Anti-microbial principles of selected remedial plants from Southern India

Objective:To examine the anti-bacterial activity of leaf extracts of Morus alba L.(Moraceae)and Piper betel L.(Piperaceae),and seed extracts of Bombax ceiba L.(Borabacaceae).Methods:We have partially purified plant extracts by solvent extraction method,and evaluated the effect of individual fractions on bacterial growth using Escherichia coli(***),Pseudomonas aeruginosa(***) and Staphylococcus aureus(***) bacterial ***:Compared with Morus and Bombax fractions,Piper fractions showed significant growth inhibition on all the three types of bacteria *** EtOAc-hexane fractions of Piper leaves exhibited significant antibacterial activity with minimum inhibitory concentrations(MIC) of 50 μg/mL culture against both gram-positive and gram-negative *** EtOAc-fractions Ⅰ,Ⅱ,and Ⅳ inhibited bacterial colony formation on soft agar in addition to growth inhibition.A combination treatment of piper fractions with ampicillin resulted in significant growth inhibition in *** and ***,and combination with anticancer drug geldanamycin(2 μg/mL) showed selective growth inhibition against *** and *** major compounds,i.e.,eugenol,3-hexene-ol and stigmasterol,were primarily identified from Piper betel leaf *** the individual compounds,eugenol treatment showed improved growth inhibition compared with stigmasterol and ***:We are reporting potential anti-bacterial compounds from Piper betel against both gram-positive and gram-negative bacteria either alone or in combination with drug treatment.

The present study focused on the estimation of submarine groundwater discharge(SGD)and the effects of nutrient fluxes due to the SGD *** parameters of SGD such as magnitude,character,and nutrient flux in Punnakayal region of South East coast of India were evaluated using multiple tracers of groundwater inputs in *** was found that the elevated values for the tracers in the study area,displayed a gradational change in the values as move from estuarine part to the *** occurrence of fresh and saline SGD is observed on the study ***,indicated that the SGD fluxes ranged from 0.04 to 0.12 m^3 m^-2d^-1 at the estuary and0.03-0.15 m^3 m^-2d^-1at the groundwater site.A substantially increased value for 222 Rn activities is distinguished in the estuary to values over 312 dpm L^-*** embellishments were generally greatest at locations with substantial meteoric elements in groundwater;however,the recirculation of saltwater through the geological formation could provide a way of transferring terrestrially-derived nutrients to the coastal zone at many places.

Experimental Investigations and Theoretical Modeling Aspects in Column Studies for Removal of Cr(VI) from Aqueous Solutions Using Activated Tamarind Seeds

Continuous adsorption experiments are conducted using fixed-bed adsorption column to evaluate the performance of the adsorbent developed (from activated tamarind seeds) for the removal of Cr(VI) from aqueous solutions and the results obtained are validated with a model developed in this study. The effects of significant parameters such as flow rate, mass of adsorbent, and initial Cr(VI) concentration are studied and breakthrough curves are obtained. As the flow rate increases from 10 to 20 mL min-1, the breakthrough time decreases from 210 to 80 min. As the mass of adsorbent increases, breakthrough time gets delayed. The breakthrough times are obtained as 110, 115 and 210 min for 15, 20 and 25 g of activated tamarind seeds. As the initial Cr(VI) concentration increases from 100 to 200 mgL-1, the break point time decreases from 210 to 45 min. The process parameters for fixed-bed adsorption such as breakthrough time, total percentage removal of Cr(VI), adsorption exhaustion rate and fraction of unused bed length are calculated and the performance of fixed-bed adsorption column is analyzed. The mechanism for Cr(VI) adsorption on activated tamarind seeds is proposed. At low value of solution pH (= 1), the increase in Cr(VI) adsorption is due to the electrostatic attraction between positively charged groups of activated tamarind seeds and the HCrO4-. A mathematical model for fixed-bed adsorption column is proposed by incorporating the effect of velocity variation along the bed length in the existing model. Pore and surface diffusion models are used to describe the intra-particle mechanism for Cr(VI) adsorption. The breakthrough curve obtained theoretically from pore diffusion model and surface diffusion model are compared with experimental results for different operating conditions. The standard deviation values obtained for pore diffusion model and solid diffusion model are 0.111 and 0.214 respectively.

Stability Indicating RP-UPLC Method for Quantification of Glycopyrrolate, Methylparaben and Propylparaben Assay in Liquid Oral Formulation

Stability indicating RP-UPLC technique was developed for the simultaneous quantification of glycopyrrolate, methylparaben, and propylparaben in glycopyrrolate oral solution. The method was established by using gradient UPLC and a Waters Acquity UPLC BEH C18, 100 mm 2.1 mm, i.d 1.7 μm particle size column with a gradient program of mobile phase A and mobile phase B with a flow rate of 0.25 mL/minute, UV wavelength detection at 222 nm, column temperature of 40°C, injection volume of 2 μL, mobile phase A contains 0.05% trifluoro acetic acid in water and Acetonitrile (90:10) v/v and mobile phase-B contains 0.05% trifluoro acetic acid in water and Acetonitrile (10:90) v/v. The current research describes a single UPLC method for developing an assay method for Glycopyrrolate Oral solution that includes Glycopyrrolate (Active), Methylparaben (Preservative), and Propylparaben (Preservative). The assay method was validated in accordance with ICH guidelines. The retention times of glycopyrrolate, methyl paraben and propylparaben were 6.051 min, 3.458 min and 8.095 min, respectively. Linearity range of glycopyrrolate, methyl paraben and propylparaben were in the range of 4 - 32 μg per mL, 35 - 290 μg per mL and 4 - 32 μg per mL, respectively. Recovery of glycopyrrolate, methylparaben and propylparaben ranged from 100.1% to 98.9%, 100.2% - 100.8%, and 100.2% - 100.8%. Validation of analytical method demonstrated that the method is suitable, specific, linear, accurate, precise, rugged and stability indicating for estimating three components in the pharmaceutical dosage form.

Development and Substantiation of a RP-HPLC Method for Monitoring of Impurities in Pirfenidone Drug Substance

A simple, rapid and rugged RP-HPLC method was developed for evaluation and quantification of impurities present in Pirfenidone (PFD) drug substance. Impurities were separated and determined on a Zorbax RX-C18 column (250 mm length, 4.6 mm inner diameter and 5.0 μm particle size, octadecylsilane chemically bonded to porous silica) with 0.02 M KH2PO4 buffer and acetonitrile as mobile phase using a simple gradientelution program. The column flow rate of 1.0 mL per minute was used for the separation. The detection wave length was fixed at 220 nm. The method was substantiated with respect to specificity, precision, linearity, range, accuracy, ruggedness, limit of detection and quantitation. The impurities were identified as 2-hydroxy-5-methylpyridine and Iodobenzene. The linearity range obtained was 0.017 to 0.380 μg/mL for 2-hydroxy-5-methylpyridine, 0.047 to 0.382 μg/mL for Pirfenidone and 0.030 to 0.99 μg/mL for Iodobenzene with the retention times of 3.248 min, 10.608 min and 24.241 min for 2-hydroxy-5-methylpyridine, Pirfenidone and Iodobenzene, respectively. The percentage recoveries of 2-hydroxy-5-methylpyridine and Iodobenzene were in the range of 94.08% - 104.12%. The LOD and LOQ values were found 0.000005 mg/mL, 0.000017 mg/mL for 2-hydroxy-5-methylpyridine and 0.009 μg/mL, 0.030 μg/mL for Iodobenzene, respectively. The method is found to be suitable for the quantitation of impurities along with Pirfenidone drug substance. The method was validated as per the International Conference on Harmonization (ICH) guidelines.

Tuberculous Arthritis of Knee Presenting as Bakers Cyst: A Case Report

Tuberculosis (TB) is no longer confined to undeveloped or developing nations. Today, one third of world’s population suffers from tuberculosis. Tuberculosis (TB) of the musculoskeletal system is an uncommon infection caused by tuberculous bacilli and constitutes 1%-5% of all forms of TB. 30% of skeletal TB involves the joints, the knee being the third most commonly affected after the spine and the hip. Knee tuberculosis commonly presents as synovitis or arthritis. Baker’s cyst is a rather rare complication of knee joint tuberculosis. We report a case of a 50-year-old man with tuberculous arthritis of the right knee who presented to us as baker’s cyst.

Anomalous Left Coronary Artery from Pulmonary Artery with Good Ventricular Function in an Adult. A Rare Scenario

Anomalous origin of the left coronary artery from the pulmonary artery (ALCAPA) with preserved ventricular function is a rare congenital coronary artery anomaly when diagnosed in an adult patient and remains an important cause of sudden cardiac death. We report a 42-year-old patient with ALCAPA managed with intrapulmonary interruption of the left main coronary artery (LMCA) ostea and grafting of the LMCA with left internal mammary artery so as to restore antegrade coronary flow. This restores a dual-coronary-artery system and also allows antegrade blood flow as in a normal coronary artery to a large area of viable myocardium. This strategy is simple to accomplish and is more physiological in an anteriorly placed and dilated LMCA as seen in our case.

Relationship between serum copper and protein thiols in hyperlipidemic patients

Objective:To measure serum copper and protein thiols along with lipid profile in hyperlipidemic patients and to establish the relationship between serum copper,protein thiols and lipid profile ***: The study group consisted of 26 newly diagnosed hyperlipidemic patients not associated with any other systemic diseases and 26 age and sex matched healthy *** lipid profile was done by enzymatic method using automated *** copper and protein thiols were measured ***: Serum copper and protein-SH levels were significantly decreased in cases(P<0.01,P<0.01) compared to *** copper correlated positively with protein thiols(r=0.800,P<0.01) and negatively with LDL cholesterol(r=-0.351 P<0.05).Conclusion: Serum copper and protein thiols were decreased in hyperlipidemia and they correlated positively indicating protective role of copper in preventing homocysteinylation of protein and hence in preventing atherosclerosis.
