Biochemical Estimation of Glucose 6 Phosphate Dehydrogenase Deficiency in Saudi Adults: Different methods and Its Rationalization

Background: Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the most common human enzymopathy affecting 400 million people, globally. G6PD deficiency is an X-linked genetic condition, which is more likely to affect males than females. Heterozygous females go undetected in a commonly used method. The aim of the study was to identify & rationalize different biochemical methods for detections of G6PD deficiency. methods: Cross section retrospective study was conducted on 1584 (800 males, 784 females) blood samples collected from King Abdulaziz University Hospital (KAUH) and King Fahd Armed force hospital (KFAFH) in Jeddah, Western Saudi Arabia. Blood samples were screened for G6PD activity by fluorescence spot test, semi quantitative color reduction test and spectrometric quantitative evaluation. Hemoglobin (Hb) was measured on the same sample by BC-3200 Auto hematology Analyser. G6PD activity was recorded as U/g Hb. Samples identified as deficient with cutoff ≤4.6 U/gHb. Results: The prevalence of G6PD deficiency identified by fluorescence spot test was 73(4.6%) and all were deficient male. By semi quantitative method, the prevalence rate was 51(3.2%) and again all were male deficit patients. However, when quantitative spectrometric method was used, the prevalence was found in 90(5.7%), where in 73(4.6%) deficient patients were males and 17(1.1%) were females. Conclusion: Since the fluorescence spot test did not miss any G6PD deficient male, it should be restricted to males and quantitative test should be done on females. Each ethnic group should cultivate their own cutoff value for categorization of deficient patients.

This study investigated the feasibility of using some natural clay minerals available in Egypt like kaolinite, bentonite, also using of iron oxide, rock phosphate and mixture of these types to minimize the rate of Ni, Cu, Zn and mn desorption from sewage soils and to evaluate the effect of these mixtures on Zn equivalent constant values before and after remediation. The obtained results indicated that all mixtures used were minimizing the rate of potential toxic elements （PTE＇s） release from sewage soils compared to control treatment. According to the decreasing order of different treatments, data indicated that application of the mixture treatment of bentonite, kaolinite and rock phosphate （RP） in sewage soils becomes the best treatment compared to other treatments used. However, the lowest and save Zn equivalent constant value which represents the hazard indicator in the study was observed in sewage soil treated with the mixture of bentonite and RP. The kinetic constants of Elovich, modified Freundlich and Hoerl equations, the best fitted models, were significantly decreased compared to control treatments with different percent of minimization according to type of pollutants and remediation material used. To understand the mechanisms of PTE＇s retained in treated sewage soil, distribution study was applied which showed that different pollutants studied were removed to hardly available form different mechanisms of PTE＇s undergo in treated sewage soils were discussed in the study.

Radionuclides naturally occurring in building materials may significantly contribute to the annual doses to the public. For instance, familiar building materials such as the concrete and gypsum board have been reported to produce a dose of about 0.04 mSv per year for a typical person (NCRP 1987c). External as well as internal exposures are two pathways of radiation dose imparted to human beings from the building materials. As information on the radioactivity of such materials is lacking, the study of gypsum materials used in Egypt was carried out in order to estimate the annual dose to the Egyptian population due to natural radionuclides in building materials. During the study, 18 samples of commonly used gypsum raw materials were collected and measured. The activity concentrations were determined by gamma ray spectrometry. Their mean values were in the ranges of 499.29 ± 11.53 Bq·kg-1 for 40K, 91.97 ± 2.61 Bq·kg-1 for 226Ra, 37.62 ± 1.67 Bq·kg-1 for 238U and 42.27 ± 2.22 Bq·kg-1 for 232Th. The activity indexed Iγ for 18, different gypsum samples varied from 0.31 ± 0.03 to 2.3 ± 0.19 and the radium equivalent activity (Raeq), from 38.81 ± 1.68 to 324.7 ± 9.42. These values are lower than the limit of 370 Bq·kg-1 adopted by the Organization for Economic Cooperation and Development (OECD). The average indoor annual effective gamma dose rate (DE) in (mSv/y) for the people, caused by the building materials of dwellings, was assessed for most commonly gypsum materials. It was estimated to be in the range from 0.10 ± 0.003 mSv/y to 0.74 ± 0.08 mSv/y. The internal and external hazard indices (Hin, Hex) and the absorbed dose rate in air D in each sample were evaluated to assess the radiation hazard for people living in dwelling made of the studied materials. The absorbed dose rate of indoor air in samples G1, G2, G11, G17 and G18 is less than the international recommended value which is 55 nGyh-1. Zearalenone (ZEN) is a non steroidal estrogenic mycotoxin produced by Fusarium species of fungi which contaminate human foods and animal feeds worldwide. In this study hepatotoxicity of ZEN was evaluated in mice by oral adminis-tration of single and repeated doses of ZEN mycotoxin (2.7 mg/kg b.w.). The protective effect of crude venom extracted from jellyfish Cassiopea andromeda was also assessed. mice were divided into four groups (N = 10). G1: receiving the toxin once and sacrificed 48 h later, G2: toxin administered twice for one week, G3: toxin administered twice a week for two weeks, G4: pretreated orally by a single dose of crude venom (1.78 mg/20g) 24 hours prior to administration of ZEN twice a week for two weeks. Each treated group had its corresponding control which received 1% DmSO *** treatment significantly increased alanine aminotransferase (ALT), aspartateaminotrnsferase (AST) and alka-line phosphatase (ALP) activities after 48 hours and two weeks, while ALT was also significantly increased after one week. Tumor necrosis factoralpha (TNF-α) level was undetected in treated and control groups except the group treated with ZEN for one week. Alphafetoprotein (AFP) level was increased significantly only after two weeks. The activity of antioxidants was significantly increased in all groups. ZEN was also found to modify the serum proteins especially gamma-globulin which showed a significant decrease after 48 h and two weeks. Improvement in liver func-tion occurred in the group pretreated with the crude venom, and AFP and antioxidants returned to normal level, while TNF-α level was also undetected. Gamma globulin was significantly increased. The recovery observed in the group which was pretreated with crude venom may related to bradykinin content of this venom which exhibits a hepatoprotective effect. 在离体条件下测试了81株芽孢杆菌分离物抑制番茄青枯病菌的能力,有4个菌株(B2、B5、B7和BS)显示了较好的抑菌效果,经细菌生物学性状、Biolog和16S rDNA序列分析,B2菌株鉴定为短短小芽孢杆菌(Brevibacillus brevis),B5、B7 和B8鉴定为枯草芽孢杆菌(Bacillus subtilis)。在温室环境中,4个菌株都能不同程度的促进灭菌土和自然土的番茄生长, 并能减轻番茄青枯病的发生。B2菌株显示了显著的抑菌作用,在灭菌土和自然土中分别降低了80．0％和87．4％的番茄青枯病发生率。

Yield Attributes and Oil Safety in the Hyperaccumulator Canola Plant Grown in a Bioremediated Sewaged Soil

A field experiment was carried out at Abu-Rawash sewage farm to appraise the effect of certain novel remediative amendments on the quality of oil as well as the vegetative parameters and yield criteria of canola plant used as hyperaccumulator for the remediation of sewaged soils. The treatments included fallow soil （irrigated without growing canola）, soil cultivated with canola （Brassica napus L.） and inoculated with arbuscular mycorrhiza （Am）, soil inoculation with Thiobacillus sp. （a mixture of Thiobacillus ferrooxidans and Thiobacillus thiooxidant）, soil treated with a mixture of 250 mg bentonite plus 250 mg rock phosphate/kg soil and inoculated with phosphate dissolving bacteria （PDB）, and soil treated with all the aforementioned remediative amendments. Results indicated that the vegetative parameters and yield criteria of canola plant did not exhibit any serious adverse impact under all treatments applied. The concentrations of Zn and Cu in canola oil extracted from plants grown in soil inoculated with Am and/or Thiobacillus sp. far exceeded the safe permissible levels. On the other hand, the content of both PTEs in the oil extracted from canola plants grown in soil treated with either probentonite or with mixture of all remediative amendments followed the permissible safe levels.

We present algorithms for computing the differential geometry properties of Frenet apparatus {t,n,b,κ,τ} and higher-order derivatives of intersection curves of implicit and parametric surfaces in R3 for transversal and tangential intersection. This work is considered as a continuation to Ye and maekawa [1]. We obtain a classification of the singularities on the intersection curve. Some examples are given and plotted.

The hepatoprotective activity of the ethanol extract of Astragalus kahiricus(Fabaceae) roots against ethanol-induced liver apoptosis was evaluated and it showed very promising hepatoprotective actions through different *** extract counteracted the ethanol-induced liver enzymes leakage and glutathione *** addition,it demonstrated anti-apoptotic effects against caspase-3 activation and DNA fragmentation that were confirmed by liver histopathological ***,the phytochemical study of this extract led to the isolation of four cycloartane-type triterpenes identified as astrasieversianin II(1),astramembrannin II(2),astrasieversianin XIV(3),and cycloastragenol(4).The structures of these isolates were established by HRESI-mS and 1D and 2D NmR *** antimicrobial,antimalarial,and cytotoxic activities of the isolates were further evaluated,but none of them showed any activity.

Background: The urokinase-type plasminogen activator system which consists of a proteinase (the urokinase-type plasminogen activator, uPA), a receptor (the urokinase-type plasminogen activator receptor uPAR or CD87) and inhibitors (PAI1) is enriched in several types of tumors and involved in proteolysis, cell migration and invasion. High expression of uPA and uPAR is associated with an increased relapse rate and shorter survival in breast cancer and colorectal carcinomas. Aims: This study shed light on the expression of uPAR in adult acute myeloid leukemia (AmL) and its prognostic relevance. methods: Peripheral blood and bone marrow samples are obtained from 54 newly diagnosed AmL adult patients, 20 healthy controls stained with anti CD87 and estimated on flow cytometry. Results: CD87 expression was heterogeneous in different FAB subtypes of AmL with high expression in monocytic leukemia (m4/m5) (P value 0.001). High expression of CD87 was associated with shorter survival and poor response to therapy (P = 0.028, 0.002 respectively). The most discriminating cut off value of CD87 expression was 20%, patients with less than 20% expression had 4.6 times better treatment outcome than those with more than or equal to 20%. On the multivariate analysis, CD87 was the most significant single variable that affect treatment outcome compared to total leucocytic count, hemoglobin level, platelets count, cytogenetic and FLT expression. Summary/Conclusion: High CD87 expression is an independent prognostic parameter associated with poor response and shorter overall survival in adult AmL patients.
