Dear Editor,In this letter,a vision-based fixed-time control is proposed for an unmanned aerial vehicle(UAV)with actuator saturation to track an uncooperative target.The fixed-time control is designed in backstepping framework.The relative states between UAV and the target are not directly measured,and are estimated by an onboard monocular camera.

Microorganisms in sediments are critical to marine ecosystems.The microbial communities in marine sediments of the Bohai Sea(BS)and Yellow Sea(YS),the eastern Chinese marginal seas,were uncovered in employing the metagenomic approach.In addition,the taxonomic and functional compositions of microbes were compared among various sediment core depths.Results showing the compositions,diversities,and functions of the microbial communities showed no significant variations with sea areas,and significant seasonal variations was observed in station 3500-7 only on functions of the microbial communities.Moreover,the compositions,diversities,and functions of the microbial communities changed noticeably in different sediment depths in close correlation with physical and chemical properties of sediments.However,the large fraction of the variation in functional communities remained unexplained.From bioinformatic analysis of the metagenomic data,the carbon-metabolismrelated genes such as glycosyl transferase(GT),glycoside hydrolase(GH),and carbohydrate esterase(CE)genes were rich in the microbial community,especially in the top sediment depth.Additionally,in this N-polluted habitat,nitrification and anaerobic ammonia oxidation(anammox)were dominant in the top sediment depth,whereas dissimilatory nitrate reduction to ammonium(DNRA)and denitrification were dominant in the middle and bottom depth,respectively.Further identification of possible biogeochemical links suggested that Gammaproteobacteria,Alphaproteobacteria,Nitrospirae,Nitrospinae,Chloroflexi,and Methanomicrobia might promote effective circulation of carbon and nitrogen cycling.This study expanded our knowledge about the structure and functional potential of microbial communities associated with different sediment depth,and provided further comprehensive information on element cycles in marine environments.

Perovskite solar cells(PSCs)with a light-harvesting three-dimensional perovskite bulk layer as backbone component have achieved great progress in performance.Nonradiative recombination is one major place to improve efficiency and stability as they cause significant energy loss in PSCs.Additionally,an imperfection in grain boundaries will initiate device degradation.One of the most successful strategies to decrease nonradiative recombination in PSCs is the introduction of reduced dimensional perovskite(e.g.,perovskite quantum wells),benefiting the device's efficiency and stability tremendously.Here,instead of quantum wells,mixed-cation perovskites with ligand-contained CsPbBr_(x)I_(3−x)quantum dots(QDs)are prepared,which is shown to function as perovskite healing“surface patches.”Benefiting from the“surface patches”effect,the QDs-film shows reduced defects and enhancing film quality which lead to the excellent performance of solar cells(enhancing the power conversion efficiency from 19.21%of the control device to 21.71%[22.1%in reverse scan]).

DDX39B drives colorectal cancer progression by promoting the stability and nuclear translocation of PKM2

Metastasis is a major cause of colorectal cancer(CRC)mortality,but its molecular mechanisms are still not fully understood.Here,we show that upregulated DDX39B correlates with liver metastases and aggressive phenotypes in CRC.DDX39B is an independent prognostic factor associated with poor clinical outcome in CRC patients.We demonstrate that Sp1 potently activates DDX39B transcription by directly binding to the GC box of the DDX39B promoter in CRC cells.DDX39B overexpression augments the proliferation,migration,and invasion of CRC cells,while the opposite results are obtained in DDX39B-deficient CRC cells.Mechanistically,DDX39B interacts directly with and stabilizes PKM2 by competitively suppressing STUB1-mediated PKM2 ubiquitination and degradation.Importantly,DDX39B recruits importin a5 to accelerate the nuclear translocation of PKM2 independent of ERK1/2-mediated phosphorylation of PKM2,leading to the transactivation of oncogenes and glycolysis-related genes.Consequently,DDX39B enhances glucose uptake and lactate production to activate Warburg effect in CRC.We identify that Arg319 of DDX39B is required for PKM2 binding as well as PKM2 nuclear accumulation and for DDX39B to promote CRC growth and metastasis.In addition,blocking PKM2 nuclear translocation or treatment with glycolytic inhibitor 2-deoxy-D-glucose efficiently abolishes DDX39B-triggered malignant development in CRC.Taken together,ourfindings uncover akey role forDDX39B in modulating glycolytic reprogramming and aggressive progression,and implicate DDX39B as a potential therapeutic target in CRC.

Pulmonary hypertension(PH) is a life-threatening disease characterized by pulmonary vascular remodeling, in which hyperproliferation of pulmonary artery smooth muscle cells(PASMCs)plays an important role. The cysteine 674(C674) in the sarcoplasmic/endoplasmic reticulum Ca^(2+)ATPase 2(SERCA2) is the critical redox regulatory cysteine to regulate SERCA2 activity. Heterozygous SERCA2 C674 S knock-in mice(SKI), where one copy of C674 was substituted by serine to represent partial C674 oxidative inactivation, developed significant pulmonary vascular remodeling resembling human PH, and their right ventricular systolic pressure modestly increased with age. In PASMCs, substitution of C674 activated inositol requiring enzyme 1 alpha(IRE1 a) and spliced X-box binding protein 1(XBP1 s) pathway, accelerated cell cycle and cell proliferation, which reversed by IRE1 a/XBP1 s pathway inhibitor 4μ8 C. In addition, suppressing the IRE1 a/XBP1 s pathway prevented pulmonary vascular remodeling caused by substitution of C674. Similar to SERCA2 a, SERCA2 b is also important to restrict the proliferation of PASMCs. Our study articulates the causal effect of C674 oxidative inactivation on the development of pulmonary vascular remodeling and PH, emphasizing the importance of C674 in restricting PASMC proliferation to maintain pulmonary vascular homeostasis. Moreover, the IRE1 a/XBP1 s pathway and SERCA2 might be potential targets for PH therapy.

Modern pollen-vegetation relationships along altitudinal vegetation belts in the Leigong Mountain of southwestern China

Understanding the distribution,dispersal,and correlation of modern pollen with vegetation in mountainous regions is essential for establishing accurate modern analogs for fossil pollen records.This study,conducted in Leigong Mountain on the YunnanGuizhou Plateau of southwestern China,involved the collection of 35 surface soil samples from diverse vegetation communities along an elevational gradient ranging from 1210 to 1875 meters.The results reveal a close correspondence between modern pollen assemblages and vegetation zones.Principal Component Analysis(PCA)results indicate that pollen assemblages can effectively distinguish between subtropical montane evergreen broad-leaved forest(SEBF)and subtropical montane deciduous broadleaved forest(SDBF).However,both SEBF and SDBF show significant overlap with subtropical montane evergreen-deciduous broad-leaved mixed forest(SEMF).Detrended Correspondence Analysis(DCA)results clearly distinguish the three vegetation zones,and the first axis of DCA shows a significant positive correlation with elevation(plities for the SEBF zone,effectively distinguishing this vegetation zone from others.Pinus and Alnus display overrepresentation in the Leigong Mountain region,while Quercus(D,deciduous-type)and Poaceae exhibit high representation in the SEBF zone.In the SEBF zone,both pollen diversity and richness are the lowest.Our study reveals the complex relationship between the richness and diversity of pollen and vegetation.The diversity and richness of tree and shrub pollen are found to be lower than those of the corresponding plants.The pollen-vegetation relationship elucidated in this study serves as a critical reference for reconstructing ancient environments from fossil pollen retrieved in this region.

Introduction:Repeat positive severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)following COVID-19 initial viral clearance(repositivity)poses a public health management challenge.The objective was to determine factors associated with neutralizing antibody(Nab)level and re-positivity among patients infected with a single strain SARS-CoV-2.Methods:During a single strain SARS-CoV-2 cluster in Beijing,China,longitudinal individual clinical,virological,and immunological data were collected from 368 infections from June 13 to September 22,2020.Factors associated with Nab level and re-positivity were analyzed using generalized estimating equations.Results:A total of 353(96%)SARS-CoV-2 infections had demographic,clinical,and laboratory data available.Among the 353 infections,55(15.5%)were re-positive,and blood draws were taken from 346 individuals(98.0%)during hospitalization and/or during the follow-up period.Symptoms were milder for the second-time admission for the re-positives,although 36.4%of re-positives presented with radiographic appearance of pneumonia manifestation.Compared to non-re-positive patients,NAb titers were lower among re-positives;NAb was positively associated with clinical severity.Samples from the lower respiratory tract manifested higher viral load than that from the upper respiratory tract.Multivariable analysis showed re-positivity was positively associated with being female[odd ratio(OR)=1.7,95%confidence interval(CI)1.1–2.8]and being aged<18 years(OR=5.2,95%CI 1.5–18.1);having initially asymptomatic infection(OR=13.7,95%CI 1.6–116.3);and negatively associated with a higher NAb level(OR=0.9,95%CI 0.5–1.7).Conclusions:NAb may be important for sustained viral clearance.Lower respiratory tract infection was associated with higher viral load among all infections when compared to upper respiratory tract infection.Continuous lower respiratory and intermittent upper respiratory viral shedding among COVID-19 infections may occur.

Background The co-occurrence of depression and anxiety among adolescents is typically associated with suicide ideation.Aims The study aimed to investigate the symptom-level relationship between suicide ideation and the comorbidity of depression and anxiety.Methods 1501 adolescents aged 12-19 years were assessed using the Patient Health Questionnaire(PHQ-9)and the Generalized Anxiety Disorder Scale,and 716 adolescents who scored≥5 on both scales were selected as participants.Network analysis was used to identify the network structure of depressive symptoms and anxiety symptoms.Participants were categorised into either the suicide ideation or non-suicide ideation groups based on their scoring on the suicide-related item in PHQ-9.A comparison was made between the depression-anxiety symptom networks of the two groups.Results‘Restlessness’,‘sad mood’and‘trouble relaxing’were the most prominent central symptoms in the depression-anxiety symptom network,and‘restlessness’,‘nervousness’and‘reduced movement’were the bridge symptoms in this network.‘Sad mood’was found to be directly related to‘suicide ideation’with the highest variance.The network structure was significantly different in properties between the suicide ideation group and the non-suicide ideation group,with‘restlessness’and‘sad mood’exhibiting significantly higher influence in the network of the suicide ideation group than that in the non-suicide ideation group.Conclusion Restlessness and sad mood could be targeted for the intervention of depression-anxiety symptoms among adolescents with suicide ideation.
