Developmental dynamics of the single nucleus regulatory landscape of pig hippocampus

The hippocampus is a brain region associated with memory,learning and spatial navigation,its aging-related dysfunction is a common sign of Alzheimer’s *** is a good model for human neurodegenerative disease,but our understanding of the regulatory program of the pig hippocampus and its cross-species conservation in humans remains ***,we profiled chromatin accessibility in 33,409 high-quality nuclei and gene expression in 8,122 high-quality nuclei of the pig hippocampus at four postnatal *** identified 510,908 accessible chromatin regions(ACRs)in 12 major cell types,among which progenitor cells such as neuroblasts and oligodendrocyte progenitor cells showed a dynamic decrease from early to later developmental *** revealed significant enrichment of transposable elements in cell type-specific ACRs,particularly in *** identified oligodendrocytes as the most prominent cell type with the greatest number of genes that showed significant changes during the *** identified ACRs and key transcription factors underlying the trajectory of neurogenesis(such as POU3F3 and EGR1)and oligodendrocyte differentiation(RXRA and FOXO6).We examined 27 Alzheimer’s disease-related genes in our data and found that 15 showed cell type-specific activity(TREM2,RIN3 and CLU),and 15 genes displayed age-associated dynamic activity(BIN1,RABEP1 and APOE).We intersected our data with human genome-wide association study results to detect neurological disease-associated cell *** present study provides a single nucleusaccessible chromatin landscape of the pig hippocampus at different developmental stages and is helpful for the exploration of pigs as a biomedical model in human neurodegenerative diseases.

Three discrete tetrahedral metallo-supramolecular cages were designed and constructed using truxenepended base *** to the synergistic rigidifying effect of unsymmetric cyano-substituted oligo(pphenylene-vinylene)(u-COPV)suspended by the truxene skeleton,the resulting supramolecular cages were confirmed to exhibit significant aggregation-induced emission(AIE)accompanied by an interesting solvatochromic fluorescent behavior as well as a porous honeycomb-like state during *** particular,the anti-counterfeiting performance and emission behaviors of the cages in the solid state under external hydrostatic pressure were investigated.

Object traceability based on Radio Frequency Identification(RFID)is an important capability of the Internet of Things(IoT).GS1 EPCglobal,a de-facto standard in RFID technology,develops Electronic Product Code(EPC)network architecture,which mainly includes EPC Information Service(EPCIS),Object Name Service(ONS),and Discovery Service(DS).This architecture is used to capture and share standardized events representing various aspects on EPC ***,the EPC network architecture also faces challenges;for example,the separate management of unrelated event data and master data may increase time consumption when an application extracts valuable information by combing them.A Hardware-Based Information Service(HIS)device is raised in this paper,which is installed on the specific EPC tagged object directly and this could alleviate the problem *** HIS could store the event data and master data related to the EPC identifier centrally and the application could gain the basic information based on the data stored in the HIS without utilizing ONS,DS and EPCIS.

Epoxidized methyl ricinoleate bio-plasticizer with a pendant acetate ester for PVC artificial material:circumventing existing limit on achievable migration resistance

Processing polyvinyl chloride(PVC)artificial material requires plasticizer that softens the PVC ***,utilizing unsaturated fatty acid methyl esters to obtain epoxidized fatty acid methyl ester(EFAME)bio-plasticizers constitutes an environmentally responsible solution to substitute conventional ortho-phthalates that are endocrine disruptors or probable ***,commercial EFAMEs,even with the highest epoxy value(ca.5.5-5.8%)so far,still suffer from fast leaching from the PVC matrix,burdening the environment and shortening lifespan of the artificial ***,we report a proof-of-principle demonstration of a new strategy to obtain migration-resistant EFAME that harnesses the midchain hydroxyl of methyl ricinoleate and covalently attachment of a pendant acetate *** a low epoxy value(3.0%),the engineered bio-plasticizer displays significantly suppressed migration in multiple scenarios compared with one conventional EFAME with much higher epoxy value(5.8%).Circumventing the limit confronting previous strategy that highlights the sole contribution of epoxy value to achievable migration resistance,the rationale herein may provide guidance for designing new EFAMEs with comparable performance to ortho-phthalates,thus bringing the old and oft-maligned PVC artificial material industry one step closer to sustainability.

Exercise intervention at the early stage of type 2 diabetes mellitus(T2DM)can aid in the maintenance of blood glucose homeostasis and prevent the development of macrovascular and microvascular ***,the exercise-regulated pathways that prevent the development of T2DM remain largely *** this study,two forms of exercise intervention,treadmill training and voluntary wheel running,were conducted for high-fat diet(HFD)-induced obese *** observed that both forms of exercise intervention alleviated HFD-induced insulin resistance and glucose *** muscle is recognized as the primary site for postprandial glucose uptake and for responsive alteration beyond exercise *** profiling of the plasma and skeletal muscle in Chow,HFD,and HFD-exercise groups revealed robust alterations in metabolic pathways by exercise intervention in both *** analysis identified nine metabolites,including beta-alanine,leucine,valine,and tryptophan,which were reversed by exercise treatment in both the plasma and skeletal *** analysis of gene expression profiles in the skeletal muscle revealed several key pathways involved in the beneficial effects of exercise on metabolic *** addition,integrative transcriptomic and metabolomic analyses uncovered strong correlations between the concentrations of bioactive metabolites and the expression levels of genes involved in energy metabolism,insulin sensitivity,and immune response in the skeletal *** work established two models of exercise intervention in obese mice and provided mechanistic insights into the beneficial effects of exercise intervention on systemic energy homeostasis.

The gut microbiota plays a pivotal role in the maintenance of health for amphibians,and it has been fully recognized,but the effectiveness of various influencing factors has not yet been fully *** this association should be considered while the amphibian order Caudata is facing a severe situation of population decline and extinction,there is little understanding of the association between diets and the diversity of gut microbiota in the amphibian order ***,we conducted an extensive analysis of the gut microbiota of Cynops orientalis fed different diets using functional prediction and 16S rRNA amplicon sequencing ***,we found that wild individuals had greater gut microbial diversity and richness in comparison to captive ***,we identified the bacterial taxa associated with diets and observed differences in the relative abundance of gut microbiota among people on various ***,we have a predictive comprehension of the selection and adaptative significance of shared core ASVs in the gut microbiota in maintaining the healthy survival of *** in a large-scale spatiotemporal *** study emphasizes how diets alter the gut microbiota of Caudata and offers fresh perspectives on the conservation and captive management of species in Caudata.

Objective:The cystatins are a family of cysteine protease inhibitors which has been used in the treatment of inflammatory diseases because of its inhibitory effect on enzyme activity and negative *** infarction(MI) is an kind of aseptic inflammatory diseases which is lack of effective drugs for the treatment of *** cystatin family 1(Stefins) is involved in MI is not ***,to explore the protective effect of recombinant rSjcystatin on MI of mice,cardiac structure and function,infiltration of inflammatory cells and serum inflammatory cytokine levels were measured in this ***:120 male C57 BL/6 J wild mice were randomly divided into four groups:(1) Sham group;(2)MI+PBS group;(3) MI+stefin group;(4) MI+dexamethasone(Dex) positive control *** changes of cardiac function and the structure at the 1,3,5,7,14 th and 28 th day post MI were observed by echocardiography,masson trichrome staining,the ratio of heart weight to body weight and the survival *** infiltration of inflammatory cells and sera inflammatory factors levels of TNF-α,IL-6,IL-10 and TGF-β at the 1,3 rd,5 th and 7 day after MI were respectively observed by hematoxylin-eosin(HE) staining and enzyme-linked immunosorbent assay(ELISA).Results:Echo results showed that LVEF(left ventricular ejection fraction) and LVFS(left ventricular fractional shortening) in stefin treatment group and Dex treatment group were significantly higher than those in PBS group at the 28 day after MI(P <0.05).Masson staining results showed that at the 28 day post MI,the left ventricular wall thickness,infarct area and the diameter of left ventricle in stefin group and Dex treatment group were significantly improved compared with those in PBS group(P <0.05).HE staining showed that the inflammatory cells infiltration in PBS group was significantly higher than that in other groups at the same time point after *** results showed that TNF-alpha and IL-6 representin

Nowadays, diverse leather usage conditions and increasing demands from consumers challenge the leather industry. Traditional leather manufacturing is facing long-term challenges, including low-value threshold, confined applica-tion fields, and environmental issues. Leather inherits all the biomimetic properties of natural skin such as flexibility, sanitation, cold resistance, biocompatibility, biodegradability, and other cross-domain functions, achieving unre-mitting attention in multi-functional bio-based materials. Series of researches have been devoted to creating and developing leather-based flexible multi-functional bio-materials, including antibacterial leather, conductive leather, flame-retardant leather, self-cleaning leather, aromatic leather, and electromagnetic shielding leather. In this review, we provide a comprehensive overview of the commonly used leather-based functional materials. Furthermore, the possible challenges for the development of functional leathers are proposed, and expected development directions of leather-based functional materials are discussed. This review may promote and inspire the emerging preparation and applications of leather for flexible functional bio-based materials.

In materials science,the discovery of recipes that yield nanomaterials with defined optical properties is costly and *** this study,we present a two-step framework for a machine learning-driven high-throughput microfluidic platform to rapidly produce silver nanoparticles with the desired absorbance *** a Gaussian process-based Bayesian optimization(BO)with a deep neural network(DNN),the algorithmic framework is able to converge towards the target spectrum after sampling 120 *** the dataset is large enough to train the DNN with sufficient accuracy in the region of the target spectrum,the DNN is used to predict the colour palette accessible with the reaction *** remaining interpretable by humans,the proposed framework efficiently optimizes the nanomaterial synthesis and can extract fundamental knowledge of the relationship between chemical composition and optical properties,such as the role of each reactant on the shape and amplitude of the absorbance spectrum.
