Assessment of phytochemicals, antioxidant, anti-hemolytic, anti-inflammatory and anti-cancer potential of flowers, leaves and stem extracts of Rosa arvensis

OBJECTIVE: To evaluate phytochemicals and in vitro biological potential of flowers, leaves and stem extracts of Rosa arvensis. METHODS: Presence of twenty secondary metabolites was confirmed and then phenolic and flavonoid contents were quantified spectrophotometrically. Fourier Transform Infrared spectroscopy was conducted to ascertain functional groups and antioxidant potential was examined using 2,2-diphenyl-1-picrylhydrazyl scavenging activity, total antioxidant capacity and total reducing power assays.Human erythrocytes were used to assess anti-hemolytic activity and five bacterial strains were examined to determine antibacterial potential of plant extracts. Radish seeds were used to perform phytotoxic activity and cytotoxic potential was evaluated via brine shrimps and PC3 cell lines. RESULTS: Highest phenolic contents were detected in the methanolic extract of Rosa arvensis flower（RAFM） [（151.635 ± 0.005） gallic acid equivalent mg/g] and highest flavonoid contents in the chloroform leaf extract（RALC） [（108.228 ± 0.004） quercetin equivalent mg/g]. Fourier-transform infrared spectroscopy analysis showed the presence of wide range of functional groups. The antioxidant assays indicated highest DPPH scavenging activity [IC50（23.5 ± 0.6） μg/mL] in the methanolic stem extract（RASM）, highest total antioxidant capacity [（265.1 ± 0.9） μg/mL] in RAFM and highest reducing potential [（209.9 ± 0.6） μg/mL] in leaf extract（RALM）. Highest antihemolytic activity [（90.0 ± 0.5） μg/mL] was recorded in RAFM and brine shrimp cytotoxicity potential [（52.3 ± 0.3） μg/mL] in RASM. The antimicrobial activity was detected highest [（21.1 ± 0.5） mm inhibition zones] in RALM against Streptococcus aureus. In the end, antiinflammatory and anti-cancer activity results depicted less than 50 % inhibition in the methanolic extracts. CONCLUSIONS: Our findings will be helpful in designing pharmaceutical regimens and therefore, more studies can be recommended to isolate and characterize compounds associated with the biological activities of Rosa

Phytochemical investigation of medicinally important plants of the Pothohar region of Pakistan

OBJECTIVE:To report the phytochemical composition(flavonoids,phenolics,saponins,tannins,and terpenoids)and biological activities(antioxidant,phytotoxic,cytotoxic,antifungal activities)of medicinally important plants of Pothohar region.METHODS:In the current study various qualitative and quantitative phytochemical tests were conducted to investigate the phytochemical composition of plant extracts prepared in benzene,chloroform and methanol.The same extracts were further utilized in different biological activities i.e.,antioxidant[total antioxidant capacity,total reducing power,and 2,2-diphenyl-1-picrylhydrazyl(DPPH)scavenging assay],phytotoxicity(Radish seed bioassay),brine shrimp cytotoxicity and antifungal activities(against four pathogenic fungal strains).RESULTS:Phytochemical analysis indicate flavonoids,phenolics,saponins,tannins and terpenoids were present in almost all plants however,maximum amount of total phenolic and flavonoid contents was reported in of Malva parviflora and Lepidium pinnatifidum.Extracts of Malva parviflora and Lepidium pinnatifidum also depicted the superlative antioxidant activity(total antioxidant capacity,total reducing power,and DPPH scavenging activity).Maximum fungal growth inhibition was demonstrated by extracts of Centaurea calcitropa,Vicia sativa and Malva parviflora.The maximum cytotoxicity was shown by the extracts of Centaurea calcitropa.Moreover,for phytotoxicity assay the highest allelopathic potential was exhibited by extracts of Lepidium pinnatifidum.Solvents prejudiced results;the best results were obtained by Methanol followed by Chloroform and Benzene.CONCLUSION:Centaurea calcitropa,Vicia sativa,Malva parviflora and Lepidium pinnatifidum are most promising medicinal plants of Pothohar region for future studies.

Microbiostatic, antioxidative and cytotoxic potentiation of some grasses of Bahawalpur, Pakistan

OBJECTIVE: To evaluate biological potential of methanol and n-hexane extracts of aerial parts in seven species of family Poaceae.METHODS: Qualitative phytochemical tests were done by using standard protocols. In vitro antioxidant activity was performed via different assays and antimicrobial potential was observed via disc diffusion method. Cytotoxic activity was carried out using brine shrimps’ assay.RESULTS: Phytochemical studies revealed the presence of alkaloids, flavonoids, glycosides, phenols,steroids, saponins, tannins, anthocyanins and coumarins in most of the plant extracts. Maximum antioxidant and antimicrobial potential were observed in Cymbopogon citratus methanol extract and Cymbopogon citratus n-hexane extract along with significant number of total flavonoids and phenols contents. However, Polypogon monspeliensis methanol extract and Polypogon monspeliensis n-hexane extract showed minimum antioxidant as well as antimicrobial potential. Moreover, methanol extracts showed a cytotoxic effect with their effectiveness ranked Polypogon monspeliensis methanol extract > Cymbopogon citratus methanol extract > Phalaris minor n-hexane extract > Aristida funiculata methanol extract > Stipagrostis plumosa methanol extract > Cenchrus ciliaris methanol extract > Panicum antidotale methanol extract. Similarly, n-hexane plant extracts revealed cytotoxic activity in decreasing order Cenchrus ciliaris n-hexane extract > Stipagrostis plumosa n-hexane extract > Phalaris minor n-hexane extract > Aristida funiculata n-hexane extract > Polypogon monspeliensis n-hexane extract > Panicum antidotale n-hexane extract > Cymbopogon citratus n-hexane extract respectively.CONCLUSION: Methanol extracts exhibit significant antioxidant and antimicrobial potential which can be correlated to their medicinal values. The observed brine shrimp’s lethality of the plant extracts revealed the presence of potent cytotoxic components in these plants.

Biological evaluation of some important medicinal plants from Poonch valley, Azad Kashmir, Pakistan

OBJECTIVE:To estimate the phytochemical composition,antioxidant,antifungal,cytotoxic and phytotoxic activities of three important medicinal plants of Poonch valley,Azad Kashmir Pakistan.METHODS:Phytochemicals were investigated for both qualitative(phenols,flavonoids,alkaloids and saponins)and quantitative(total phenolic and flavonoid contents)analysis of plant extracts prepared in ethanol,ethyl acetate and benzene.Moreover,antioxidant potential was investigated by screening botanical extracts against 2,2-diphenyl-1-picrylhydrazyl scavenging assay,phosphomolybdenum and total reducing power assay.While,disc diffusion method was used for determination of antifungal potential of all extracts against four pathogenic strains.Furthermore,brine shrimp cytotoxicity assay was conducted for cytotoxic potential and radish seed germination assay was carried out for estimation of allelopathic potential of plant extracts.RESULTS:Phytochemical evaluation showed the presence of phenolics,flavonoids,alkaloids and saponins in almost all plants.In addition to significant antioxidant potential,Rubia cordifolia(ethanolic extract)revealed highest amount of phenolics and flavonoids.The said plant also exhibited good cytotoxic activity.Among plant extracts highest antifungal activity and allelopathic potential was recorded by Oxalis latifolia(ethyl acetate extract).Ethanol and ethyl acetate were observed to be the best choice of solvents.The best solvents-based results were obtained by Ethanol followed by Ethyl acetate and Benzene.CONCLUSION:For further studies,Rubia cordifolia and Oxalis latifolia are the most promising medicinal plants of Poonch valley,Azad Kashmir Pakistan at molecular level in order to isolate the compounds concerning with pronounced activities.

Plant-derived anticancer agents: A green anticancer approach

Cancer is a frightful disease and represents one of the biggest health-care issues for the human race and demands a proactive strategy for cure. Plants are reservoirs for novel chemical entities and provide a promising line for research on cancer. Hitherto, being effective, chemotherapy is accompanied by certain unbearable side effects. Nevertheless,plants and plant derived products is a revolutionizing field as these are Simple, safer, ecofriendly, low-cost, fast, and less toxic as compared with conventional treatment methods.Phytochemicals are selective in their functions and acts specifically on tumor cells without affecting normal cells. Carcinogenesis is complex phenomena that involves many signaling cascades. Phytochemicals are considered suitable candidates for anticancer drug development due to their pleiotropic actions on target events with multiple manners. The research is in progress for developing potential candidates(those can block or slow down the growth of cancer cells without any side effects) from these phytochemicals. Many phytochemicals and their derived analogs have been identified as potential candidates for anticancer therapy. Effort has been made through this comprehensive review to highlight the recent developments and milestones achieved in cancer therapies using phytomolecules with their mechanism of action on nuclear and cellular factors. Furthermore, drugs for cancer treatment and their limitations have also been discussed.

Role of dietary phytochemicals in modulation of miRNA expression:Natural swords combating breast cancer

The National Cancer Institute had projected breast cancer(BC) as one of the topmost prevalent malignancies around the globe.In many cases,BC becomes resistant to chemotherapy,radiation and hormonal therapies.Traditional BC therapies are associated with adverse side effects,drug resistance and recurrence.Extensive research work has shown that these dietary phytochemicals(DPs) may exert therapeutic effects by regulating the miRNA expression.A large number of DPs have been researched as miRNA regulatory agents against BC and some other DPs have not yet been tested against BC.We have discussed the effects of curcumin,diallyl disulphide,3,3′ diindolylmethane,ellagic acid,genistein,indole-3-carbinol,quercetin,resveratrol,and sulforaphane on regulation of expression of BC miRNAs in a wide range of in vitro and in vivo models.We have also shown some of the possible DPs(Oleanolic acid,capsaicin,benzyl isothiocyanate,epigallocatechin gallate,phenethyl isothiocyanate and ursolic acid) that have shown miRNA regulatory activities and have not yet been tested against BC miRNAs.Finally,current limitations,challenges,future perspectives of DPs and BC research are also critically discussed.

Dietary isoflavones,the modulator of breast carcinogenesis:Current landscape and future perspectives

Breast cancer is a frightful disease and serious concern in women around the world causing significant health care burden in both developed and developing countries. Extensive research work has shown that breast cancer provides strong resistance to chemical agents, U V radiation,and hormonal treatments. It is generally accepted that cell genetics is not the only main reason for breast cancer and genetic risk factors, for example, mutations in RRCAI and BRCA2 genes constitute 5%-10% of all breast cancer rates. Other related factors include age, gender,race, ethnicity, weight, reproductive factors, exo-and endogenous hormonal exposures, oral contraceptives use, ultraviolet radiation, diet, and night work(circadian disruption). Many studies have revealed that dietary isoflavones regulate breast cancer occurrence, recurrence and prognosis. Dietary isoflavones have long been part of Asian population diet and there is a significant increase as compared to dietary isoflavones intake among other populations. Dietary isoflavones are natural phytoestrogens having both estrogenic and anti-estrogenic potentials on breast cancer cells in culture, animal models and in experimental trials. This literature survey provides a comprehensive overview on the tumor preventive and tumor promoting potentials of dietary isoflavones on breast cancer. In addition, this paper provides a literature review of dietary isoflavones and their effects on up-regulation and down-regulation of different signaling pathways, genes and proteins. Finally, future perspectives of dietary isoflavones and breast cancer researchers are also critically discussed, which will provide a deeper insight regarding the inner molecular mechanisms of action.

Glinus lotoides ethanolic extract alleviates LPS-induced anxiety and depression-like behavior by modulating antioxidant and inflammatory biomarkers in rats

Objective:To explore the protective role of Glinus lotoides ethanolic extract in a depression model through modulating oxidant/antioxidant enzyme system and inflammatory status.Methods:Phytochemical constituents of Glinus lotoides ethanolic extract were evaluated qualitatively and quantitatively along with HPLC.Rats were divided into six groups.The normal control and the intoxicated groups received normal saline,and the standard group received imipramine,while the remaining groups received 100,300,and 500 mg/kg Glinus lotoides ethanolic extract.All groups received treatments for 14 d.Lipopolysaccharides(LPS)were then administered i.p.(0.83 mg/kg)to all groups except the normal control group.After 24 h,anxiety and depression-like behaviors were evaluated by performing behavioral analysis(open field,tail suspension,forced swim,sucrose preference test),and determining total oxidant status,total antioxidant capacity,catalase,and biochemical parameters[malondialdehyde,glutathione,superoxide dismutase,tumor necrosis factor(TNF)-alpha and interleukin(IL)-6].Results:Phytochemical studies confirmed the presence of phenols and flavonoids and HPLC analysis showed the presence of gallic acid,quercetin,chlorogenic,and caffeic acid.Total oxidant status was significantly decreased,while total antioxidant capacity was significantly increased in the Glinus lotoides ethanolic extract treated groups.Moreover,Glinus lotoides ethanolic extract diminished malondialdehyde,IL-6,and TNF-alpha levels,while increasing superoxide dismutase,catalase,and glutathione activities.Conclusions:Glinus lotoides ethanolic crude extract shows significant antidepressant activity by modulating oxidative and biochemical parameters that supports its folkloric use in traditional systems of medicine.

Locally Advanced Breast Cancer in Pakistani Women: What Is Different from Rest of the World and Why It Is Difficult to Manage

Locally Advance Breast Cancer refers to a heterogeneous group of breast cancer with locally extensive disease, which may or may not involve the nodes, without any distant metastases. The study was conducted at Faisalabad Medical University (FMU), Oncology, Allied Hospital Faisalabad (Pakistan). Data of 100 patients with LABC was collected. Demographics were recorded in the form of age, socio-economic status. In clinical data, time of presentation, family history of breast cancer, the presenting symptom in the form of lump, ulceration and other skin changes were noted. Histo-pathological variables including tumor size, histopathology, Bloom & Richardson grading, estrogen receptor (ER), progesterone receptor status (PR) and HER2 status. Results showed that after following a standard trimodality treatment approach in LABC patients, 30 percent died within two years. Disease free survival for more than two years was observed in only 25% of patients. Whereas, 70% patients had eventful (Recurrence/metastases) survival. This poor outcome was observed due to lack of health care facilities, awareness and poor socioeconomic status.
