Low-Density Co-Inoculation of Myanmar <i>Bradyrhizobium yuanmingense</i>MAS34 and <i>Streptomyces griseoflavus</i>P4 to Enhance Symbiosis and Seed Yield in Soybean Varieties

This study examined whether low-density co-inoculation of Myanmar Bradyrhizobium yuanmingense strain MAS34 and Streptomyces griseoflavus P4 would enhance nodulation, N2 fixation, and seed yield in two soybean varieties. A field experiment was conducted during the July to November 2012 growing season at Kyushu University Farm, Japan, using a split-plot design with three replications and the following four treatments: T1, an uninoculated treatment with peat moss (uninoculated);T2, a single inoculation with S. griseoflavus P4 (P4);T3, a single inoculation of B. yuanmingense MAS34 (MAS34);and T4, a dual inoculation of P4 with MAS34 (P4 + MAS34). Two varieties of soybean, Yezin-3 (Rj4) and Yezin-6 (non-Rj), were used. The N2 fixation activity of soybean was evaluated by the relative ureide method using xylem solute from root bled sap at the early pod-fill stage (R3.5). Dry matter production, N2 fixation, and seed yield were significantly (P 0.01) different between the inoculated treatments. The effect of variety was also significant (P 0.05) for nodule dry weight at the V6 stage, percentage of N derived from the atmosphere at the R3.5 stage, and seed yield at the maturity stage. The number of nodules on the tap roots was significantly higher in Yezin-3 than in Yezin-6. The single inoculation of P4 did not have a significant effect on dry matter production, N2 fixation, and seed yield in either soybean variety. The dry matter production, relative ureide index, percentage of N derived from the atmosphere, and seed yield were significantly (P 0.01) enhanced by a single inoculation of MAS34 in Yezin-3 and by dual inoculation of P4 + MAS34 in Yezin-6. These results indicate that low inoculum concentrations (105 cells seed-1) increase N2 fixation and seed yield in these soybean varieties under open field conditions. Myanmar B. yuanmingense MAS34 and S. griseoflavus P4 are expected to be useful biofertilizers for soybean production.

Assessment of Farmer’s Practices on the Rubber Nursery Management Using Growing Medium and Varietal Selection in Mon State

Rubber is an economically important perennial crop in Myanmar for latex production. As the rubber plantation area increases yearly, the requirement of vigorous rubber seedlings for its establishment plays a central role. The success of rubber plantations depends on some farming practices such as using different compost or other materials in the potting soil-medium, varietal selection for stock and scion in the budding process, and fertilizer application methods. The objective of this study was to assess the farmer’s practices in the establishment of rubber nurseries in mostly rubber planted areas in Myanmar. The survey interviewed 60 respondents from three townships in Mon State, namely Mudon, Kyaikmaraw, and Thanbyuzayat. The response data were analyzed through the descriptive method. This survey exposed the potentially active operators (middle age of 30 - 60 years) in rubber nursery production. Local experienced farmers usually raised the budded seedlings with 15 cm × 23 cm polyethylene bag in all study regions. Most farmers selected multi-clonal seed for stock and BPM 24 for scion according to the local market demand and high latex yield. All survey areas used both organic and inorganic fertilizers for nutrient management. Compost is a chief component of growing medium in their nursery production. However, making compost and high demand of the compost were local constraints. Therefore, this survey suggested improving the proper composting method for rubber nursery establishment.

Trachypithecus,which currently contains 20 species divided into four groups,is the most speciose and geographically dispersed genus among Asian *** several morphological and molecular studies,however,its evolutionary history and phylogeography remain poorly ***’s langur(Trachypithecus phayrei) is one of the most widespread members of the genus,but details on its actual distribution and intraspecific taxonomy are limited and ***,to elucidate the evolutionary history of Trachypithecus and to clarify the intraspecific taxonomy and distribution of ***,we sequenced 41 mitochondrial genomes from georeferenced fecal samples and museum specimens,including two *** analyses revealed a robustly supported phylogeny of Trachypithecus,suggesting that the *** group branched first,followed by the *** group,and the *** and *** groups most *** four species groups diverged from each other 4.5-3.1 million years ago(Ma),while speciation events within these groups occurred much more recently(1.6-0.3 Ma).within ***,we found three clades that diverged 1.0-0.9 Ma,indicating the existence of three rather than two *** the phylogenetic species concept and based on genetic,morphological,and ecological differences,we elevate the *** subspecies to species level,describe a new species from central Myanmar,and refine the distribution of the three ***,our study highlights the importance of museum specimens and provides new insights not only into the evolutionary history of *** but the entire Trachypithecus genus as well.

An Adaptive Response Compensation Technique for the Constant-Current Hot-wire Anemometer

An adaptive response compensation technique has been proposed to compensate for the response lag of the constant-current hot-wire anemometer (CCA) by taking advantage of digital signal processing technology. First, we have developed a simple response compensation scheme based on a precise theoretical expression for the frequency response of the CCA (Kaifuku et al. 2010, 2011), and verified its effectiveness experimentally for hot-wires of 5 μm, 10 μm and 20 μm in diameter. Then, another novel technique based on a two-sensor probe technique—originally developed for the response compensation of fine-wire thermocouples (Tagawa and Ohta 1997;Tagawa et al. 1998)—has been proposed for estimating thermal time-constants of hot-wires to realize the in-situ response compensation of the CCA. To demonstrate the usefulness of the CCA, we have applied the response compensation schemes to multipoint velocity measure- ment of a turbulent wake flow formed behind a circular cylinder by using a CCA probe consisting of 16 hot-wires, which were driven simultaneously by a very simple constant-current circuit. As a result, the proposed response compensation techniques for the CCA work quite successfully and are capable of improving the response speed of the CCA to obtain reliable measurements comparable to those by the commercially-available constant-temperature hot-wire anemometer (CTA).

为明确棉铃虫Helicoverpa armigera丝氨酸蛋白酶抑制剂(serine protease inhibitor,serpin)的种类及其表达特性,利用PCR技术克隆棉铃虫的serpin基因,使用生物信息学软件预测其结构并进行系统进化分析,采用实时荧光定量PCR(real time quantitative PCR,RT-qPCR)技术比较serpin基因在棉铃虫不同发育阶段和组织中的表达量及取食Cry1Ac后其表达量的变化。结果表明,共获得serpin-a、serpin-b、serpin-c、serpin-e四个棉铃虫serpin基因,全长为1119~1254 bp,编码373~418个氨基酸,均包含一段具有反应中心环的保守结构域,且与斜纹夜蛾Spodoptera litura、草地贪夜蛾***等鳞翅目昆虫serpin的同源性较高。serpin-a和serpin-e在棉铃虫4龄幼虫期的表达量最高,serpin-b和serpin-c分别在成虫期和蛹期表达量最高。serpin-a在中肠和围食膜中表达量最高,serpin-b在头、中肠和表皮中表达量最高,serpin-c在头部表达量最高,serpin-e在中肠和血淋巴中的表达量显著高于其他组织。棉铃虫取食低浓度Cry1Ac后,中肠的serpin-b和serpin-e的表达量显著增加。推测不同serpin基因在棉铃虫不同发育时期和组织中可能发挥不同的作用,其中serpin-b和serpin-e可能参与棉铃虫对Cry1Ac的解毒过程。

自1996年转基因作物商业化生产以来,表达苏云金芽孢杆菌杀虫蛋白的转基因棉花有效地控制了主要靶标害虫。但是,随着转基因棉花种植后棉田杀虫剂使用量的下降,绿盲蝽(Apolygus lucorum)(Hemiptera:Miridae)等次要害虫的种群数量上升。另一主要原因是当前种植的Bt作物对刺吸式害虫几乎没有毒性。最近有报道,属于β-成孔蛋白家族的Bt Cry51Aa1和Cry51Aa2蛋白对盲蝽具有的潜在毒性,Cry51Aa1序列修饰的Cry51Aa2突变体(Cry51Aa2.834＿16)对西部牧草盲蝽Lygus Hesperus和美国牧草盲蝽Lygus lineolaris具有很高的毒性。本研究中,我们测定了Cry51Aa1和Cry51Aa2对绿盲蝽***的杀虫活性,并对杀虫机制进行了初步研究。我们通过生物测定检测了Cry51Aa1和Cry51Aa2蛋白对绿盲蝽若虫的毒性,测定了绿盲蝽中肠匀浆液、中肠酶液、胰蛋白酶或胰凝乳蛋白酶对Cry51Aa1和Cry51Aa2原毒素的活化,利用ELISA法测定了绿盲蝽中肠蛋白与Cry51Aa1和Cry51Aa2的结合能力,通过Ligand blot法明确了Cry51Aa1和Cry51Aa2原毒素和活化毒素与绿盲蝽中肠受体蛋白的结合,并利用配体垂钓和LC-MS/MS进一步分析了可能的受体蛋白种类。主要结果如下:Cry51Aa1原毒素蛋白是在携带全长基因的重组Bt菌株BMB171表达获得,。通过离子交换层析法纯化产生的Bt原毒素蛋白,纯化后的Cry51Aa1原毒素蛋白大约35 k Da。具有His-tag的重组Cry51Aa2在p ET30a＿51Aa2质粒构建的载体转化至BL21(DE3)感受态细胞中表达,表达的重组蛋白用Ni柱纯化,利用肠激酶消化切去重组Cry51Aa2蛋白N末端的His标签区域,再次用离子交换层析法纯化,纯化的Cry51Aa2毒素约为35 k Da。1.利用混合饲料法进行生物测定,结果表明,绿盲蝽若虫对Cry51Aa1和Cry51Aa2毒素都很敏感,且Cry51Aa1(LC=11.87μgml)原毒素毒性高于Cry51Aa2原毒素(LC=61.34μgml)。***51Aa1和Cry51Aa2毒素都能够被胰蛋白酶和胰凝乳蛋白酶活化,活化的Cry51Aa1和Cry51Aa2毒素大小为25 k Da左右。当毒素与绿盲蝽中肠匀浆液或中肠酶液一起孵育后,用western blot进行检测,也得到了相同的结果。此外,我们发现了60 k Da的Cry51Aa1和Cry51Aa2毒素的自二聚体。绿盲蝽若虫取食亚致死剂量的原毒素1小时后,间隔不同时间收集中肠,通过蛋白免疫印迹(western blot)分析原毒素在体内的活化进程,结果发现绿盲蝽取食原毒素1小时,在3小时和6小时后取样,Cry51Aa1和Cry51Aa2原毒素部分被活化,同时有原毒素、预活化和活化毒素三种形式;在12、24或48小时后取样,Cry51Aa1和Cry51Aa2原毒素被完全活化。***分析结果表明,绿盲蝽若虫中肠蛋白能够与Cry51Aa1和Cry51Aa2毒素结合,结合能力与浓度有关。当使用100 n M和200 n M的Cry51Aa1时,结合能力随着中肠浓度的增加而增加,并在中肠蛋白20μg时结合能力达到最高。但是,当Cry51Aa125n M和50 n M时,与中肠蛋白的结合能力较低。所检测的Cry51Aa2,浓度为25、50、100和200 n M时,随中肠蛋白浓度的增加结合能力增加,与20μg和30μg中肠蛋白结合能力最高。20μg中肠蛋白与Cry51Aa1特异性结合的解离常数(kd)为248.6±47.4n M,与Cry51Aa2的解离常数为113.8±25.4 n M。*** blot分析结果显示,原毒素和活化的Cry51Aa1毒素主要与绿盲蝽中肠上分子大小约25 k Da的蛋白结合。与Cry51Aa2原毒素和活化的毒素与中肠蛋白的结合主要也显示出相同的条带,而且与活化的Cry51Aa2还与一个约50 k Da的蛋白结合。5.通过pull-down分析能够结合Cry51Aa1和Cry51Aa2原毒素的绿盲蝽中肠蛋白。切下在SDS-PAGE检测中结合的蛋白条带,并通过LC-MS/MS分析鉴定。在所有鉴定出的蛋白中,与两种毒素都结合的共有15种质膜蛋白,包括ABCA3、ABCB6、ABCC7、ABCD1、ABCG1、ABCG4(86.8 k Da)、ABCF1和一种氨肽酶等蛋白;还鉴定到分别与Cry51Aa1和Cry51Aa2特异性结合的质膜蛋白各24个,其中与Cry51Aa2结合的片段中包含两个氨肽酶和ABCF2蛋白等,与Cry51Aa2结合的片段中含ABCB

危险货物运输,尤其是原油,通过船舶从一个地方转运至别处,对全球经济的发展至关重要。与此同时,由于自然灾害、人为等因素,各主要海域的船舶水运事故频率大幅上升,这其中就包括船舶碰撞。油轮发生碰撞事故,首要的威胁是船员的人身财产安全,并有可能造成当地环境污染。本文主要通过国际海事组织(IMO)的正式安全评估(FSA),对缅甸皎漂深水港内的双层壳油轮碰撞事故进行定量风险评估,利用ANSYS 14.0软件的显示动力学模块,通过碰撞模型,模拟各型油轮(VLCC、苏伊士型、阿芙拉型、巴拿马型等)真实尺寸的碰撞,预测碰撞事故对船体结构损害情况,并分析原油泄漏入海可能的量,然后根据当地油轮类型、船舶流量、水文等数据,以及相关历史事故统计,分析油轮发生碰撞的频率、污染物泄漏频率、船体损害频率和船员死亡频率等。油轮碰撞事故后果分析同样重要,本文通过蒙特卡洛模拟,定量化的分析十万次的随机变量事件,尽可能的将事故后果造成的环境损害、社会和经济损害,用货币损失的方式呈现出来,直观感受某次油轮事故的危害程度。统计每个可能情境的风险后,分析总的风险潜在人命损失(PLL)、潜在污染损失(PLC)和潜在财产损失(PLP),并得到三种风险对应的FN曲线、FT曲线和FP曲线。本文研究发现缅甸皎漂深水海港油轮碰撞事故的社会风险、环境风险和财产损失风险的最大可接受风险水平分别是死亡数1.00E-03个/年、泄漏量0.9E-02吨/年和1.2E+00 MUSD/年。最后,本文旨在将IMO的正式安全评估(FSA)发展为一个简化方法,快速预评某一海域发生油轮碰撞事故的风险标准。

Implementation of Capacity Development and Public Awareness for CTBT Verification Regime in Myanmar

Myanmar signed the Comprehensive Nuclear-Test-Ban Treaty(CTBT)in 1996 and ratified it in *** a CTBT ratifying state,Myanmar needs to implement the capability development and public awareness concerning with CTBT verification *** implementing the CTBT verification regime,Myanmar considered the linkage between CTBT and sustainable development goals(SDGs).A National Data Centre(NDC)is needed in the country for collecting the data from the stations,differentiating the data quality,prioritizing and storage of the data to conduct with International Data Centre(IDC)of Comprehensive Nuclear-Test-Ban Treaty Organization(CTBTO).The seismic,hydroacoustic,infrasound and radionuclide technologies are important for identifying the signals generated by a nuclear *** capacity development and public awareness are important for initial step before the ratification process of the state *** society,change school curriculum,participation in process,organizing workshops and seminars are carried out by policy makers’*** collaboration and citizen awareness are needed to understand the CTBT verification system for monitoring the nuclear explosions in a global *** Atomic Energy Division(AED)will collaborate with external funding agents like CTBTO to improve skill of human resource for NDC implementation in *** scientists,science administrators and representative will be achieved from this national development *** is focused on research and development in areas potentially relevant to the Treaty’s verification regime.
