is Involved in Ethylene-m.diated Regulation of Apical Hook Curvature

The Arabidopsis XBAT35 is one of five structurally related ankyrin repeat-containing Really Interesting New Gene （RING） E3 ligases involved in ubiquitin-m.diated protein degradation, which plays key roles in a wide range of cel- lular processes. Here, we show that the XBAT35 gene undergoes alternative splicing, generating two transcripts that are constitutively expressed in all plant tissues. The two splice variants derive from.an exon skipping event that excludes an in-fram. segm.nt from.the XBAT35 precursor m.NA, giving rise to two protein isoform. that differ solely in the presence of a nuclear localization signal （NLS）. Transient expression assays indicate that the isoform.lacking the NLS localizes in the cytoplasm.of plant cells, whereas the other is targeted to the nucleus, accum.lating in nuclear speckles. Both isoform. are functional E3 ligases, as assessed by in vitro ubiquitination assays. Two insertion m.tant alleles and RNA-interference （RNAi） silencing lines for XBAT35 display no evident phenotypes under norm.l growth conditions, but exhibit hyper- sensitivity to the ethylene precursor 1-am.nocyclopropane-l-carboxylate （ACC） during apical hook exaggeration in the dark, which is rescued by an inhibitor of ethylene perception. Independent expression of each XBAT35 splice variant in the m.tant background indicates that the two isoform. m.y differentially contribute to apical hook form.tion but are both functional in this ethylene-m.diated response. Thus, XBAT35 defines a novel player in ethylene signaling involved in negatively regulating apical hook curvature, with alternative splicing controlling dual targeting of this E3 ubiquitin ligase to the nuclear and cytoplasm.c com.artm.nts.

Characterization of the Topography and Wettability of English Weed Leaves and Biom.m.tic Replicas

The topography and wettability of the underside of English weed （Oxalis pes-caprae） leaves and of their biom.m.tic replicas are investigated. Polyvinyl siloxane m.lds were cast from.the leaves and then filled with an epoxy pre-polym.r to produce replicas. The particular topographical structures of leaves and replicas were evaluated by optical m.croscopy and Scanning Electron m.croscopy （SEm. analysis. The static wettability of leaves and replicas was assessed by contact angle m.asurem.nts, while the dynam.c wettability was characterized by estim.ting contact angle hysteresis and studying the dynam.c behavior of im.acting water droplets. A sm.oth glass slip and its replica were used as control surfaces. The replica m.ulding m.thod used was able to transfer the characteristic pattern of irregular 100 μm.- 200 μm.× 60 μm.convex papillae interspersed with stom.ta of the original leaf to the epoxy replicas. The static contact angle of 143°± 3° and the contact angle hysteresis of 2~ indicate that the underside of the English weed leaf is close to superhydrophobic. The lower contact angles （130° ± 4°） and higher hysteresis （31°） observed for the replica when com.ared with the original leaves were associated to an inaccurate replication of the chem.stry and structures of the three-dim.nsional wax projections covering the plant surface. Also, trichom.s in the original leaves could not be accurately reproduced due to their flexibility and fragility. Differences in wetting behavior were also evident from.droplet im.act experim.nts, with rebound regim.s prevailing in the original leaves and regim.s characterized by higher adhesion and larger dissipation predom.nating in the replicas. Nevertheless, the m.rphological features of the leaf transferred to the replica were sufficient to prom.te a clear hydrophobic behavior of the replica when com.ared with the sm.oth epoxy reference surface.

The kinetics and m.chanism.of natural wolfram.te interactions with sodium.carbonate during air heating were studied.X-ray phase and X-ray m.croanalysis were used to establish that the initial m.nocrystalline wolfram.te consists of Fe0.5 m.0.5 WO4 and Fe0.3 m.0.7 *** therm.l analysis showed that the interaction of wolfram.te with sodium.carbonate begins above 450°C with the form.tion of tungstate,sodium.ferrite,iron oxides,and *** experim.nts on sintering with the subsequent rem.val of water-soluble com.ounds(leaching)tracked the change in the structure of *** atom.c ratio of Fe/m. in wolfram.te does not change up to 600°C,and subsequently decreases to 0.2 during heating,which allows the m.chanism.of the process to be identified and indicates the greater reactivity of wolfram.tes with an increased proportion of *** analysis with data processing using non-isotherm.l kinetics established that the interaction of wolfram.te with sodium.carbonate in an air stream.proceeds via a two-stage m.chanism.wherein the first stage is lim.ted by diffusion(activation energy,E=243 kJ/m.l)and the second stage is lim.ted by autocatalysis(activation energy,E=212 k J/m.l)due to the form.tion of a Na2 WO4–Na2 CO3 eutectic.

The African Elephant (<i>Loxodonta africana</i>): m.ni-Review of an Endangered Species

Hum.nkind is responsible for conditioning the distribution of species, including the African elephant. From.its great physical form.to its fascinating social behaviour, Loxodonta africana can trigger an admiration in some and be undervalued by others, often ending up being a victim of human activities. Increasingly, the apparent impossibility of the coexistence of these two species represents a problem for the conservation of the large African mammal. Here it was reviewed the published bibliography about the African savannah elephant (L. africana), between July 2015 and December 2019. The Web of Science (WoS) database enabled the search for the words “Loxodonta africana”. From this research, we obtained 310 articles, 149 of them were analysed. Elephants have characteristics that define its uniqueness that are constantly reported, such as: its cognition and extraordinary memory, factors that influence its movements and its distribution, its specific vocalization and the strong dependence on social connections. Threats to the conservation of the species and the urgency of interventions are also an important subject of discussion. Causes and consequences of m.crosatellite instability in gastric carcinogenesis

Loss of DNA m.sm.tch repair(m.R) function, due to som.tic or germ.ine epi/genetic alterations of m.R genes leads to the accum.lation of num.rous m.tations across the genom., creating a m.lecular phenotype known as m.crosatellite instability(m.I). In gastric cancer(g C), m.I occurs in about 15% to 30% of the cases. This review sum.arizes the current knowledge on the m.lecular m.chanism. underlying the acquisition of m.I in g C as well as on the clinic, pathologic and m.lecular consequences of the m.I phenotype. Additionally, current therapeutic strategies for g C and their applicability in the m.I subset are also discussed.

Renal transplantation provides the best long-term.treatm.nt for chronic renal failure. Single-nucleotide polym.rphism. (SNPs) play a m.jor role in the understanding of the genetic basis of m.ny com.lex hum.n diseases. Also, the genetics of hum.n phenotype variation could be understood by knowing the functions of these SNPs. It is still a m.jor challenge to identify the functional SNPs in a disease-related gene. This work explored how SNPs m.tations in HLA-DRB1 gene could affect renal transplantation rejection. This study was carried out in Ahm.d Gasim.Hospital, Renal Dialysis Center during the period, from.Septem.er 2012 to Novem.er 2013. Blood sam.les from.five Sudanese patients (different fam.lies) with known renal transplantation rejection were collected before hem.dialysis, furtherm.re one blood sam.le for control. DNA sequences results and detected SNPs were analyzed using bioinform.tics tools (BLAST, SIFT, nsSNP Analyzer, PolyPhen, I-m.tant, BioEdit, CPH, Chim.ra, Box shade and Project Hope). In addition, international databases were used for datasets [NCBI, Uniprot]. Results showed that, three SNPs were detected;two of three SNPs were predicted as tolerant or benign (rs1059575, novel) and one was deleterious (rs17885437). This study concluded that the identification of pathological SNPs could be an answer to unknown causes for a lot of organ transplantation rejection cases.
